    Statistička analiza obrade podataka u nekim seizmičkim refrakcijskim metodama: Primjer sintetičkih podataka

    The delay time method has gained attention in shallow seismic refraction survey because it has the capability to map the lateral thickness of overburden and relief of bedrock. This study addresses the comparison between the performances of the plus-minus and conventional reciprocal methods using a synthetic data. The interpretations obtained from both methods are reasonably comparable to the actual geophysical models. This suggests that either of the methods can be used to construct a geologic section. However, the result of randomized complete block design (RCBD) experiment shows a significant difference in the type of method used and this necessitate the need for further test. The pairwise comparison suggests that the plus-minus method produces a model that better mimics the actual data than the conventional reciprocal method.Metoda vremenskog kašnjenja često se koristi u istraživanju plitke seizmičke refrakcije jer ima mogućnost kartiranja lateralne debljine nadsloja i reljefa stijena. Ovaj rad bavi se usporedbom svojstava plus-minus i konvencionalnih recipročnih metoda korištenjem sintetičkih podataka. Interpretacije dobivene objema metodama razumno su usporedive sa stvarnim geofizičkim modelima. To sugerira da se bilo koja od metoda može koristiti za konstrukciju geološke sekcije. Međutim, rezultat eksperimenta randomiziranog cjelovitog projekta blokiranja (RCBD) pokazuje značajne razlike ovisno o korištenoj metodi, a to nameće potrebu za daljnjim ispitivanjima. Uparena usporedba sugerira da plus-minus metoda daje model koji bolje oponaša stvarne podatke u odnosu na konvencionalnu recipročnu metodu

    Review of immunohistochemical typing of endometrial carcinoma at the Lagos University Teaching Hospital

    Background: Categorization of endometrial carcinomas as type I and II provides useful insights into their different risk factors, pathogenesis and biologic behaviours. Aim: To determine the immunohistochemical classifications of endometrial carcinomas in Nigerian women. Design: A retrospective review of histopathologic slides of cases of endometrial carcinomas seen at the Lagos University Teaching Hospital (LUTH) over a 5-year period. The slides were reviewed, and the diagnoses made according to the WHO nomenclature. The classification of endometrial carcinomas into Type I and II was made by immunohistochemistry using antibodies to ER, PR, p53 and Ki-67. Results: Eight cases of endometrial adenocarcinoma were reported accounting for 53.3% of all endometrial malignancies. Of these, only 1 case showed the classic type I immunophenotype while type II staining pattern was seen in 4 cases. The remaining 3 cases had equivocal immunophenotypes: one was p53+ but showed ER+, PR+ and high Ki-67 index; the second was p53-, ER+, PR+ but had a high Ki-67 expression; while the last was p53-, but ER-, PR- and had high Ki-67 expression. Conclusion: Endometrial carcinomas in Nigerian women are more likely to be type II carcinomas. A reasonable proportion of the cases were equivocal thus requiring further categorization with molecular studies

    Collaborative Molecular Epidemiology Study of Metabolic Dysregulation, DNA Methylation, and Breast Cancer Risk Among Nigerian Women: MEND Study Objectives and Design

    PURPOSE To elucidate the role of metabolic dysregulation and associated DNA methylation changes on breast cancer risk and aggressive subtypes among Nigerian women. We describe the design and methods of a collaborative molecular epidemiology study of breast cancer in Nigerian hospitals. METHODS The Mechanisms for Novel and Established Risk Factors for Breast Cancer in Women of Nigerian Descent (MEND) study was designed as a matched case-control study of 350 patients, age 18 to 75 years, with newly diagnosed, treatment-naïve breast cancer and 350 age-matched healthy controls from surrounding geographic areas. Patients with breast cancer seen for initial diagnosis at four large tertiary hospitals in southwest Nigeria and one affiliated private hospital were recruited. Healthy female controls were selected from a cohort of 4,000 healthy women recruited as part of the Human Heredity and Health (H3) in Africa Chronic Kidney Disease Case-Control Study in Nigeria. Tumor and adjacent normal tissue, and blood and saliva samples were collected for molecular and epigenetic assays. RESULTS Although recruitment is ongoing, a total of 416 patients have been recruited to date, with tumor and blood samples obtained from at least 310 patients. Data on age-matched (± 6 months) controls have also been obtained and harmonized. Lipid assays for 350 pathologically verified cases and 350 age-matched controls is underway, and pathologic characterization of tumors (including immunohistochemistry for subtyping) is ongoing. Data on DNA methylation for tumors and adjacent normal tissue are expected by the end of the study period. CONCLUSION The MEND study will provide a unique, high-quality source of data to evaluate the contribution of metabolic dysregulation such as obesity, diabetes, hypertension, and metabolic syndrome to the biology of breast cancer among Nigerian women and foster collaborative studies relevant for women of African descent globally

