    Is Oak Establishment in Old‐fields and Savanna Openings Context Dependent?

    Multiple factors are known to influence tree seedling establishment, yet the degree to which these factors depend on each other and on spatial context is largely unknown. We examined the influence of herbaceous competition and water and nitrogen limitations on tree seedling establishment as functions of distance from trees (within‐site spatial context) and site history (between‐site spatial context; as old‐fields vs. savanna openings). We grew Quercus ellipsoidalis E.J. Hill (pin oak) and Q. macrocarpa Michx. (bur oak) seedlings for 3 years in abandoned agricultural fields and savannas in central Minnesota, USA, near and distant from adult oak trees, with and without water and nitrogen resource additions, and with and without clipping of herbaceous vegetation (reducing above‐ground competition). The strongest treatment effects were found in response to distance from trees and clipping herbaceous vegetation. Ectomycorrhizal infection, year 1 foliar N concentrations, and survival were greater in seedlings growing near vs. distant from adult trees, while clipping herbaceous vegetation increased above‐ground seedling biomass but reduced seedling heights, regardless of distance from adult trees. There were conflicting effects of resource addition, which were dependent on clipping of herbaceous vegetation and site (savanna vs. old‐field). Distance from adult trees and clipping herbaceous vegetation appear to have largely independent effects. Thus, while being near trees benefits seedlings, probably via increased mycorrhizal infection, competition from herbaceous vegetation limits seedlings regardless of distance from trees. In contrast, the effects of resource addition were more context dependent, interacting significantly with herbaceous context and site. The factors influencing seedling success can perhaps be best conceptualized as a series of largely independent environmental filters: seedlings near trees have increased mycorrhizal infection, nutrient uptake and survival, but face competition from herbaceous vegetation regardless of distance from trees. The slow encroachment of woody vegetation into old‐fields and savanna openings in this region is likely to be the result of the net cumulative effect of such filters

    Multi-channel pre-beamformed data acquisition system for research on advanced ultrasound imaging methods

    The lack of open access to the pre-beamformed data of an ultrasound scanner has limited the research of novel imaging methods to a few privileged laboratories. To address this need, we have developed a pre-beamformed data acquisition (DAQ) system that can collect data over 128 array elements in parallel from the Ultrasonix series of research-purpose ultrasound scanners. Our DAQ system comprises three system-level blocks: 1) a connector board that interfaces with the array probe and the scanner through a probe connector port; 2) a main board that triggers DAQ and controls data transfer to a computer; and 3) four receiver boards that are each responsible for acquiring 32 channels of digitized raw data and storing them to the on-board memory. This system can acquire pre-beamformed data with 12-bit resolution when using a 40-MHz sampling rate. It houses a 16 GB RAM buffer that is sufficient to store 128 channels of pre-beamformed data for 8000 to 25 000 transmit firings, depending on imaging depth; corresponding to nearly a 2-s period in typical imaging setups. Following the acquisition, the data can be transferred through a USB 2.0 link to a computer for offline processing and analysis. To evaluate the feasibility of using the DAQ system for advanced imaging research, two proof-of-concept investigations have been conducted on beamforming and plane-wave B-flow imaging. Results show that adaptive beamforming algorithms such as the minimum variance approach can generate sharper images of a wire cross-section whose diameter is equal to the imaging wavelength (150 Όm in our example). Also, planewave B-flow imaging can provide more consistent visualization of blood speckle movement given the higher temporal resolution of this imaging approach (2500 fps in our example). © 2012 IEEE.published_or_final_versio

    Promoting Value Practice in Museums Creates Impact

    This article examines how museological value discussion can offer a tool for museum professionals to engage themselves in the current discourse regarding building sustainable futures. The focus of the article is on collection care and collection development. It describes the latest interview and workshop results regarding museum values in the field of collection development among Finnish museum professionals and students. In addition, it emphasizes the integration of theoretical knowledge and its practical application. Promoting and creating opportunities for value discussion among museum professionals increases the ability of these professionals to further engage in such value-related discourse with various stakeholders. Eventually, the benefits of this kind of value-based discussions are to be seen in the more coherent and focused ones regarding museological values between and among various parties, be they museum professionals, politicians, students or museum visitors. The initial idea for the interviews, and subsequently the workshops as well, emerged from a collection development survey conducted in 2012 among Finnish art museums, which was published in 2016 by the author. Based on the material analyzed at that time, it became clear that the issue of active values in Finnish museums would need further study.Peer reviewe

    Complex facilitation and competition in a temperate grassland: loss of plant diversity and elevated CO\u3csub\u3e2\u3c/sub\u3e have divergent and opposite effects on oak establishment

    Encroachment of woody vegetation into grasslands is a widespread phenomenon that alters plant community composition and ecosystem function. Woody encroachment is often the result of fire suppression, but it may also be related to changes in resource availability associated with global environmental change. We tested the relative strength of three important global change factors (CO2 enrichment, nitrogen deposition, and loss of herbaceous plant diversity) on the first 3 years of bur oak (Quercus macrocarpa) seedling performance in a field experiment in central Minnesota, USA. We found that loss of plant diversity decreased initial oak survival but increased overall oak growth. Conversely, elevated CO2 increased initial oak seedling survival and reduced overall growth, especially at low levels of diversity. Nitrogen deposition surprisingly had no net effect on survival or growth. The magnitude of these effects indicates that long-term woody encroachment trends may be most strongly associated with those few individuals that survive, but grow much larger in lower diversity patches. Further, while the CO2 results and the species richness results appear to describe opposing trends, this is due only to the fact that the natural drivers are moving in opposite directions (decreasing species richness and increasing CO2). Interestingly, the mechanisms that underlie both patterns are very similar, increased CO2 and increased species richness both increase herbaceous biomass which (1) increases belowground competition for resources and (2) increases facilitation of early plant survival under a more diverse plant canopy; in other words, both competition and facilitation help determine community composition in these grasslands

    Characterising a human endogenous retrovirus(HERV)-derived tumour-associated antigen: enriched RNA-Seq analysis of HERV-K(HML-2) in mantle cell lymphoma cell lines.

