    Immuno-Cytochemistry with Backscattered Electrons

    Some cytochemical reaction products are visible inside the cytoplasm of cells observed with the scanning electron microscope (SEM) using the backscattered electron imaging (BEI) mode. Methods can be utilized whenever they result in the deposition of heavy metal, like silver, lead or osmium at the sites of the enzymatic reaction. More recently the BEI mode of the SEM has been demonstrated to improve the detection of immunogold labeled cell surface antigens. Colloidal gold particles, 40 to 15 nm in diameter can be efficiently used for immuno-specific labeling. Moreover, cytochemical reactions can be applied to previously immunogold labeled cells, therefore combining the results of enzyme cytochemistry and of surface labeling at the level of each individual cell. The choice of fixative, incubation media, dehydration and drying methods should be guided by considerations on the sample characteristics for optimal electron scattering. Cytochemical as well as immuno-labeling reactions are not used per se but in combination with the study of cell surface morphology which needs, therefore, to be sufficiently well preserved. Coating should provide good conductivity and secondary electron emission, while emitting a minimal number of backscattered electrons. The application of these methods considerably enhances our capacity to characterize with the SEM the surface morphology of precisely identified subpopulations of many cell types

    FM Continuous Monitoring of Intraocular Pressure, an Engineering Perspective

    This chapter discusses the problem of continuously monitoring intraocular pressure (IOP) from an engineering perspective. It is aimed to all public in general although we think that medical staff and engineers may benefit the most from it. Although equations are included for engineers to get a glimpse of how the system works, this chapter does not go into great detail in mathematics and physics to make it understandable to medical staff. It provides though references for engineers who wish to get a better understanding of key subjects tackled in this chapter. The chapter is organized as follows: Section 1 introduces intraocular pressure (IOP) and need for its continuous monitoring. Section 2 describes the most recent efforts to develop a continuous IOP monitoring system. Section 3 shows what medical and engineering considerations must be taken into account to effectively measure IOP. Section 4 deals with health issues due to tissue warming and how to prevent them. Section 5 explains how an implant can be fabricated using either passive electronic components or active ones. Finally, Section 6 explains how the pressure sensor and the electronic circuits can be integrated

    Light Ions Response of Silicon Carbide Detectors

    Silicon carbide (SiC) Schottky diodes 21 mum thick with small surfaces and high N-dopant concentration have been used to detect alpha particles and low energy light ions. In particular 12C and 16O beams at incident energies between 5 and 18 MeV were used. The diode active-region depletion-thickness, the linearity of the response, energy resolution and signal rise-time were measured for different values of the applied reverse bias. Moreover the radiation damage on SiC diodes irradiated with 53 MeV 16O beam has been explored. The data show that SiC material is radiation harder than silicon but at least one order of magnitude less hard than epitaxial silicon diodes. An inversion in the signal was found at a fluence of 10^15 ions/cm^2.Comment: 20 pages, 16 figures, submitted for publication to Nuclear Instruments and Methods in Physics Research

    The backscattered electron imaging (BEI) mode of scanning electron microscopy (SEM) has been applied to study various histo-cytochemical reactions in biological specimens since the early seventies. Due to numerous, recent technical improvements the BEI mode of SEM now belongs to the routine of many SEM laboratories. For cytochemistry, BEI has been mainly used to: visualize intracellular structures and organelles; recognize the different cell types in heterogeneous populations or tissues; study the correlations between enzymatic activities and cell surface features. We have evaluated the most relevant results obtained in the study of blood cells and the possible future applications of these techniques