    Association of Body Composition with Odds of Breast Cancer by Molecular Subtype: Analysis of the Mechanisms for Established and Novel Risk Factors for Breast Cancer in Nigerian Women (MEND) Study

    BACKGROUND: The association between obesity and breast cancer (BC) has been extensively studied among US, European and Asian study populations, with often conflicting evidence. However, despite the increasing prevalence of obesity and associated conditions in Africa, the continent with the highest age-standardized BC mortality rate globally, few studies have evaluated this association, and none has examined in relation to molecular subtypes among African women. The current analysis examines the association between body composition, defined by body mass index (BMI), height, and weight, and BC by molecular subtype among African women. METHODS: We estimated odds ratios (ORs) and 95% confidence intervals (95% CI) for the association between measures of body composition and BC and molecular subtypes among 419 histologically confirmed cases of BC and 286 healthy controls from the Mechanisms for Established and Novel Risk Factors for Breast Cancer in Women of Nigerian Descent (MEND) case-control study. RESULTS: Higher BMI (aOR: 0.79; 95% CI: 0.67, 0.95) and weight (aOR: 0.83; 95% CI: 0.69, 0.98) were associated with reduced odds of BC in adjusted models, while height was associated with non-statistically significant increased odds of BC (aOR: 1.07, 95% CI: 0.90, 1.28). In pre/peri-menopausal, but not post-menopausal women, both higher BMI and weight were significantly associated with reduced odds of BC. Further, higher BMI was associated with reduced odds of Luminal A, Luminal B, and HER2-enriched BC among pre/peri-menopausal women, and reduced odds of triple-negative BC among post-menopausal women. CONCLUSIONS: Higher BMI and weight were associated with reduced odds of BC overall and by molecular subtype among West African women. Larger studies of women of African descent are needed to definitively characterize these associations and inform cancer prevention strategies

    Acute toxicity effect of Croton penduliflorus (Euphorbiaceae) Seed Oil

    Background; Croton penduliforus (Euphorbiaceae) is a tropical evergreen plant widely distributed in Africa. Its seeds are used in folklore medicine as laxative, as well as a major component of herbal contraceptive and antifibroid decoction.Objective: This study is to investigate the oral acute toxicity and histological effects of Croton penduliflorus seed oil in mice.Methods: Croton penduliflorus seed oil (CPSO) was prepared by Soxhlet extraction of shelled, oven dried, ground seed sample with 40-60°C petroleum ether. Albino mice of both sexes aged 6-7 weeks old were randomly divided into seven groups of five. Graded doses of 200, 400, 600, 800, 1000 and 1200 mg/kg body weight of the extract were administered orally to groups 2-7 respectively. The control group was administered with 0.1 ml of Tween 20. The numbers of deaths over a period of 24 hours were recorded. Acute toxicity (lethal dose) was estimated from the graph of % cumulative death against log dose of the extract. The animals that survived after 24 hours were monitored daily for 14 days for appearance of delayed toxicity signs. At the end of 14 days all the animals were sacrificed and blood sample were collected from each animal into a plain sterilized bottle. Internal organs namely kidney, liver, heart and lungs were isolated and fixed in 10 % formal saline for histopathological examination.Results: The LD50  was estimated to be 570 mg/kg body weight. CPSO caused weight loss at doses greater than 600 mg/kg with significant increases in AST and ALP activities and fluctuation of serum electrolytes.Conclusion: Croton penduliflorus seed oil is toxic to the kidney and liver of mice. Keywords: Croton penduliflorus, seed, oral acute toxicity, mice

    Correlation between Fine-Needle Aspiration Cytology and Histology for Palpable Breast Masses in a Nigerian Tertiary Health Institution

    Background. Management of breast lumps can be challenging in resource poor settings. Fine-needle aspiration cytology (FNAC) especially when used with cell block can help improve affordability for the patients. Objective. To determine the diagnostic accuracy of FNAC of palpable breast lesions within a 5-year period. Methods. The findings obtained from FNAC of palpable breast lumps seen at the FNAC clinic of our department from January 2007 to December 2011 were retrieved and correlated with findings on histology of excisional biopsies. Results. A total of 1790 patients had FNAC of breast lumps during the 5-year period; 436 of them subsequently had biopsies. Our results compare favourably with the measures of test performance of the UK NHS Breast Screening Programme shown in brackets: absolute sensitivity 95.4% (>70%), complete sensitivity 99.2% (>90%), full specificity 88.9% (>65%), positive predictive value 99.6% (>99%), false-negative rate 0.8% (<4%), false-positive rate 0.4% (<0.5%), inadequate rate 3.2% (<15%), and suspicious rate 10.2% (<15%). Conclusion. Breast FNACs compare very well with histology of excisional biopsies and in experienced hands are extremely useful in the management of breast lumps. Further studies assessing the diagnostic accuracy of FNAC and cell blocks in our setting are recommended
