    Sulfoquinovose is produced by photosynthetic organisms at a rate of 1010 tons per annum and is degraded by bacteria as a source of carbon and sulfur. We have identified Escherichia coli YihQ as the first dedicated sulfoquinovosidase and the gateway enzyme to sulfoglycolytic pathways. Structural and mutagenesis studies unveiled the sequence signatures for binding the distinguishing sulfonate residue and revealed that sulfoquinovoside degradation is widespread across the tree of life

    Detecting a stochastic gravitational wave background with the Laser Interferometer Space Antenna

    The random superposition of many weak sources will produce a stochastic background of gravitational waves that may dominate the response of the LISA (Laser Interferometer Space Antenna) gravitational wave observatory. Unless something can be done to distinguish between a stochastic background and detector noise, the two will combine to form an effective noise floor for the detector. Two methods have been proposed to solve this problem. The first is to cross-correlate the output of two independent interferometers. The second is an ingenious scheme for monitoring the instrument noise by operating LISA as a Sagnac interferometer. Here we derive the optimal orbital alignment for cross-correlating a pair of LISA detectors, and provide the first analytic derivation of the Sagnac sensitivity curve.Comment: 9 pages, 11 figures.     Estradiol Regulates Expression of Estrogen Receptor ERα46 in Human Macrophages

    BACKGROUND:Monocytes and macrophages are key innate immune effector cells that produce cytokines and chemokines upon activation. We and others have shown that 17beta-estradiol (E2) has a direct role in the modulation of monocyte and macrophage immune function. However, relatively little is known about the ability of E2 to regulate isoform expression of estrogen receptors (ERs) in these cells. METHODOLOGY/PRINCIPAL FINDINGS:In this study, we quantify expression of ERalpha and ERbeta in human monocytes and macrophages. We also show for the first time that the N-terminal truncated ERalpha variant, ERalpha46, is expressed in both cell types. Promoter utilization studies reveal that transcription of ERalpha in both cell types occurs from upstream promoters E and F. Treatment with E2 induces ERalpha expression in macrophages but has no effect on ERbeta levels in either cell type. During monocyte-to-macrophage differentiation, ERalpha is upregulated in a time-dependent manner. Previous studies by our group demonstrated that E2 treatment attenuates production of the chemokine CXCL8 in an ER-dependent manner. We now show that ERalpha expression levels parallel the ability of E2 to suppress CXCL8 production. CONCLUSIONS/SIGNIFICANCE:This work demonstrates for the first time that human macrophages predominantly express the truncated ER variant ERalphap46, which is estradiol-inducible. This is mediated through usage of the ERalpha F promoter. Alternative promoter usage may account for tissue and cell type-specific differences in estradiol-induced effects on gene expression. These studies signify the importance of ERalpha expression and regulation in the ability of E2 to modulate innate immune responses

    Key signalling nodes in mammary gland development and cancer. Mitogen-activated protein kinase signalling in experimental models of breast cancer progression and in mammary gland development

    Seven classes of mitogen-activated protein kinase (MAPK) intracellular signalling cascades exist, four of which are implicated in breast disease and function in mammary epithelial cells. These are the extracellular regulated kinase (ERK)1/2 pathway, the ERK5 pathway, the p38 pathway and the c-Jun N-terminal kinase (JNK) pathway. In some forms of human breast cancer and in many experimental models of breast cancer progression, signalling through the ERK1/2 pathway, in particular, has been implicated as being important. We review the influence of ERK1/2 activity on the organised three-dimensional association of mammary epithelial cells, and in models of breast cancer cell invasion. We assess the importance of epidermal growth factor receptor family signalling through ERK1/2 in models of breast cancer progression and the influence of ERK1/2 on its substrate, the oestrogen receptor, in this context. In parallel, we consider the importance of these MAPK-centred signalling cascades during the cycle of mammary gland development. Although less extensively studied, we highlight the instances of signalling through the p38, JNK and ERK5 pathways involved in breast cancer progression and mammary gland development

    Simulating textual scenarios using state charts

    This paper describes an approach that enables early analysis of the dynamic behaviour of textual scenarios by simulation. It is commonly accepted that detecting defects as early as possible is central in the development process. However, for the early phases, the only available techniques are usually static ones, such as inspections, making analysis of the dynamic behaviour difficult. This paper presents an approach where textual scenarios, which represent system requirements, are transformed into state charts. The state charts are then simulated, making an analysis of the dynamic behaviour possible. Early experiences indicate that many defects, otherwise difficult to detect, are found both in the transformation and in the simulation. As the transformation is straightforward and relatively inexpensive, the benefits seem to outweigh the costs

    Assessing the Impact of Active Guidance for Defect Detection: A Replicated Experiment

    Scenario-based reading (SBR) techniques have been proposed as an alternative to checklists to support the inspectors throughout the reading process in the form of operational scenarios. Many studies have been performed to compare these techniques regarding their impact on the inspector performance. However, most of the existing studies have compared generic checklists to a set of specific reading scenarios, thus confounding the effects of two SBR key factors: separation of concerns and active guidance. In a previous work we have preliminarily conducted a repeated case study at the University of Kaiserslautern to evaluate the impact of active guidance on inspection performance. Specifically, we compared reading scenarios and focused checklists, which were both characterized as being perspective-based. The only difference between the reading techniques was the active guidance provided by the reading scenarios. We now have replicated the initial study with a controlled experiment using as subjects 43 graduate students in computer science at University of Bari. We did not find evidence that active guidance in reading techniques affects the effectiveness or the efficiency of defect detection. However, inspectors showed a better acceptance of focused checklists than reading scenarios
