Gratia Adami, gratia Christi. La nature, la Loi et la grâce dans le premier augustinisme

La question de la Loi, rapportée à la nature et à la grâce, fut l’une des principales préoccupations de ceux qui, après sa mort, défendirent Augustin contre les « semipélagiens », de Cassien à Fauste de Riez. Une analyse des probables filiations entre les textes d’Augustin et ceux de ses défenseurs, Prosper d’Aquitaine et Fulgence de Ruspe, permet d’observer les modalités et les motivations de l’émergence d’un augustinisme, entendu au sens d’un durcissement, conscient ou non, de la doctrine originelle. Si les termes et les développements restent en apparence identiques et profondément ancrés dans la théologie d’Augustin, leur application à des objets variés et leur insertion dans des contextes inédits contribuent à donner de la pensée d’Augustin une représentation biaisée.The question of the Law as related to nature and grace was one of the main concerns of those who, after Augustine’s death, defended him against the so-called “Semipelagians”, from Cassian to Faustus of Riez. An analysis of the probable filiations between the texts by Augustine and those of his defenders, Prosper of Aquitaine and Fulgentius of Ruspe, allows us to observe the terms and reasons for the emergence of Augustinianism, in the sense of a partly conscious and partly unconscious hardening of the original doctrine. BIOACTIVITIES OF HOP-DERIVED PRENYLFLAVONOIDS IN RELATION TO PROSTATE CANCER

Prostate cancer (PC) is highly common among aging men causing bothersome symptoms or developing into a life-threatening disease. Phytoestrogens could influence hormone-dependent diseases including PC by their varying bio-activities. Recently, five prenylflavonoids xanthohumol (X), desmethylxanthohumol (DMX), isoxanthohumol (IX), 8-prenylnaringenin (8PN), and 6-prenylnaringenin (6PN) were identified in hops (Humulus lupulus L.) which were investifated in this research project for their chemopreventive or therapeutic properties on PC. The compounds were found to inhibit the proliferation of the human prostate cancer cells PC-3, DU145, and LNCaP.FGC in a dose-dependent manner. Antiproliferative activities of X and 6PN were found to be strongest for PC-3 and DU145, while, on LNCaP.FGC, these compounds first exerted a strong induction of cell proliferation followed by a proliferation shut-off. 8PN, which has previously been identified as a highly potent phytoestrogen, showed weaker antiproliferative effects on PC-3 and DU145. However, on LNCaP.FGC, 8PN demonstrated a more potent growth inhibition. IX exhibited activities on PC-3 and DU145 comparable to those of 8PN, but it was more effective on LNCaP.FGC. DMX was the least active compound in all cell lines. No quantitative relation between the estrogenic potency as measured in the recombinant yeast estrogen screen and antiproliferative effects could be concluded. The strong antiproliferative activity of X in endothelial cells suggests potential antiangiogenic activity compromising tumor growth and metastasis. Further research into the underlying mechanisms of growth inhibition revealed that there was no evidence for induction of apoptotic cell death in PC-3 and DU145 upon treatment with X, IX, 8PN, and 6PN. However, considering the formation of vacuoles in PC-3 when treated with IX and 6PN, and in DU145 when treated with IX, 6PN, and 8PN, it is suggested that autophagy is induced, which could consequently lead to cell death in this model. It can be concluded that X, IX, 8PN, and 6PN appear to be promising candidates for further investigation in prostate anticancer therapy and chemoprevention, especially in the final stages

From Lateral Flow Devices to a Novel Nano-Color Microfluidic Assay

Improving the performance of traditional diagnostic lateral flow assays combined with new manufacturing technologies is a primary goal in the research and development plans of diagnostic companies. Taking into consideration the components of lateral flow diagnostic test kits; innovation can include modification of labels, materials and device design. In recent years, Resonance-Enhanced Absorption (REA) of metal nano-particles has shown excellent applicability in bio-sensing for the detection of a variety of bio-molecular binding interactions. In a novel approach, we have now integrated REA-assays in a diagnostic microfluidic setup thus resolving the bottleneck of long incubation times inherent in previously existing REA-assays and simultaneously integrated automated fabrication techniques for diagnostics manufacture. Due to the roller-coating based technology and chemical resistance, we used PET-co-polyester as a substrate and a CO2 laser ablation system as a fast, highly precise and contactless alternative to classical micro-milling. It was possible to detect biological binding within three minutes – visible to the eye as colored text readout within the REA-fluidic device. A two-minute in-situ silver enhancement was able to enhance the resonant color additionally, if required
