    Activation of Stress Kinases in the Brain of Mucopolysaccharidosis IIIB mice

    The accumulation of heparan sulfate (HS) in lysosomes is the primary consequence of the enzyme defect (α-N-acetylglucosaminidase) in mucopolysaccharidosis type IIIB. This accumulation triggers a cascade of pathological events that progressively leads to CNS pathology. Here we examined the activation of the three major stress kinases in the neuronal tissue of a murine model of the disease. ERK1/2 was significantly higher in the cortex of 1–2-month-old affected animals compared with wild-type (Wt) littermates. Similarly, ERK1/2 was stimulated in neurons cultured from MPS IIIB mice. SAPK/JNK was also found to be activated in the cortex of 1–2-month-old affected animals compared with Wt subjects, and the same was found for cultured neurons. In contrast, the active form of p38MAPK was lower in the cortex of 1-month-old MPS IIIB mice compared with Wt animals, but no significant difference was found between the two p38MAPK analyzed in normal and affected neurons cultured in vitro. These data indicate the possible involvement of MAPK dysregulation in the early stage of MPS IIIB brain disease. © 2011 Wiley-Liss, Inc

    Fully discrete polynomial de Rham sequences of arbitrary degree on polygons and polyhedra

    In this work, merging ideas from compatible discretisations and polyhedral methods, we construct novel fully discrete polynomial de Rham sequences of arbitrary degree on polygons and polyhedra. The spaces and operators that appear in these sequences are directly amenable to computer implementation. Besides proving exactness, we show that the usual sequence of Finite Element spaces forms, through appropriate interpolation operators, a commutative diagram with our sequence, which ensures suitable approximation properties. A discussion on reconstructions of potentials and discrete L2L^2-products completes the exposition

    Extra Virgin Olive Oil and Cardiovascular Protection in Chronic Kidney Disease

    The high mortality related to chronic kidney disease (CKD) is not only due to the disease itself; in fact, CKD also represents an important risk factor for cardiovascular (CV) morbidity and mortality. Among the functional foods that seems to have cardioprotective action, extra virgin olive oil (EVOO) plays a pivotal health-promoting role. The aim of this study was to evaluate the possible cardioprotective effects of an EVOO containing a very high content (>900 ppm) of minor phenolic compounds (MPCs). The selected EVOO was analyzed by HPLC-DAD-MS to establish the MPC content. The Olea extract obtained from the selected EVOO was tested against the RAW 264.7 cell line in order to investigate its anti-inflammatory activity. We enrolled 40 CKD patients under conservative therapy for in vivo clinical testing. All CKD patients consumed 40 mL/day of raw EVOO for 9 weeks (T1). At baseline (T0) and at T1, we monitored the patients' blood and urinary parameters. The patients' body composition was assessed using bioelectrical impedance analysis and the carotid intima-media thickness (CIMT) using ultrasound imaging. At T1, we observed a decrease in inflammatory parameters, CIMT, and oxidative stress biomarkers. We also noticed improvements in lipid and purine metabolism, atherogenic indices, and body composition. Thus, this study highlighted the cardioprotective action of EVOO in nephropathic patients

    Determination of pesticides in the respirable fraction of airborne particulate matter by high-performance liquid chromatography–tandem mass spectrometry

    Potential harmful effects of pesticides include risks to human health of workers involved in the wet spray application in cultivated areas. Inhalation exposure depends on several factors including pesticide concentrations in the respirable fraction of airborne particulate matter (PM4). To ensure a high level of protection, the use of tractors with cabins provides protection against dust, aerosols, and vapors. Since tractors not providing maximum protection are still in use, PM4 was sampled during spreading operations in agricultural fields inside and outside tractor cabins. Sample preparation technique based on accelerated solvent extraction and solid-phase extraction cleanup was optimized before analysis of nine pesticides in PM4. Meptyldinocap, deltamethrin, myclobutanil, fluopyram, methoxyfenozide, dimethomorph, fluopicolide, cyflufenamid, and metrafenone were simultaneously determined by high-performance liquid chromatography–electrospray ionization–tandem mass spectrometry (HPLC–ESI–MS–MS). The results demonstrated the efficacy of the tractor cabs used in the sampling sites. © 2017 Taylor & Francis

    Prenatal exome sequencing: background, current practice and future perspectives - A systematic review

