    OBJECTIVE: In the present investigation, the anticariogenic effect of fluoride released by two products commonly applied in infants was evaluated. METHODS: Bovine sound enamel blocks were randomly allocated to each one of the treatment groups: control (C), varnish (V) and diamine silver fluoride solution (D). The blocks were submitted to pH cycles in an oven at 37ºC. Next, surface and cross-sectional microhardness were assessed to calculate the percentage loss of surface microhardness (%SML) and the mineral loss (deltaZ). The fluoride present in enamel was also determined.     OBJETIVES: The aim of this study was to verify the anticariogenic effect of acidulate solutions with low NaF concentration, using pH-cycling model and bovine enamel. MATERIAL AND METHODS: Enamel blocks were submitted to the surface microhardness (SMH) test and randomly divided in 12 experimental and one placebo groups. The blocks were submitted to pH cycling for 7 days, with daily applications once/day of 0.05% NaF and 0.1% NaF and twice/day of 0.02% NaF solutions. Four different pH: 4.0, 5.0, 6.0 and 7.0 were used. Next, SMH test was again used to determine the surface microhardness percentage change (%SMH). Data obtained for %SMH were homogeneous and passed through variance analyses and Tukey's test (5%) as far as fluoride concentrations and pH. RESULTS:The results showed that pH influenced %SMH in 0.02% NaF and 0.05% NaF solutions with pH 4.0, which had less mineral loss compared to pH 7.0 (p;0.05). A dose-response relationship was observed among the tested solutions, with better anticariogenic effect for the 0.1% NaF solution. CONCLUSION: The results suggest that the addition of citric acid to acidulate mouth rinses reduce mineral loss.OBJETIVO: O objetivo do presente estudo foi verificar o efeito anticariogênico de soluções aciduladas com baixa concentração de flúor, usando um modelo de ciclagem de pH e esmalte bovino. Os blocos foram submetidos à ciclagem de pH por 7 dias e a aplicações diárias de soluções de NaF a 0,05% e 0,1% (1x) e de 0,02% (2x), com pHs 4.0, 5.0, 6.0 e 7.0. A seguir, determinou-se a SMH novamente para o cálculo da porcentagem de alteração da microdureza de superfície (%SMH). Os dados de %SMH foram homogêneos e submetidos a análise de variância e teste de Tukey (5%) considerando as concentrações de flúor e pH. RESULTADOS: O pH influenciou a %SMH nas soluções de NaF 0,02% e 0,05% com pH 4.0, no qual a perda mineral foi menor quando comparado ao pH 7.0 (p;0,05). A relação dose-resposta foi observada entre as soluções testadas, com melhor efeito anticariogênico para a solução de NaF 0,1%.     Fluoride release by restorative materials before and after a topical application of fluoride gel

