    Regulation of local expression of cell adhesion and motility-related mRNAs in breast cancer cells by IMP1/ ZBP1

    Summary Metastasis involves tumor cell detachment from the primary tumor, and acquisition of migratory and invasive capabilities. These capabilities are mediated by multiple events, including loss of cell-cell contact, an increase in focal adhesion turnover and failure to maintain a normal cell polarity. We have previously reported that silencing of the expression of the zipcode-binding protein IMP1/ZBP1 in breast tumor patients is associated with metastasis. IMP1/ZBP1 selectively binds to a group of mRNAs that encode important mediators for cell adhesion and motility. Here, we show that in both T47D and MDA231 human breast carcinoma cells IMP1/ZBP1 functions to suppress cell invasion. Binding of ZBP1 to the mRNAs encoding E-cadherin, b-actin, a-actinin and the Arp2/3 complex facilitates localization of the mRNAs, which stabilizes cell-cell connections and focal adhesions. Our studies suggest a novel mechanism through which IMP1/ZBP1 simultaneously regulates the local expression of many cell-motility-related mRNAs to maintain cell adherence and polarity, decrease focal adhesion turnover and maintain a persistent and directional motility

    GPX8 regulates pan-apoptosis in gliomas to promote microglial migration and mediate immunotherapy responses

    IntroductionGliomas have emerged as the predominant brain tumor type in recent decades, yet the exploration of non-apoptotic cell death regulated by the pan-optosome complex, known as pan-apoptosis, remains largely unexplored in this context. This study aims to illuminate the molecular properties of pan-apoptosis-related genes in glioma patients, classifying them and developing a signature using machine learning techniques.MethodsThe prognostic significance, mutation features, immunological characteristics, and pharmaceutical prediction performance of this signature were comprehensively investigated. Furthermore, GPX8, a gene of interest, was extensively examined for its prognostic value, immunological characteristics, medication prediction performance, and immunotherapy prediction potential. ResultsExperimental techniques such as CCK-8, Transwell, and EdU investigations revealed that GPX8 acts as a tumor accelerator in gliomas. At the single-cell RNA sequencing level, GPX8 appeared to facilitate cell contact between tumor cells and macrophages, potentially enhancing microglial migration. ConclusionsThe incorporation of pan-apoptosis-related features shows promising potential for clinical applications in predicting tumor progression and advancing immunotherapeutic strategies. However, further in vitro and in vivo investigations are necessary to validate the tumorigenic and immunogenic processes associated with GPX8 in gliomas

    Research on the Influence of Traction Load on Transient Stability of Power Grid Based on Parameter Identification

    The traction load of electrified railways is a special user of the power system, which will have a significant impact on and pose a challenge to the power system after grid connection. Considering the impact and fluctuation of traction load connected to the power grid, this paper proposed a method to study the influence of traction load on the transient stability of the power grid based on parameter identification. Firstly, according to the operation characteristics of traction load, a comprehensive traction load model based on the measurement-based method of induction motor parallel power function model is selected, and the objective function of parameter identification is determined. Then, the specific steps of using the improved gray wolf optimizer (IGWO) algorithm to achieve the parameter identification of the traction comprehensive load model are introduced, including chaotic map, nonlinear convergence factor, and individual position update. Next, with the IEEE39 bus system as the network background, the load parameters identified by the measured data of a line of the Beijing–Shanghai high-speed railway are imported into the Power System Analysis Software Package (PSASP) 7.0 for transient simulation to analyze the impact of traction load on the transient stability of the power grid. Finally, three typical load models are compared with the traction load model to draw relevant conclusions, and it is verified that the traction load characteristics are different from the general load model, which provides reference for the connection of electrified railway traction load to the power grid and the economic and technical construction of the power system

    Traction Load Modeling and Parameter Identification Based on Improved Sparrow Search Algorithm

    In this paper, a traction load model parameter identification method based on the improved sparrow search algorithm (ISSA) is proposed. According to the load characteristics of the AC traction power supply system under transient disturbance, the model structure of the traction load is equated to the composite load model structure of the static load shunt induction motor's dynamic load. The traditional sparrow search algorithm is improved to enhance its accuracy and convergence. The generalization ability of the model was tested, and the accuracy of the proposed model was verified. Using the ISSA to determine the load model from the measured data, the results can verify the effectiveness of the ISSA for comprehensive load model parameter identification. Comparing the ISSA with the traditional SSA and PSO algorithms, it shows that the ISSA has better accuracy and convergence

    Comparative Analysis of Free Amino Acids and Nucleosides in Different Varieties of Mume Fructus Based on Simultaneous Determination and Multivariate Statistical Analyses

    Mume Fructus (MF) contains a variety of organic acids, free amino acids, and nucleoside components, and studies have not yet analyzed the relationship between the components of free amino acids and nucleosides with the varieties of MF. A rapid and sensitive method was established for simultaneous determination of 21 free amino acids and 9 nucleosides in MF by ultrafast liquid chromatography-mass spectrometry. The analysis was carried out on a Waters XBridge Amide column (100 mm × 2.1 mm, 3.5 μm) with elution by the mobile phase of 0.2% aqueous formic acid (A) and 0.2% formic acid acetonitrile (B) at a flow rate of 0.2 mL/min with 1 μL per injection. The column temperature was maintained at 30°C. The target compounds were analyzed by the positive ion multiple reaction monitoring (MRM) mode. The comprehensive evaluation of the samples was carried out by principal component analysis (PCA) and technique for order preference by similarity to an ideal solution (TOPSIS) analysis. Results showed the method could simultaneously determine 30 components in MF. The content of total analytes in six mainstream varieties was different, exhibited the order Nangao > Daqingmei > Zhaoshuimei > Yanmei > Shishengme > Baimei, and aspartic acid and adenosine were the most abundant amino acid and nucleoside. PCA and OPLS-DA could easily distinguish the samples, and 11 components could be chemical markers of sample classification. TOPSIS implied that the quality of Nangao and Daqingmei was superior to the other varieties. The results could provide a reliable basis for quality evaluation and utilisation of medicinal and edible MF

    An integrated metabolomic approach to elucidate the mechanism of Chrysanthemi Flos processed products in ameliorating metabolic syndrome

    As an important medicinal and dietary flower tea, Chrysanthemi Flos has hypotensive, hypolipidemic and hypoglycaemic benefits. This study investigated the effects of two types of processed Chrysanthemi Flos products—oven-dried Chrysanthemi Flos (DCF) and shade-dried Chrysanthemi Flos (SCF)—on rats with metabolic syndrome (MetS) and their potential mechanisms of action. The pharmacodynamic analysis showed that DCF and SCF improved hypertension, regulated lipid profiles, and reduced liver and kidney damage. Metabolomics analysis revealed eight metabolites, including L-serine, oxaloacetate and succinate as potential biomarkers for mitigating metabolic disorders in MetS rats. Our findings provide scientific evidence for the integration of Chrysanthemi Flos-based products into clinical dietary interventions, offering insights into the mechanisms through which these products can ameliorate MetS. These results have important implications for the development of functional foods that can improve metabolic health