    The introduction of Next Generation Sequencing (NGS) technologies has exerted a significant impact on prenatal diagnosis. Prenatal Exome Sequencing (pES) is performed with increasing frequency in fetuses with structural anomalies and negative chromosomal analysis. The actual diagnostic value varies extensively, and the role of incidental/secondary or inconclusive findings and negative results has not been fully ascertained. We performed a systematic literature review to evaluate the diagnostic yield, as well as inconclusive and negative-result rates of pES. Papers were divided in two groups. The former includes fetuses presenting structural anomalies, regardless the involved organ; the latter focuses on specific class anomalies. Available findings on non-informative or negative results were gathered as well. In the first group, the weighted average diagnostic yield resulted 19%, and inconclusive finding rate 12%. In the second group, the percentages were extremely variable due to differences in sample sizes and inclusion criteria, which constitute major determinants of pES efficiency. Diagnostic pES availability and its application have a pivotal role in prenatal diagnosis, though more homogeneity in access criteria and a consensus on clinical management of controversial information management is envisageable to reach widespread use in the near future

    Employment insecurity and mental health during the economic recession : an analysis of the young adult labour force in Italy

    Background and objective. A growing body of scientific literature highlights the negative consequences of employment insecurity on several life domains. This study focuses on the young adult labour force in Italy, investigating the relationship between employment insecurity and mental health and whether this has changed after years of economic downturn. It enhances understanding by addressing differences in mental health according to several employment characteristics; and by exploring the role of respondents’ economic situation and educational level. Data and Methods. Data from a large-scale, nationally representative health survey are used to estimate the relationship between employment insecurity and the Mental Health Inventory (MHI), by means of multiple linear regressions. Results and Conclusions. The study demonstrates that employment insecurity is associated with poorer mental health. Moreover, neither temporary workers nor unemployed individuals are a homogeneous group. Previous job experience is important in differentiating the mental health risks of unemployed individuals; and the effects on mental health vary according to occupational status and to the amount of time spent in a condition of insecurity. Further, the experience of financial difficulties partly explains the relationship between employment insecurity and mental health; and different mental health outcomes depend on respondents’ educational level.     A low-cost radiation detection system to monitor radioactive environments by unmanned vehicles

    AbstractUnconventional scenarios with hazardous radioactive levels are expected as consequences of accidents in the industrial sector of the nuclear energy production or following intentional releases of radioactive materials for terrorist purposes (dirty bombs, indoor contaminations, etc.). Nowadays, the need to balance the high standards of safety and security through an effective detection network is a matter of paramount importance. In this work, the authors' challenge has been to design, realize and test a low-cost gamma detection and spectroscopy system which may be used in unmanned vehicles in general and/or drones with low payload capabilities. The designed platform may be used to carry out mapping or localization operations in order to reduce the risk factor for first responders or for the population affected by radiological and nuclear events. In this paper, the design process of a gamma ray detection and spectroscopy system based on affordable and commercially available technologies is presented along with the results of our ongoing characterization of the prototype

    A Pipeline for the Implementation of Immersive Experience in Cultural Heritage Sites in Sicily

    Modern digital technologies allow potentially to explore Cultural Heritage sites in immersive virtual environments. This is surely an advantage for the users that can better experiment and understand a specific site, also before a real visit. This specific approach has gained increasing attention during the extreme conditions of the recent COVID-19 pandemic. In this work, we present the processes that lead to the implementation of an immersive app for different kinds of low and highcost devices, which have been attained in the context of the 3DLab-Sicilia project. 3DLab-Sicilia’s main objective is to sponsor the creation, development, and validation of a sustainable infrastructure that interconnects three main Sicilian centers specialized in augmented and virtual reality. The project gives great importance to the cultural heritage, as well as to the tourism related areas. Despite the presentation of the case study of the Santa Maria La Vetere church, the process of the final app implementation guided by the general pipeline here presented is general and can be applied to other cultural heritage sites

    A Case Study for the Design and Implementation of Immersive Experiences in support of Sicilian Cultural Heritage

    Virtual Reality (VR) is a robust tool for sponsoring Cultural Heritage sites. It enables immersive experiences in which the user can enjoy the cultural assets virtually, behaving as he/she would do in the real world. Covid-19 pandemic has shed light on the importance of using VR in cultural heritage, showing advantages for the users that can visit the site safely through specific devices. In this work, we present the processes that lead to the creation of an immersive app that makes explorable a famous cultural asset in Sicily, the church of SS. Crocifisso al Calvario. The application creation process will be described in each of its parts, beginning from the digital acquisition of the cultural asset to the development of the user interface. The application is provided for three different VR devices: smartphones equipped with cardboards, headsets, and CAVE. The paper is supported by the 3dLab-Sicilia project, whose main objective is to sponsor the creation, development, and validation of a sustainable infrastructure that interconnects three main Sicilian centres specialized in augmented and virtual reality
