Real-Time Particle Radiography by Means of Scintillating Fibers Tracker and Residual Range Detectors
In this chapter, a detailed description of the construction and the procedure for the measurement of performances of a charged particle imaging system is given. Such a system can be realized by the combined use of a position sensitive detector and a residual range detector. The position sensitive detector is made up of two superimposed and right-angled planes, each of which subsists of two layers of pre-aligned and juxtaposed scintillating fibers. The selected 500Â Îźm square section fibers are optically coupled to two silicon photomultiplier arrays adopting a channel reduction system patented by the Istituto Nazionale di Fisica Nucleare. The residual range detector consists of 60 parallel layers of the same fibers used in the position detector, each of which is optically coupled to a channel of silicon photomultiplier array by means of two wavelength-shifting fibers. The sensitive area of both detectors is 90 Ă 90Â mm2. The performance of the prototypes was tested in different facilities with protons and carbon ions at energy up to about 250Â MeV and rate up to about 109 particles per second. The comparison between simulations and measurements confirms the validity of this system. In vitro and in vivo inhibition of breast cancer cell growth by targeting the Hedgehog/GLI pathway with SMO (GDC-0449) or GLI (GANT-61) inhibitors.
Aberrant Hedgehog (Hh)/glioma-associated oncogene (GLI) signaling has
been implicated in cancer progression. Here, we analyzed GLI1, Sonic Hedgehog
(Shh) and NF-ÎşB expression in 51 breast cancer (ductal carcinoma) tissues using
immunohistochemistry. We found a positive correlation between nuclear GLI1
expression and tumor grade in ductal carcinoma cases. Cytoplasmic Shh staining
significantly correlated with a lower tumor grade. Next, the in vitro effects of two
Hh signaling pathway inhibitors on breast cancer cell lines were evaluated using the
Smoothened (SMO) antagonist GDC-0449 and the direct GLI1 inhibitor GANT-61.
GDC-0449 and GANT-61 exhibited the following effects: a) inhibited breast cancer
cell survival; b) induced apoptosis; c) inhibited Hh pathway activity by decreasing
the mRNA expression levels of GLI1 and Ptch and inhibiting the nuclear translocation
of GLI1; d) increased/decreased EGFR and ErbB2 protein expression, reduced p21-
Ras and ERK1/ERK2 MAPK activities and inhibited AKT activation; and e) decreased
the nuclear translocation of NF-ÎşB. However, GANT-61 exerted these effects more
effectively than GDC-0449. The in vivo antitumor activities of GDC-0449 and GANT-
61 were analyzed in BALB/c mice that were subcutaneously inoculated with mouse
breast cancer (TUBO) cells. GDC-0449 and GANT-61 suppressed tumor growth of
TUBO cells in BALB/c mice to different extents. These findings suggest that targeting
the Hh pathway using antagonists that act downstream of SMO is a more efficient
strategy than using antagonists that act upstream of SMO for interrupting Hh signaling
in breast cancer
Experimental study for evaluation of a suitable ground displacement monitoring system: Pilot hole Campi Flegrei Deep Drilling Project case
The paper presents an experimental study carried out in 2012 during the drilling activity for a pilot hole performed in the framework of the Campi Flegrei Deep Drilling Project. A monitoring network has been installed to test and choose a suitable ground deformation system for the subsequent deep drilling of about 3.5 km in the Campi Flegrei Caldera (Italy). We describe the seismic network installed to characterize the structure of the pilot hole area and the ground deformation monitoring system chosen for the small drilling area. Data analysis and results obtained indicate that Total Station is a suitable tool for this case.Published4V. Dinamica dei processi pre-eruttiviN/A or not JC

