    Last Men Standing: Chlamydatus Portraits and Public Life in Late Antique Corinth

    Notable among the marble sculptures excavated at Corinth are seven portraits of men wearing the long chlamys of Late Antique imperial office. This unusual costume, contemporary portrait heads, and inscribed statue bases all help confirm that new public statuary was created and erected at Corinth during the 4th and 5th centuries. These chlamydatus portraits, published together here for the first time, are likely to represent the Governor of Achaia in his capital city, in the company of local benefactors. Among the last works of the ancient sculptural tradition, they form a valuable source of information on public life in Late Antique Corinth

    Influence of antisynthetase antibodies specificities on antisynthetase syndrome clinical spectrum time course

    Antisynthetase syndrome (ASSD) is a rare clinical condition that is characterized by the occurrence of a classic clinical triad, encompassing myositis, arthritis, and interstitial lung disease (ILD), along with specific autoantibodies that are addressed to different aminoacyl tRNA synthetases (ARS). Until now, it has been unknown whether the presence of a different ARS might affect the clinical presentation, evolution, and outcome of ASSD. In this study, we retrospectively recorded the time of onset, characteristics, clustering of triad findings, and survival of 828 ASSD patients (593 anti-Jo1, 95 anti-PL7, 84 anti-PL12, 38 anti-EJ, and 18 anti-OJ), referring to AENEAS (American and European NEtwork of Antisynthetase Syndrome) collaborative group's cohort. Comparisons were performed first between all ARS cases and then, in the case of significance, while using anti-Jo1 positive patients as the reference group. The characteristics of triad findings were similar and the onset mainly began with a single triad finding in all groups despite some differences in overall prevalence. The "ex-novo" occurrence of triad findings was only reduced in the anti-PL12-positive cohort, however, it occurred in a clinically relevant percentage of patients (30%). Moreover, survival was not influenced by the underlying anti-aminoacyl tRNA synthetase antibodies' positivity, which confirmed that antisynthetase syndrome is a heterogeneous condition and that antibody specificity only partially influences the clinical presentation and evolution of this condition

    Small-angle X-ray scattering and dynamic light scattering investigations on proteins denatured by acid or by guanidine hydrochloride (proteins apo-cytochrome c, streptokinase, histon H1, staphylokinase, phosphoglycerate kinase) proved that the denatured proteins have the conformation of a perturbed random coil with repulsive potentials between the chain segments. Structural models are proposed for streptokinase and staphylokinase in solution. Both consist of compact, separately folded domains (four domains in streptokinase; two domains in staphylokinase) linked by mobil segments of the protein chain. The molecules exhibit the conformation of a flexible string-of-beds in solution by which crystallization is subdued. In the frame of this work computer programs and theoretical methods have been developed for an improved interpretation of small- and intermediate-angle X-ray scattering results.     Based on x-ray angle scattering methods, dynamical light scattering, ultra centrifugation, circular dichroism in the UV region, fluorescence spectroscopy and theoretical methods of structure prediction from amino acid sequences the structures of staphylokinase and streptokinase have been determined. Both are flexible multi-domain proteins with modules arranged in straight lines, which in solution can bloat against each other. In addition it is demonstrated that the thermostability of natural variants of the bacterial plasminogen activator staphylokinase can largely differ from each other.
