    Selective non-steroidal glucocorticoid receptor agonists attenuate inflammation but do not impair intestinal epithelial cell restitution in vitro

    Introduction: Despite the excellent anti-inflammatory and immunosuppressive action of glucocorticoids (GCs), their use for the treatment of inflammatory bowel disease (IBD) still carries significant risks in terms of frequently occurring severe side effects, such as the impairment of intestinal tissue repair. The recently-introduced selective glucocorticoid receptor (GR) agonists (SEGRAs) offer anti-inflammatory action comparable to that of common GCs, but with a reduced side effect profile. Methods: The in vitro effects of the non-steroidal SEGRAs Compound A (CpdA) and ZK216348, were investigated in intestinal epithelial cells and compared to those of Dexamethasone (Dex). GR translocation was shown by immunfluorescence and Western blot analysis. Trans-repressive effects were studied by means of NF-κB/p65 activity and IL-8 levels, trans-activation potency by reporter gene assay. Flow cytometry was used to assess apoptosis of cells exposed to SEGRAs. The effects on IEC-6 and HaCaT cell restitution were determined using an in vitro wound healing model, cell proliferation by BrdU assay. In addition, influences on the TGF-β- or EGF/ERK1/2/MAPK-pathway were evaluated by reporter gene assay, Western blot and qPCR analysis. Results: Dex, CpdA and ZK216348 were found to be functional GR agonists. In terms of trans-repression, CpdA and ZK216348 effectively inhibited NF-κB activity and IL-8 secretion, but showed less trans-activation potency. Furthermore, unlike SEGRAs, Dex caused a dose-dependent inhibition of cell restitution with no effect on cell proliferation. These differences in epithelial restitution were TGF-β-independent but Dex inhibited the EGF/ERK1/2/MAPK-pathway important for intestinal epithelial wound healing by induction of MKP-1 and Annexin-1 which was not affected by CpdA or ZK216348. Conclusion: Collectively, our results indicate that, while their anti-inflammatory activity is comparable to Dex, SEGRAs show fewer side effects with respect to wound healing. The fact that SEGRAs did not have a similar effect on cell restitution might be due to a different modulation of EGF/ERK1/2 MAPK signalling

It has proven to be a valuable plasma source for industry as well as research and commercial use in plasma medicine, leading to very successful therapeutic results and its certification as a medical device. This topical review presents the different kINPen plasma sources available. Diagnostic techniques applied to the kINPen are introduced. The review summarizes the extensive studies of the physics and plasma chemistry of the kINPen performed by research groups across the world, and closes with a brief overview of the main application fields

    Differential expression of protein kinase C isoforms in coronary arteries of diabetic mice lacking the G-protein Gα11

    <p>Abstract</p> <p>Background</p> <p>Diabetes mellitus counts as a major risk factor for developing atherosclerosis. The activation of protein kinase C (PKC) is commonly known to take a pivotal part in the pathogenesis of atherosclerosis, though the influence of specific PKC isozymes remains unclear. There is evidence from large clinical trials suggesting excessive neurohumoral stimulation, amongst other pathways leading to PKC activation, as a central mechanism in the pathogenesis of diabetic heart disease. The present study was therefore designed to determine the role of G<sub>q</sub>-protein signalling via Gα<sub>11 </sub>in diabetes for the expression of PKC isozymes in the coronary vessels.</p> <p>Methods</p> <p>The role of Gα<sub>11 </sub>in diabetes was examined in knockout mice with global deletion of Gα<sub>11 </sub>compared to wildtype controls. An experimental type 1-diabetes was induced in both groups by injection of streptozotocin. Expression and localization of the PKC isozymes α, βII, δ, ε, and ζ was examined by quantitative immunohistochemistry.</p> <p>Results</p> <p>8 weeks after induction of diabetes a diminished expression of PKC <b>ε </b>was observed in wildtype animals. This alteration was not seen in Gα<sub>11 </sub>knockout animals, however, these mice showed a diminished expression of PKCζ. Direct comparison of wildtype and knockout control animals revealed a diminished expression of PKC δ and ε in Gα<sub>11 </sub>knockout animals.</p> <p>Conclusion</p> <p>The present study shows that expression of the nPKCs δ and <b>ε </b>in coronary vessels is under control of the g-protein Gα<sub>11</sub>. The reduced expression of PKC ζ that we observed in coronary arteries from Gα<sub>11</sub>-knockout mice compared to wildtype controls upon induction of diabetes could reduce apoptosis and promote plaque stability. These findings suggest a mechanism that may in part underlie the therapeutic benefit of RAS inhibition on cardiovascular endpoints in diabetic patients.</p

    Single cavitation bubble dynamics in a stagnation flow

    ABSTRACT: Jetting of collapsing bubbles is a key aspect in cavitation-driven fluid–solid interactions as it shapes the bubble dynamics and additionally due to its direct interaction with the wall. We study experimentally and numerically the near-wall collapse and jetting of a single bubble seeded into the stagnation flow of a wall jet, i.e. a jet that impinges perpendicular onto a solid wall. High-speed imaging shows rich and rather distinct bubble dynamics for different wall jet flow velocities and bubble-to-wall stand-off distances. The simulations use a volume-of-fluid method that allows us to numerically determine the microscopic and transient pressures and shear stresses on the wall. It is shown that a wall jet at moderate flow velocities of a few metres per second already shapes the bubble ellipsoidally inducing a planar and convergent jet flow. The distinct bubble dynamics allow us to tailor the wall interaction. In particular, the shear stresses can be increased by orders of magnitude without increasing impact pressures the same way. Interestingly, at small seeding stand-offs, the bubble during the final collapse stage can lift off the wall and migrate against the flow direction of the wall jet such that the violent collapse occurs away from the wall
