    Structure-Activity Relationship for the Oxadiazole Class of Antibacterials

    A structure-activity relationship (SAR) for the oxadiazole class of antibacterials was evaluated by syntheses of 72 analogs and determination of the minimal-inhibitory concentrations (MICs) against the ESKAPE panel of bacteria. Selected compounds were further evaluated for in vitro toxicity, plasma protein binding, pharmacokinetics (PK), and a mouse model of methicillin-resistant Staphylococcus aureus (MRSA) infection. Oxadiazole 72c shows potent in vitro antibacterial activity, exhibits low clearance, a high volume of distribution, and 41% oral bioavailability, and shows efficacy in mouse models of MRSA infection.Fil: Boudreau, Marc A..     Prevalence and Determinants of Metabolic Syndrome among Women in Chinese Rural Areas

    BACKGROUND AND AIMS: Metabolic syndrome (MS) is prevalent in recent years but few data is reported in the rural areas in China. The aim of this study was to examine MS prevalence and its risk factors among women in rural China. METHODS AND RESULTS: The Nantong Metabolic Syndrome Study (NMSS), a population based cross-sectional study, was conducted during 2007-2008 in Nantong, China. In person interviews, blood glucose and lipid measurements were completed for 13,505 female participants aged 18-74 years. The International Diabetes Federation (IDF), the US Third Report of the National Cholesterol Education Program, the Adult Treatment Panel (ATPIII) and modified ATPIII for Asian population has determined three criteria of MS. These criteria for MS were used and compared in this study. The prevalence of MS was 22.0%, 16.9% and 23.3% according to IDF, ATPIII and ATPIII-modified criteria, respectively. Levels of agreement of these criteria for MS were above 0.75. We found that vigorous-intensity of occupational physical activity was associated with a low prevalence of MS with OR of 0.76 (95% confidence interval (CI): 0.63-0.91). Rice wine drinkers (alcohol >12.8 g/day) had about 34% low risks of developing MS with OR of 0.66 (95% CI: 0.48-0.91), compared with non-drinkers. Odds ratio of MS was 1.81 (95% CI: 1.15-2.84) in women who smoked more than 20 pack-years, compared to non-smokers. Odds ratio of MS was 1.56 (95% CI: 1.25-1.95) in women who had familial history of diseases, including hypertension, diabetes and stroke, compared to women without familial history of those diseases. CONCLUSION: MS is highly prevalent among women in rural China. Both physical activity and rice wine consumption play a protective role, while family history and smoking are risk factors in MS development. Educational programs should be established for promoting healthy lifestyles and appropriate interventions in rural China

    Characteristics and Source Apportionment of Summertime Volatile Organic Compounds in a Fast Developing City in the Yangtze River Delta, China

    Volatile organic compounds (VOCs) are crucial for ozone formation in the Yangtze River Delta (YRD) in China. The characteristics of ambient VOCs in Nantong, a fast developing city in the YRD, were studied. Sixty ambient air samples were taken at five sites in three sampling days during summer time. One hundred and five VOCs were measured, showing that VOC concentration varied between 27.5 ppbv to 33.1 ppbv at five sites; these levels were generally lower than those for some big Chinese cities like Beijing, Shanghai and Nanjing. With larger fractions of alkanes, the Zilang (ZL) and Sanqu (SQ) sites had relatively higher VOC concentrations among the five sites. The oxidation formation potential (OFP) and secondary aerosol formation potential (SOAFP) were estimated to be 125 μg/m3 and 0.76 μg/m3, respectively. These two values were smaller than those in other big cities, as they were dominated by aromatic compounds (e.g., toluene and benzene) of which concentrations in Nantong were found to be lower. The highest toluene concentration was measured in ZL, implying substantial effects of surface coating industry near the site. Through the Positive Matrix Factorization (PMF) model, the identified sources of VOCs included LPG (Liquefied petroleum gas) combustion (13.9%), chemical industry (8.5%), natural gas use (15.6%), gasoline evaporation (12.8%), petrol industry use (11.8%), solvent use (16.2%) vehicle exhausts (12.1%) and surface coating (9.2%). A relatively small contribution from vehicles was found in Nantong compared with other big cities. Moreover, LPG emissions were identified to be relatively important in Nantong, indicated by the large mass fraction of propane and ethane concentrations in the atmosphere