    Avaliou-se a liberação de flúor de materiais restauradores (Vitremer, Ketac-Fil, Fuji II LC e Freedom) durante 2 períodos de 15 dias, antes e após a aplicação tópica de fluorfosfato acidulado (FFA). Foram confeccionados, para cada material, 6 corpos-de-prova que foram imersos em 2 ml de água deionizada. As dosagens da concentração de flúor das soluções foram realizadas em intervalos de 24 horas durante 15 dias. Após esse período, os corpos-de-prova de cada material receberam tratamento com gel de FFA durante 4 minutos. O flúor liberado foi analisado em intervalos de 24 horas durante 15 dias. A análise de variância e o teste de Tukey (p < 0,05) mostraram que a média total de flúor liberado durante os 15 dias iniciais foram maiores para o Vitremer e o Ketac-Fil e menores para o Fuji II LC e Freedom; e nos 15 dias finais apresentaram diferença de liberação, com maior valor para o Vitremer, seguido pelo Fuji II LC, Ketac-Fil e Freedom. A comparação dos resultados entre o 1º dia e o 16º dia (após aplicação do gel) mostrou uma maior liberação de flúor no 16º para o Vitremer, Fuji II LC e Freedom e igual para o Ketac-Fil. Embora todos os materiais avaliados tenham ganhado flúor pela aplicação de fluorfosfato acidulado, os dados sugerem que os cimentos de ionômero de vidro modificados por resina são mais eficientes para liberar o flúor do que os outros materiais.The release of fluoride from restorative materials (Vitremer, Ketac-Fil, Fuji II LC and Freedom) was evaluated during two 15-day periods, before and after a topical application of acidulated phosphate fluoride gel (APF). For each material, 6 specimens were made, which were immersed in 2 ml of deionized water. The fluoride concentration dosages in the solutions were read at intervals of 24 hours for 15 days. After this period, the specimens of each material received treatment with APF gel for 4 minutes and the fluoride released was analyzed at 24-hour intervals during the following 15 days. The analysis of variance and the Tukey test (p < 0.05) showed that the total mean fluoride released during the initial 15 days was greater for Vitremer and Ketac-Fil and lower for Fuji II LC and Freedom; and in the final 15 days there was a difference in release readings, with the greatest value for Vitremer, followed by Fuji II LC, Ketac-Fil and Freedom. The comparison of the results between the 1st day and the 16th day (after gel application) showed a greater fluoride release on the 16th day for Vitremer, Fuji II LC and Freedom and was equal for Ketac-Fil. Although all the materials evaluated gained fluoride with the application of APF, the data suggest that the resin-modified ionomers are more efficient in releasing fluoride to the medium than the other materials

    Comparação in vitro do efeito cariostático entre a aplicação tópica de flúor e dentifrício fluoretado

    The aim of this study was to compare the effect of topical fluoride products [acidulated phosphate fluoride (APF) or neutral gel (NF) x fluoride toothpaste (MFP)], in respect to fluoride uptake and anticariogenic action. One hundred and twenty five blocks of human teeth, sorted in 5 groups according to the treatment, were submitted to pH cycling for ten days. The parameters analyzed were: fluoride uptake before and after pH cycling and surface (SMH) and cross-sectional (CSMH) microhardness of the enamel blocks. The results of fluoride concentration in enamel after the pH cycling showed an enhancement of fluoride uptake for all groups compared to sound control. No significant differences between APF and MFP were observed for surface microhardness, percentage change of surface microhardness and mineral loss. The volume percent mineral obtained from cross-sectional microhardness demonstrated that APF has a different lesion progression rate regarding subsurface carious lesion. The results suggest that professionally applied fluoride gel or frequent fluoride application in low concentration is a positive preventive measure for the control of dental caries.O objetivo do presente estudo foi comparar in vitro o efeito da aplicação tópica de flúor na forma de gel acidulado (FFA) ou neutro (FFN) com um dentifrício fluoretado (MFP), na incorporação e ação anticariogênica do flúor. Foram utilizados 125 blocos obtidos de esmalte humano, divididos em 5 grupos de acordo com o tratamento e submetidos à ciclagem de pH durante dez dias. Analisou-se o flúor incorporado antes e depois da ciclagem de pH; a microdureza superficial (SMH) e a microdureza interna do esmalte (CSMH). Os resultados da concentração de flúor no esmalte após a ciclagem indicou uma maior quantidade de flúor incorporado para todos os grupos comparados ao grupo controle sadio. A microdureza superficial entre APF e MFP, assim como a porcentagem de alteração da microdureza superficial e a perda mineral não mostraram diferenças estatísticas. A porcentagem de volume mineral obtida a partir da microdureza em secção longitudinal demonstrou que o APF tem um padrão melhor na formação da cárie subsuperficial. Os resultados sugerem que o uso de géis acidulados ou a freqüente aplicação de flúor em baixa concentração são medidas eficazes para o controle da cárie dental

    Effects of polyphosphates and fluoride on hydroxyapatite dissolution:A pH-stat investigation