    BACKGROUND: The cell-surface attachment protein (Env) of the HERV-K(HML-2) lineage of endogenous retroviruses is a potentially attractive tumour-associated antigen for anti-cancer immunotherapy. The human genome contains around 100 integrated copies (called proviruses or loci) of the HERV-K(HML-2) virus and we argue that it is important for therapy development to know which and how many of these contribute to protein expression, and how this varies across tissues. We measured relative provirus expression in HERV-K(HML-2), using enriched RNA-Seq analysis with both short- and long-read sequencing, in three Mantle Cell Lymphoma cell lines (JVM2, Granta519 and REC1). We also confirmed expression of the Env protein in two of our cell lines using Western blotting, and analysed provirus expression data from all other relevant published studies. RESULTS: Firstly, in both our and other reanalysed studies, approximately 10% of the transcripts mapping to HERV-K(HML-2) came from Env-encoding proviruses. Secondly, in one cell line the majority of the protein expression appears to come from one provirus (12q14.1). Thirdly, we find a strong tissue-specific pattern of provirus expression. CONCLUSIONS: A possible dependency of Env expression on a single provirus, combined with the earlier observation that this provirus is not present in all individuals and a general pattern of tissue-specific expression among proviruses, has serious implications for future HERV-K(HML-2)-targeted immunotherapy. Further research into HERV-K(HML-2) as a possible tumour-associated antigen in blood cancers requires a more targeted, proteome-based, screening protocol that will consider these polymorphisms within HERV-K(HML-2).     Renewing Criminalized and Hegemonic Cultural Landscapes

    The Mafia's long historical pedigree in Mezzogiorno, Southern Italy, has empowered the Mafioso as a notorious, uncontested, and hegemonic figure. The counter-cultural resistance against the mafiosi culture began to be institutionalized in the early 1990s. Today, Libera Terra is the largest civil society organization in the country that uses the lands confiscated from the Mafia as a space of cultural repertoire to realize its ideals. Deploying labor force through volunteer participation, producing biological fruits and vegetables, and providing information to the students on the fields are the principal cultural practices of this struggle. The confiscated lands make the Italian experience of anti-Mafia resistance a unique example by connecting the land with the ideals of cultural change. The sociocultural resistance of Libera Terra conveys a political message through these practices and utters that the Mafia is not invincible. This study draws the complex panorama of the Mafia and anti-Mafia movement that uses the ‘confiscated lands’ as cultural and public spaces for resistance and socio-cultural change. In doing so, this article sheds new light on the relationship between rural criminology and crime prevention policies in Southern Italy by demonstrating how community development practice of Libera Terra changes the meaning of landscape through iconographic symbolism and ethnographic performance

    Performance of masonry buildings and churches in the 22 february 2011 christchurch earthquake

    As part of the „Project Masonry‟ Recovery Project funded by the New Zealand Natural Hazards Research Platform, commencing in March 2011, an international team of researchers was deployed to document and interpret the observed earthquake damage to masonry buildings and to churches as a result of the 22nd February 2011 Christchurch earthquake. The study focused on investigating commonly encountered failure patterns and collapse mechanisms. A brief summary of activities undertaken is presented, detailing the observations that were made on the performance of and the deficiencies that contributed to the damage to approximately 650 inspected unreinforced clay brick masonry (URM) buildings, to 90 unreinforced stone masonry buildings, to 342 reinforced concrete masonry (RCM) buildings, to 112 churches in the Canterbury region, and to just under 1100 residential dwellings having external masonry veneer cladding. In addition, details are provided of retrofit techniques that were implemented within relevant Christchurch URM buildings prior to the 22nd February earthquake and brief suggestions are provided regarding appropriate seismic retrofit and remediation techniques for stone masonry buildings.The authors acknowledge the financial support for Project Masonry from the New Zealand Natural Hazards Research Platform.     Ectomycorrhizal fungal communities of oak savanna are distinct from forest communities

    Abstract: Oak savanna is one of the most endangered ecosystems of North America, with less than 0.02% of its original area remaining. Here we test whether oak savanna supports a unique community of ectomycorrhizal fungi, a higher diversity of ectomycorrhizal fungi or a greater proportional abundance of ascomycete fungi compared with adjacent areas where the absence of fire has resulted in oak savanna conversion to oak forest. The overall fungal community was highly diverse and dominated by Cenococcum geophilum and other ascomycetes, Cortinarius, Russula, Lactarius and Thelephoraceae. Oak savanna mycorrhizal communities were distinct from oak forest communities both aboveground (sporocarp surveys) and belowground (RFLP identification of ectomycorrhizal root tips); however total diversity was not higher in oak savanna than oak forests and there was no evidence of a greater abundance of ascomycetes. Despite not having a higher local diversity than oak forests, the presence of a unique fungal community indicates that oak savanna plays an important role in maintaining regional ectomycorrhizal diversity