    Novel biphasic role of resolvin D1 on expression of cyclooxygenase-2 in lipopolysaccharide-stimulated lung fibroblasts is partly through PI3K/AKT and ERK2 pathways

    Fibroblasts, far frombeing merely bystander cells, are known to play a specific role in inflammation resolution after an acute injury. As the endogenous “braking signal,” resolvins possess potent anti-inflammatory and proresolution actions. We demonstrated that the expression of COX-2 protein was significantly peaked initially at 6 hours but then also at 48 hours after LPS stimulation in lung fibroblasts. PGE2 levels also peaked at 6 hours, and PGD2 levels were increased and peaked at 48 hours. However, no significant change in the protein expression of COX-1 was observed after treatment with LPS in lung fibroblasts. Exogenous resolvin D1 inhibited the first peak of COX-2 expression as well as the production of PGE2 induced by LPS. In contrast, exogenous resolvin D1 increased the second peak of COX-2 expression as well as the production of PGD2 induced by LPS. In addition, resolvin D1 inhibited COX-2 expression at 6 hours, which was partly through PI3K/AKT and ERK2 signalling pathways

    A Potent and Narrow-Spectrum Antibacterial against <i>Clostridioides difficile</i> Infection

    Clostridioides difficile is an anaerobic Gram-positive bacterium that colonizes the gut of patients treated with broad-spectrum antibiotics. The normal gut microflora prevents C. difficile colonization; however, dysbiosis by treatment with broad-spectrum antibiotics causes recurrent C. difficile infection (CDI) in 25% of patients. There are no fully effective antibiotics for multiple recurrent CDIs. We report herein that oxadiazole antibiotics exhibit bactericidal activity against C. difficile vegetative cells. We screened a library of 75 oxadiazoles against C. difficile ATCC 43255. The findings from this collection served as the basis for the syntheses of an additional 58 analogs, which were tested against the same strain. We report a potent (MIC50 = 0.5 μg/mL and MIC90 = 1 μg/mL values for 101 C. difficile strains) and narrow-spectrum oxadiazole (3-(4-(cyclopentyloxy)­phenyl)-5-(4-nitro-1H-imidazol-2-yl)-1,2,4-oxadiazole; compound 57), which is not active against common gut bacteria or other tested organisms. Compound 57 is selectively bactericidal against C. difficile and targets cell-wall synthesis

    Polymer electrolyte glue : a universal interfacial modification strategy for all-solid-state Li batteries

    In recent years solid Li+ conductors with competitive ionic conductivity to those of liquid electrolytes have been reported. However, the incorporation of highly conductive solid electrolytes into the lithium-ion batteries is still very challenging mainly due to the high resistance existing at the solid-solid interfaces throughout the battery structure. Here, we demonstrated a universal interfacial modification strategy through coating a curable polymer-based glue electrolyte between the electrolyte and electrodes, aiming to address the poor solid-solid contact and thus decrease high interfacial resistance. The liquid glue exhibits both great wettability as well as chemical/electrochemical stability to most of the electrodes, and it can be easily solidified into a polymer electrolyte layer through a "post-curing" treatment. As a result, symmetric Li batteries with the glue modification exhibit much smaller impedance and enhanced stability upon plating/stripping cycles compared to the batteries without glue modification. The all-solid-state Li-S batteries with glue modification show significantly enhanced performances. The strategy of developing glue electrolytes to improve the electrode-electrolyte interface contact provides an alternative option for improving many other solid-state batteries.Accepted versio
