Their biochemical responses to L-arginine and when they dimerize could help clarify this pathway, so I sought to develop a periplasmic dimerization sensor to better monitor these biochemical interactions. Similar to STM1987, the ToxR transcriptional regulator from Vibrio cholera is also activated by dimerization. By switching out the periplasmic domain of ToxR for the periplasmic regions of interest, I can better evaluate the cyclic-di-GMP response to L-arginine. This research aims to find the specific responses in this pathway to be able to use this in combatting bacterial virulence. I was able to successfully show that the STM1987 periplasmic domain dimerizes in response to L-arginine, providing an important insight into this signaling pathway

    Constitutional Implications on Block Pricing in the California Water Market

    Due to the severity of the recent drought, those residing in California have realized the devastating effects of climate change. As California continues to experience amplified weather conditions and diminished water supply, the state must adapt to drought conditions and mitigate the impacts of droughts. In order to do so, municipal water pricing mechanisms that send price signals to consumers to conserve water should be enforced to ensure the longevity of California\u27s water supply. In particular, increasing block-pricing structures are a favorable market-based solution that disincentivizes heavy water consumption while allowing essential water use to remain affordable. The California Constitution limits conservation water pricing to the at-cost requirement where municipalities must prove that rates reflect the actual cost of service. This Note looks at the constitutional and judicial limitations on municipal block-pricing structures in light of the recent Fourth District Court of Appeals decision, Capistrano Taxpayers Association, Inc. v. City of San Juan Capistrano, and concludes that the case casts constitutional doubt on tiered price structures because the opinion: (1) embarked on a lengthy and unnecessary discussion on the nexus of Proposition 218 and Article X, section 2 of the California Constitution; (2) relies on incomparable precedent in analyzing conservation prices and the at-cost requirement; (3) inconsistently uses the term true cost; (4) fundamentally fails to understand the nature and scope of the duties of water service providers; and (5) did not limit its discussion of penalty rates to the facts of the case. Despite these shortcomings, block water pricing structures remain constitutional so long as the municipality proves this at-cost requirement at each corresponding level of usage. This Note looks at Capistrano\u27s implications on municipal block-pricing structures, and explains how municipalities can continue to implement these pricing mechanisms within the judicial limitations set forth in Capistrano

    Tin dioxide nanoparticle based sensor integrated with microstrip antenna for passive wireless ethylene sensing

    In this dissertation, we present the development and integration of a passive ethylene gas sensor with triangular microstrip patch antenna for wireless monitoring of climacteric fruit freshness. The existing ethylene sensors are mostly SnO2 resistor based active sensors, fabricated on rigid substrates requiring high fabrication temperatures and cannot be used for wireless applications. The proposed passive ethylene gas sensor is a novel nanoparticle based SnO2 capacitive sensor which, unlike the other existing SnO2resistor based active thick film and thin film sensors, consists of 10 nm to 15 nm SnO2 nanoparticles coated as a thin dielectric film of 1300 nm thickness. The nanoscale particle size and film thickness of the sensing dielectric layer in the capacitor model aids in sensing ethylene at room temperature and eliminates the need for micro hotplates used in existing SnO2 based resistive sensors. In comparison to the high sintering deposition temperatures used for many currently available ethylene sensors fabricated on rigid substrates, the SnO2 sensing layer is deposited using a room temperature dip coating process on flexible polyimide substrates. The capacitive sensor fabricated with pure SnO2 nanoparticles as the dielectric showed a 5 pF change in capacitance when ethylene gas concentration was increased from 0 to 100 ppm. The change in capacitance was increased to 7 pF by introducing a 10 nm layer of platinum (Pt) and palladium (Pd) alloy deposited by sputter deposition. This also improved the selectivity of the sensor to ethylene mixed in a CO2 gas environment. The response time was decreased to 3 min for SnO2 samples with Pt/Pd layer (5 min for pure SnO2samples) and its recovery time was decreased to 5 min compared to 7 min for pure SnO2 samples. The passive SnO2 capacitive ethylene sensor is integrated with a triangular microstrip patch antenna using capacitively loaded integration methodology which represents a one of a kind passive wireless sensor tag used for detecting freshness of climacteric fruit. The integration methodology adapted also reduced the size of the triangular patch antenna by 63 percent. The decrease in sensor capacitance due to the presence of ethylene (0 to 100 ppm) changes the antenna resonant frequency by 7 MHz and return loss by 9.5 dB, which makes the system reliable for far field wireless ethylene monitoring applications. The sensor tag output was also detected using an RFID reader showing a change in demodulated signal amplitude of 3 mV. Experimental result is presented for detecting multiple sensor tags at varying distances based on the wireless measurement of return loss which eliminates the common distance problem existing in backscatter signal based tags

It restricts the most important segment of society from demonstrating their full potential and their right to work and attain equal opportunity. The quantum of this behaviour in a society can only be evaluated when women in general are fully aware of the nature of this behaviour and the legal protection available. Sexual harassment is an ambiguous term for many; when hearing of an incident of sexual harassment, many might not understand exactly what crime has been committed. Under which category does it fall? What if the complainant is lying? Pakistan has enacted special and general laws to fight the epidemic of sexual harassment. This article presents the appraisal and implementation of all the prevailing laws related to sexual harassment of women in Pakistan. It examines the hurdles, restraints, and resistance women have faced during the course of adjudication and determination of such matters due to various factors including social barricades that persist in this society. This qualitative and quantitative study examined whether the prevailing laws sufficiently cover the full nature, scope, and underlying themes of sexual harassment through implementation, by analyzing the procedures adopted at the time of implementations. I also explored whether sexual harassment is really a form of discrimination and what prevailing laws should entail to combat the practice and whether the law is generally accepted by the social community because of certain intrinsic differences. The study also recommended changes and additions to the existing methods and procedures in the laws, policies, and guidelines and implementations and examined how rampant sexual harassment is in the workplace, and whether employers address harassment through policies, training, and acting on complaints