    AbstractObjectivesThis study investigated the immediate and sustained effect of sodium trimetaphosphate (TMP) and sodium hexametaphosphate (HMP) associated or not with fluoride (F) on hydroxyapatite (HA) dissolution using an erosion-like model, considering as well as the influence of salivary coating.DesignBaseline dissolution rates were determined for HA discs using a pH-stat system. In the first set of experiments, HA discs were treated with 1100μgF/mL, 1% or 8% of HMP, 1% or 8% of TMP and 1100μgF/mL associated with 1% or 8% of HMP or TMP, totaling 9 groups (n=8). In a second phase, HA discs were kept in pooled human saliva at 37°C for 2h before treatment with deionised water and 1100μgF/mL associated with 1% or 8% of HMP or TMP, totaling 5 groups (n=8). The post-treatment dissolution rate was determined from three consecutive 30-min assays. Data were analysed using 2 and 3-way ANOVA followed by Fisher and Holm–Sidak methods, respectively (α=0.05).ResultsAll test solutions promoted reduction in HA dissolution rate when compared to baseline control in the first post-treatment run (p<0.001). However, a synergistic effect was only observed between fluoride and 1% HMP. Moreover, the duration of inhibitory effect was greater when 8% HMP and 1 or 8% HMP associated with F were assessed (p<0.001). The presence of salivary coating led to higher protection for all groups when compared to discs without coating (p<0.001).ConclusionThe reduction of HA dissolution rate, as well as the duration of this effect were influenced by fluoride, type and concentration of phosphate salt and the presence of a salivary coating

    Fluoride Concentration of Some Brands of Fermented Milks Available in the Market

    Objectives: To evaluate the fluoride ion concentration in some fermented milks present in the market. Methods: Three brands of 6 fermented milks (Parmalat®-uva, Chamyto®, Paulista®, Batavito®, Yakult®, Vigor Club®) were analyzed. Fluoride concentration was evaluated after facilitated microdiffusion by HDMS. Results: Parmalat® products ranged from 0.022 μgF/g to 0.031 μgF/g, Chamyto® from 0.228 μgF/g to 0.272 μgF/g, Paulista® from 0.182 μgF/g to 0.220 μgF/g, Batavito® from 0.028 μgF/g to 0.030 μgF/g, Yakult® from 0.115 μgF/g to 0.206 μgF/g and Vigor Club® from 0.808 μgF/g to 1.171 μgF/g. Conclusions: The presence of fluoride could be observed in all of the fermented milks analyzed which can contribute with the total fluoride daily intake

    Several pathologies have been diagnosed in children of hypertensive mothers; however, some studies that evaluated the alterations in their oral health are not conclusive. This study analyzed the salivary gland activity and dental mineralization of offsprings of spontaneously hypertensive rats (SHR). Thirty-day-old SHR males and Wistar rats were studied. The salivary flow was evaluated by injection of pilocarpine, the protein concentration and salivary amylase activity, by the Lowry method and kinetic method at 405 nm, respectively. Enamel and dentin mineralization of the mandibular incisors was quantified with aid of the microhardness meter. The results were analyzed by the ANOVA or Student's t test (pDiversas patologias têm sido diagnosticadas em filhos de mães hipertensas, entretanto alguns estudos que avaliaram as possíveis alterações na saúde oral não são conclusivos.     New insights into the anti-erosive property of a sugarcane-derived cystatin: different vehicle of application and potential mechanism of action

