    Air Pollution Exposure in Relation to the Commute to School: A Bradford UK Case Study

    Walking School Buses (WSBs) provide a safe alternative to being driven to school. Children benefit from the contribution the exercise provides towards their daily exercise target, it gives children practical experience with respect to road safety and it helps to relieve traffic congestion around the entrance to their school. Walking routes are designed largely based in road safety considerations, catchment need and the availability of parent support. However, little attention is given to the air pollution exposure experienced by children during their journey to school, despite the commuting microenvironment being an important contributor to a child’s daily air pollution exposure. This study aims to quantify the air pollution exposure experienced by children walking to school and those being driven by car. A school was chosen in Bradford, UK. Three adult participants carried out the journey to and from school, each carrying a P-Trak ultrafine particle (UFP) count monitor. One participant travelled the journey to school by car while the other two walked, each on opposite sides of the road for the majority of the journey. Data collection was carried out over a period of two weeks, for a total of five journeys to school in the morning and five on the way home at the end of the school day. Results of the study suggest that car commuters experience lower levels of air pollution dose due to lower exposure and reduced commute times. The largest reductions in exposure for pedestrians can be achieved by avoiding close proximity to traffic queuing up at intersections, and, where possible, walking on the side of the road opposite the traffic, especially during the morning commuting period. Major intersections should also be avoided as they were associated with peak exposures. Steps to ensure that the phasing of lights is optimised to minimise pedestrian waiting time would also help reduce exposure. If possible, busy roads should be avoided altogether. By the careful design of WSB routes, taking into account air pollution, children will be able to experience the benefits that walking to school brings while minimizing their air pollution exposure during their commute to and from school

    An alginate-layer technique for culture of Brassica oleracea L. protoplasts

    Ten accessions belonging to the Brassica oleracea subspecies alba and rubra, and to B. oleracea var. sabauda were used in this study. Protoplasts were isolated from leaves and hypocotyls of in vitro grown plants. The influence of selected factors on the yield, viability, and mitotic activity of protoplasts immobilized in calcium alginate layers was investigated. The efficiency of protoplast isolation from hypocotyls was lower (0.7 ± 0.1 × 106 ml−1) than for protoplasts isolated from leaf mesophyll tissue (2 ± 0.1 × 106 ml−1). High (70–90%) viabilities of immobilized protoplasts were recorded, independent of the explant sources. The highest proportion of protoplasts undergoing divisions was noted for cv. Reball F1, both from mesophyll (29.8 ± 2.2%) and hypocotyl (17.5 ± 0.3%) tissues. Developed colonies of callus tissue were subjected to regeneration and as a result plants from six accessions were obtained

    MECP2 Isoform-Specific Vectors with Regulated Expression for Rett Syndrome Gene Therapy

    BACKGROUND:Rett Syndrome (RTT) is an Autism Spectrum Disorder and the leading cause of mental retardation in females. RTT is caused by mutations in the Methyl CpG-Binding Protein-2 (MECP2) gene and has no treatment. Our objective is to develop viral vectors for MECP2 gene transfer into Neural Stem Cells (NSC) and neurons suitable for gene therapy of Rett Syndrome. METHODOLOGY/PRINCIPAL FINDINGS:We generated self-inactivating (SIN) retroviral vectors with the ubiquitous EF1alpha promoter avoiding known silencer elements to escape stem-cell-specific viral silencing. High efficiency NSC infection resulted in long-term EGFP expression in transduced NSC and after differentiation into neurons. Infection with Myc-tagged MECP2-isoform-specific (E1 and E2) vectors directed MeCP2 to heterochromatin of transduced NSC and neurons. In contrast, vectors with an internal mouse Mecp2 promoter (MeP) directed restricted expression only in neurons and glia and not NSC, recapitulating the endogenous expression pattern required to avoid detrimental consequences of MECP2 ectopic expression. In differentiated NSC from adult heterozygous Mecp2(tm1.1Bird)+/- female mice, 48% of neurons expressed endogenous MeCP2 due to random inactivation of the X-linked Mecp2 gene. Retroviral MECP2 transduction with EF1alpha and MeP vectors rescued expression in 95-100% of neurons resulting in increased dendrite branching function in vitro. Insulated MECP2 isoform-specific lentiviral vectors show long-term expression in NSC and their differentiated neuronal progeny, and directly infect dissociated murine cortical neurons with high efficiency. CONCLUSIONS/SIGNIFICANCE:MeP vectors recapitulate the endogenous expression pattern of MeCP2 in neurons and glia. They have utility to study MeCP2 isoform-specific functions in vitro, and are effective gene therapy vectors for rescuing dendritic maturation of neurons in an ex vivo model of RTT

    Complex time series analysis of PM₁₀ and PM₂.₅ for a coastal site using artificial neural network modelling and k-means clustering

    This paper uses artificial neural networks (ANN), combined with k-means clustering, to understand the complex time series of PM 10 and PM 2.5 concentrations at a coastal location of New Zealand based on data from a single site. Out of available meteorological parameters from the network (wind speed, wind direction, solar radiation, temperature, relative humidity), key factors governing the pattern of the time series concentrations were identified through input sensitivity analysis performed on the trained neural network model. The transport pathways of particulate matter under these key meteorological parameters were further analysed through bivariate concentration polar plots and k-means clustering techniques. The analysis shows that the external sources such as marine aerosols and local sources such as traffic and biomass burning contribute equally to the particulate matter concentrations at the study site. These results are in agreement with the results of receptor modelling by the Auckland Council based on Positive Matrix Factorization (PMF). Our findings also show that contrasting concentration-wind speed relationships exist between marine aerosols and local traffic sources resulting in very noisy and seemingly large random PM 10 concentrations. The inclusion of cluster rankings as an input parameter to the ANN model showed a statistically significant (p < 0.005) improvement in the performance of the ANN time series model and also showed better performance in picking up high concentrations. For the presented case study, the correlation coefficient between observed and predicted concentrations improved from 0.77 to 0.79 for PM 2.5 and from 0.63 to 0.69 for PM 10 and reduced the root mean squared error (RMSE) from 5.00 to 4.74 for PM 2.5 and from 6.77 to 6.34 for PM 10 . The techniques presented here enable the user to obtain an understanding of potential sources and their transport characteristics prior to the implementation of costly chemical analysis techniques or advanced air dispersion models
