    An Experimental Examination of the Assignment Problem

    The problem of optimally assigning individuals to heterogeneous objects so that each individual is allocated at most one object (the assignment problem) has a long history. Algorithms• •based •on •ordinal preferences have been developed and several auctions using monetary transfers have been proposed. The performance of two auction mechanisms to solve the assignment problem is examined in an experimental setting. One of the auctions is a sealed-bid variant of the Vickrey auction for homogeneous objects and the other auction is an extension of the English auction. The auctions are tested in two diverse competitive environments (high and low contention). The experimental results show that the English auction generates higher revenues and efficiencies than its sealed-bid counterpart especially if there is a high level of contention. However, the efficiency gains of the English auction are at the expense of consumers' surplus. Indeed, a random assignment creates greater consumers' surplus relative to either auction outcomes in the high contention environment

    BACKGROUND: Broad patterns of HIV epidemiology are frequently used to design generic HIV programs in sub-Saharan Africa. METHODS: We reviewed the epidemiology of HIV among young people in sub-Saharan Africa, and explored the unique dynamics of infection in its different regions. RESULTS: In 2009, HIV prevalence among youth in sub-Saharan Africa was an estimated 1.4% in males and 3.4% in females, but these values mask wide variation at regional and national levels. Within countries there are further major differences in HIV prevalence, such as by sex, urban/rural location, economic status, education, or ethnic group. Within this highly nuanced context, HIV prevention programs targeting youth must consider both where new infections are occurring and where they are coming from. CONCLUSIONS: Given the epidemiology, one-size-fits-all HIV prevention programs are usually inappropriate at regional and national levels. Consideration of local context and risk associated with life transitions, such as leaving school or getting married, is imperative to successful programming for young people

    Recent Establishment of the Asian Tiger Mosquito (Aedes albopictus) in Independence County, Arkansas

    Three adult Aedes albopictus were collected on 10 August 1993 within the city limits of Batesville, Arkansas. This is the firs tknown report of this species from Independence County. Subsequent investigation revealed the presence of at least three well established populations in the county, two of which were monitored on a monthly basis from Aprilto September of 1994. Peak A. albopictus numbers were recorded in July and August when the population levels were sufficient to make this mosquito a significant pest in Batesville. Our observations suggest that the Asian tiger mosquito will likely colonize other similar communities in Arkansas

    On Simulating Concurrent Flame Spread in Reduced Gravity by Reducing Ambient Pressure

    The flammability of combustible materials in spacecraft environments is of importance for fire safety applications because the environmental conditions can greatly differ from those on earth, and a fire in a spacecraft could be catastrophic. Moreover, experimental testing in spacecraft environments can be difficult and expensive, so using ground-based tests to inform microgravity tests is vital. Reducing buoyancy effects by decreasing ambient pressure is a possible approach to simulate a spacecraft environment on earth. The objective of this work is to study the effect of pressure on material flammability, and by comparison with microgravity data, determine the extent to which reducing pressure can be used to simulate reduced gravity. Specifically, this work studies the effect of pressure and microgravity on upward/concurrent flame spread rates and flame appearance of a burning thin composite fabric made of 75% cotton and 25% fiberglass (Sibal). Experiments in normal gravity were conducted using pressures ranging between 100 and 30 kPa and a forced flow velocity of 20 cm/s. Microgravity experiments were conducted during NASAs Spacecraft Fire Experiment (Saffire), on board of the Orbital Corporation Cygnus spacecraft at 100 kPa and an air flow velocity of 20 cm/s. Results show that reductions of ambient pressure slow the flame spread over the fabric. As pressure is reduced, flame intensity is also reduced. Comparison with the concurrent flame spread rates in microgravity show that similar flame spread rates are obtained at around 30 kPa. The normal gravity and microgravity data is correlated in terms of a mixed convection non-dimensional parameter that describes the heat transferred from the flame to the solid surface. The correlation provides information about the similitudes of the flame spread process in variable pressure and reduced gravity environments, providing guidance for potential on-earth testing for fire safety design in spacecraft and space habitats

    Tissue-selective expression of a conditionally-active ROCK2-estrogen receptor fusion protein

    The serine/threonine kinases ROCK1 and ROCK2 are central mediators of actomyosin contractile force generation that act downstream of the RhoA small GTP-binding protein. As a result, they have key roles in regulating cell morphology and proliferation, and have been implicated in numerous pathological conditions and diseases including hypertension and cancer. Here we describe the generation of a gene-targeted mouse line that enables CRE-inducible expression of a conditionally-active fusion between the ROCK2 kinase domain and the hormone-binding domain of a mutated estrogen receptor (ROCK2:ER). This two-stage system of regulation allows for tissue-selective expression of the ROCK2:ER fusion protein, which then requires administration of estrogen analogues such as tamoxifen or 4-hydroxytamoxifen to elicit kinase activity. This conditional gain-of-function system was validated in multiple tissues by crossing with mice expressing CRE recombinase under the transcriptional control of cytokeratin14 (K14), murine mammary tumor virus (MMTV) or cytochrome P450 Cyp1A1 (Ah) promoters, driving appropriate expression in the epidermis, mammary or intestinal epithelia respectively. Given the interest in ROCK signaling in normal physiology and disease, this mouse line will facilitate research into the consequences of ROCK activation that could be used to complement conditional knockout models

    Flagellar membrane association via interaction with lipid rafts

    The eukaryotic flagellar membrane has a distinct composition from other domains of the plasmalemma. Our work shows that the specialized composition of the trypanosome flagellar membrane reflects increased concentrations of sterols and saturated fatty acids, correlating with direct observation of high liquid order by laurdan fluorescence microscopy. These findings indicate that the trypanosome flagellar membrane possesses high concentrations of lipid rafts: discrete regions of lateral heterogeneity in plasma membranes that serve to sequester and organize specialized protein complexes. Consistent with this, a dually acylated Ca(2+) sensor that is concentrated in the flagellum is found in detergent-resistant membranes and mislocalizes if the lipid rafts are disrupted. Detergent-extracted cells have discrete membrane patches localized on the surface of the flagellar axoneme, suggestive of intraflagellar transport particles. Together, these results provide biophysical and biochemical evidence to indicate that lipid rafts are enriched in the trypanosome flagellar membrane, providing a unique mechanism for flagellar protein localization and illustrating a novel means by which specialized cellular functions may be partitioned to discrete membrane domains