    A new sugarcane-derived cystatin (CaneCPI-5) showed anti-erosive properties when included in solutions and strong binding force to enamel, but the performance of this protein when added to gel formulations and its effect on surface free energy (SFE) requires further studies. Objective: 1) to evaluate the protective effect of gels containing different concentrations of CaneCPI-5 against initial enamel erosion (Experiment 1); and 2) to analyze the SFE (γS) after treating the enamel surface with CaneCPI-5 solution (Experiment 2). Methodology: In Experiment 1, 75 bovine enamel specimens were divided into five groups according to the gel treatments: placebo (negative control); 0.27%mucin+0.5%casein (positive control); 0.1 mg/mL CaneCPI-5; 1.0 mg/mL CaneCPI-5; or 2.0 mg/mL CaneCPI-5. Specimens were treated with the gels for 1 min, the AP was formed (human saliva) for 2 h and the specimens were incubated in 0.65% citric acid (pH=3.4) for 1 min. The percentage of surface hardness change (%SHC) was estimated. In Experiment 2, measurements were performed by an automatic goniometer using three probing liquids: diiodomethane, water and ethylene glycol. Specimens (n=10/group) remained untreated (control) or were treated with solution containing 0.1 mg/mL CaneCPI-5, air-dried for 45 min, and 0.5 µL of each liquid was dispensed on the surface to measure contact angles. Results: Gels containing 0.1 and 1.0 mg/mL CaneCPI-5 significantly reduced %SHC compared to the other treatments (p&lt;0.05). Treated enamel showed significantly lower γS than control, without changes in the apolar component (γSLW), but the polar component (γSAB=Lewis acid-base) became more negative (p&lt;0.01). Moreover, CaneCPI-5 treatment showed higher γS - (electron-donor) values compared to control (p&lt;0.01). Conclusions: Gels containing 0.1 mg/mL or 1.0 mg/mL CaneCPI-5 protected enamel against initial dental erosion. CaneCPI-5 increased the number of electron donor sites on the enamel surface, which may affect AP formation and could be a potential mechanism of action to protect from erosion.&nbsp

    In vitro assessment of an experimental coat applied over fluoride varnishes

    OBJECTIVE: The time of contact between the product and enamel surface is important in ensuring the efficacy of fluoride varnishes. Thus, some alternatives could avoid fluoride loss to saliva and improve the anticariogenic action of the product. This study evaluated the effect of an experimental coat on the anticariogenic action of fluoride varnishes. MATERIAL AND METHODS: Enamel bovine blocks were selected by evaluating surface microhardness and randomized into five groups (n=24): placebo, DuraphatTM, DuraphatTM with coat, DuofluoridTM and DuofluoridTM with coat. Twelve blocks from each group were used to analyze calcium fluoride (CaF2) formed on enamel after treatment. The other 12 blocks were subjected to pH cycling for 7 days. The varnishes were kept on enamel for 6 h. Next, the percentage change of surface microhardness (%SMHC) and mineral loss (&#916;Z) were calculated. CaF2 retained and fluoride present in the pH-cycled solutions were also measured. RESULTS: The use of the coat did not decrease %SMHC and &#916;Z, but all fluoride varnishes had better results when compared to the placebo (ANOVA and Kruskal-Wallis, respectively).     Avaliação do teor de flúor e pH em diferentes soluções para bochechos no mercado nacional

    In this study, the fluoride concentration and pH of 14 commercial brands of mouthrinses were assessed in order to compare them with the values expressed on the labels and with those established by the National Sanitary Surveillance Agency. Forty-two (42) products were obtained from three places, with different manufacturing batches. The fluoride concentration was determined in diluted solutions, using a combined specific electrode for fluoride ion (9609 BN Orion Research) and ion analyzer (290 A Orion Research). The results showed that 50% of the solutions had statistically significant differences, with higher fluoride concentrations than those expressed on the labels, and the pH ranged from 4.23 to 7.34, but only one of the products registered the pH value on its label.No presente estudo, o teor de flúor e o pH de 14 marcas comerciais de soluções para bochechos foram avaliados a fim de comparar com os valores expressos nos rótulos e com aqueles determinados pela Agência Nacional de Vigilância Sanitária. Foram adquiridos 42 produtos em três localidades, com diferentes lotes de fabricação. A concentração de flúor foi determinada em soluções diluídas, utilizando-se eletrodo específico combinado para íon flúor (9609 BN Orion Research) e analisador de íons (290 A Orion Research).