BCL11A enhancer edited hematopoietic stem cells persist in rhesus monkeys without toxicity
Gene editing of the erythroid-specific BCL11A enhancer in hematopoietic stem and progenitor cells (HSPCs) from sickle cell disease (SCD) patients induces fetal hemoglobin (HbF) without detectable toxicity as assessed by mouse xenotransplant. Here, we evaluated autologous engraftment and HbF induction potential of erythroid-specific BCL11A enhancer edited HSPCs in four non-human primates. We utilized a single guide RNA (sgRNA) with identical human and rhesus target sequences to disrupt a GATA1 binding site at the BCL11A +58 erythroid enhancer. Cas9 protein and sgRNA ribonucleoprotein complex (RNP) was electroporated into rhesus HSPCs, followed by autologous infusion after myeloablation. We found that gene edits persisted in peripheral blood (PB) and bone marrow (BM) for up to 101 weeks similarly for BCL11A enhancer or control locus (AAVS1) targeted cells. Biallelic BCL11A enhancer editing resulted in robust gamma-globin induction, with the highest levels observed during stress erythropoiesis. Indels were evenly distributed across PB and BM lineages. Off-target edits were not observed. Non-homologous end-joining repair alleles were enriched in engrafting HSCs. In summary, we find that edited HSCs can persist for at least 101 weeks post-transplant, and biallelic edited HSCs provide substantial HbF levels in PB red blood cells, together supporting further clinical translation of this approach
NURE: An ERC project to study nuclear reactions for neutrinoless double beta decay
Neutrinoless double beta decay (0νββ) is considered the best potential resource to
access the absolute neutrino mass scale. Moreover, if observed, it will signal that neutrinos are
their own anti-particles (Majorana particles). Presently, this physics case is one of the most
important research âbeyond Standard Modelâ and might guide the way towards a Grand
Unified Theory of fundamental interactions.
Since the 0νββ decay process involves nuclei, its analysis necessarily implies nuclear structure
issues. In the NURE project, supported by a Starting Grant of the European Research Council
(ERC), nuclear reactions of double charge-exchange (DCE) are used as a tool to extract
information on the 0νββ Nuclear Matrix Elements. In DCE reactions and ββ decay indeed the
initial and final nuclear states are the same and the transition operators have similar structure.
Thus the measurement of the DCE absolute cross-sections can give crucial information on ββ
matrix elements. In a wider view, the NUMEN international collaboration plans a major
upgrade of the INFN-LNS facilities in the next years in order to increase the experimental
production of nuclei of at least two orders of magnitude, thus making feasible a systematic
study of all the cases of interest as candidates for 0νββ
Recent results on heavy-ion induced reactions of interest for neutrinoless double beta decay at INFN-LNS

The possibility to use a special class of heavy-ion induced direct reactions, such as double charge exchange reactions, is discussed in view of their application to extract information that may be helpful to determinate the nuclear matrix elements entering in the expression of neutrinoless double beta decay halflife. The rapid spread of SARS-COV-2 Omicron variant in Italy reflected early through wastewater surveillance
The SARS-CoV-2 Omicron variant emerged in South Africa in November 2021, and has later been identified worldwide,
raising serious concerns.
A real-time RT-PCR assay was designed for the rapid screening of the Omicron variant, targeting characteristic mutations
of the spike gene. The assay was used to test 737 sewage samples collected throughout Italy (19/21 Regions) between
11 November and 25 December 2021, with the aim of assessing the spread of the Omicron variant in the
country. Positive samples were also tested with a real-time RT-PCR developed by the European Commission, Joint
Research Centre (JRC), and through nested RT-PCR followed by Sanger sequencing.
Overall, 115 samples tested positive for Omicron SARS-CoV-2 variant. The first occurrence was detected on 7
December, in Veneto, North Italy. Later on, the variant spread extremely fast in three weeks, with prevalence of positive
wastewater samples rising from 1.0% (1/104 samples) in the week 5â11 December, to 17.5% (25/143 samples)
in the week 12â18, to 65.9% (89/135 samples) in the week 19â25, in line with the increase in cases of infection with
the Omicron variant observed during December in Italy. Similarly, the number of Regions/Autonomous Provinces in
which the variant was detected increased fromone in the first week, to 11 in the second, and to 17 in the last one. The
presence of the Omicron variant was confirmed by the JRC real-time RT-PCR in 79.1% (91/115) of the positive samples,
and by Sanger sequencing in 66% (64/97) of PCR amplicons

New results from the NUMEN project
NUMEN aims at accessing experimentally driven information on Nuclear Matrix Elements (NME) involved in the half-life of the neutrinoless double beta decay (0νββ), by high-accuracy measurements of the cross sections of Heavy Ion (HI) induced Double Charge Exchange (DCE) reactions. First evidence about the possibility to get quantitative information about NME from experiments is found for the (18O,18Ne) and (20Ne,20O) reactions. Moreover, to infer the neutrino average masses from the possible measurement of the half-life of 0νββ decay, the knowledge of the NME is a crucial aspect. The key tools for this project are the high resolution Superconducting Cyclotron beams and the MAGNEX magnetic spectrometer at INFN Laboratori Nazionali del Sud in Catania (Italy). The measured cross sections are extremely low, limiting the present exploration to few selected isotopes of interest in the context of typically low-yield experimental runs. A major upgrade of the LNS facility is foreseen in order to increase the experimental yield of at least two orders of magnitude, thus making feasible a systematic study of all the cases of interest. peerReviewe
