    Generation of Tunable Narrow Bandwidth Nanosecond Pulses in the Deep Ultraviolet for Efficient Optical Pumping and High Resolution Spectroscopy

    Nanosecond optical pulses with high power and spectral brightness in the deep ultraviolet (UV) region have been produced by sum frequency mixing of nearly transform-limited-bandwidth IR light originating from a home-built injection-seeded ring cavityoptical parametric oscillator(OPO) and the fourth harmonic beam of an injection-seeded Nd:YAG laser used simultaneously to pump the OPO with the second harmonic. We demonstrate UV output, tunable from 204 to 207 nm, which exhibits pulse energies up to 5 mJ with a bandwidth better than [Math Processing Error]. We describe how the approach shown in this paper can be extended to wavelengths shorter than 185 nm. The injection-seeded OPO provides high conversion efficiency ([Math Processing Error] overall energy conversion) and superior beam quality required for highly efficient downstream mixing where sum frequencies are generated in the UV. The frequency stability of the system is excellent, making it highly suitable for optical pumping. We demonstrate high resolution spectroscopy as well as optical pumping using laser-induced fluorescence and stimulated emission pumping, respectively, in supersonic pulsed molecular beams of nitric oxide

    Perfluorochemical (PFC) liquid enhances recombinant adenovirus vector-mediated viral interleukin-10 (AdvIL-10) expression in rodent lung

    Adenovirus and cationic liposome mediated transfer of Interleukin-10 (IL-10), a potent anti-inflammatory cytokine, has been shown to decrease pro-inflammatory cytokine levels and overall lung inflammation in models of lung transplantation and injury. Limitations to current approaches of IL-10 gene therapy include poor vector delivery methods and pro-inflammatory properties of human IL-10 under certain conditions. We hypothesize that using perfluorochemical (PFC) liquid to deliver the highly homologous viral IL-10 (vIL-10), which is predominantly anti-inflammatory with minimal pro-inflammatory activities, can potentially be a more effective strategy to combat inflammatory lung diseases. In this study, we compare the use of PFC liquid versus aerosolized method to deliver adenovirus encoding the vIL-10 gene (AdvIL-10) in C57Bl6 mice. Detectable vIL-10 levels were measured from bronchoalveolar lavage fluid and lung homogenates at one, four, ten and thirty days after AdvIL-10. Furthermore, we determined if use of PFC liquid could allow for the use of a lower dose of AdvIL-10 by comparing the levels of detectable vIL-10 at different doses of AdvIL-10 delivered +/- PFC liquid. Results showed that PFC liquid enhanced detectable vIL-10 by up to ten fold and that PFC liquid allowed the use of ten-fold less vector. PFC liquid increased detectable vIL-10 in lung homogenates at all time points; however, the increase in detectable vIL-10 in BAL fluid peaked at four days and was no longer evident by thirty days after intratracheal instillation. In summary, this is the first report utilizing PFC liquid to enhance the delivery of a potentially therapeutic molecule, vIL-10. We believe this strategy can be used to perform future studies on the use of the predominantly anti-inflammatory vIL-10 to treat inflammatory lung diseases

    Insights and issues with simulating terrestrial DOC loading of Arctic river networks

The definitive version was published in Ecological Applications 23 (2013): 1817-1836, doi:10.1890/11-1050.1.Terrestrial carbon dynamics influence the contribution of dissolved organic carbon (DOC) to river networks in addition to hydrology. In this study, we use a biogeochemical process model to simulate the lateral transfer of DOC from land to the Arctic Ocean via riverine transport. We estimate that, over the 20th century, the pan-Arctic watershed has contributed, on average, 32 Tg C/yr of DOC to river networks emptying into the Arctic Ocean with most of the DOC coming from the extensive area of boreal deciduous needle-leaved forests and forested wetlands in Eurasian watersheds. We also estimate that the rate of terrestrial DOC loading has been increasing by 0.037 Tg C/yr2 over the 20th century primarily as a result of climate-induced increases in water yield. These increases have been offset by decreases in terrestrial DOC loading caused by wildfires. Other environmental factors (CO2 fertilization, ozone pollution, atmospheric nitrogen deposition, timber harvest, agriculture) are estimated to have relatively small effects on terrestrial DOC loading to Arctic rivers. The effects of the various environmental factors on terrestrial carbon dynamics have both offset and enhanced concurrent effects on hydrology to influence terrestrial DOC loading and may be changing the relative importance of terrestrial carbon dynamics on this carbon flux. Improvements in simulating terrestrial DOC loading to pan-Arctic rivers in the future will require better information on the production and consumption of DOC within the soil profile, the transfer of DOC from land to headwater streams, the spatial distribution of precipitation and its temporal trends, carbon dynamics of larch-dominated ecosystems in eastern Siberia, and the role of industrial organic effluents on carbon budgets of rivers in western Russia.This study was supported, in part, by the U.S. National Science Foundation under grants ARC-0531047, ARC-0531082, ARC-0531119, ARC-0554811, and ARC- 0652838; the U.S. Environmental Protection Agency under grant R833261; the U.S. Department of Energy under grant DE-FG02-08ER64597; and the U.S. National Aeronautics and Space Administration under grant NNX09A126G

    GSK-3β: A Bifunctional Role in Cell Death Pathways

    Although glycogen synthase kinase-3 beta (GSK-3β) was originally named for its ability to phosphorylate glycogen synthase and regulate glucose metabolism, this multifunctional kinase is presently known to be a key regulator of a wide range of cellular functions. GSK-3β is involved in modulating a variety of functions including cell signaling, growth metabolism, and various transcription factors that determine the survival or death of the organism. Secondary to the role of GSK-3β in various diseases including Alzheimer's disease, inflammation, diabetes, and cancer, small molecule inhibitors of GSK-3β are gaining significant attention. This paper is primarily focused on addressing the bifunctional or conflicting roles of GSK-3β in both the promotion of cell survival and of apoptosis.     Factors Associated With Ordering Laboratory Monitoring Of High-Risk Medications

    Background Knowledge about factors associated with provider ordering of appropriate testing is limited. Objective To determine physician factors correlated with ordering of recommended laboratory monitoring tests for high-risk medications, accounting for patient characteristics. Methods Analysis of the administrative claims and electronic medical records of patients prescribed a high-risk medication requiring laboratory monitoring in a large multispecialty group practice between January 1, 2008 and December 31, 2008. The outcome is a physician order for each recommended laboratory test for each prescribed medication. Key predictor variables include physician characteristics, including age, gender, specialty training, years since completing training, and prescribing volume. We used multivariable logistic regression to identify the independent association of physician and patient characteristics with ordering of laboratory tests to monitor medications after adjustment for potential confounders, taking into account clustering of drugs within patients and patients within providers. Results Physician orders for laboratory testing varied across drug-test pairs and ranged from 9% (Primidone–Phenobarbital level) to 97% (Azathioprine–CBC) with 50% of drug-test pairs in the 85-91% ordered range. Failure to order a test was associated with lower provider prescribing volume for study drugs and whether the physician was a specialist (primary care providers were more likely to order tests than specialists). Patients with lower patient comorbidity burden and younger patients were less likely to have appropriate tests ordered. Drug-test combinations with black box warnings were more likely to have tests ordered. Conclusions Interventions targeting providers should be addressed at those subgroups with the greatest potential for improvement: providers with lower frequencies of prescribing high-risk medications, and healthier and younger patients. Drug-test combinations with black box warnings have higher ordering rates, but many medications without such warnings also have evidence of harm, thus efforts to improve testing are necessary for all medications shown to be high-risk
