    PATHOGENICITY OF EIMERIA SPECIES ISOLATED FROM CHICKEN IN THUA THIEN HUE PROVINCE

    Abstract: The present study clarifies the effect of locally isolated Eimeria species on chickens in central Vietnam. Oocysts of Eimeria species were isolated from feces suspected to be infected with coccidiosis in 3 farms in Huong Thuy district, Thua Thien Hue province. A total of 54 2-week-old chickens were randomly allocated to 2 groups: 3 replicates containing 9 chickens in each replicate. The chicken in group 1 were orally inoculated with 2×104 isolated oocysts of Eimeria species, while the chickens in group 2 were inoculated with PBS as a control. Growth performance, oocyst output, gross lesions, and histopathological lesions were measured at 5, 10 and 28 days after infection. The chicks in group 1 showed general signs of ruffled feathers, anorexia, huddling together with diarrhea and/or bloody dropping from 4–7 days post-infection. Feed intake, growth performance and body weight in group 1 were lower than those of the control chicks. The oocyst was detected in the feces of chicks in group 1 from 6–14 days post-infection, among them, the number of oocysts reach the peak at 8 days post-infection. At day 5 post-infection, the most damaged part of the intestine is the ceca with a large amount of blood in the ceca content. The histopathological lesions were detected clearly by HE staining at day 5 and 10 post-infection. In conclusion, the locally isolated oocysts of Eimeria species have high virulence to the chickens. The present study provides useful information related to the pathogenicity of Eimeria species which may contribute to coccidiosis diagnosis and treatment in poultry production.Keywords: Eimeria, coccidiosis, infection, central Vietna

    Selective breeding of saline-tolerant striped catfish (Pangasianodon hypophthalmus) for sustainable catfish farming in climate vulnerable Mekong Delta, Vietnam

    peer reviewedStriped catfish (Pangasianodon hypophthalmus), a freshwater species cultured mainly in the Mekong Delta region in Southern Vietnam, is facing a significant challenge due to salinity intrusion as a result of climatic changes. Given these evolving environmental conditions, selecting new strains with a higher salinity tolerance could make production of striped catfish economically feasible in brackish environments. In this study, we carried out a selection program aimed at developing a striped catfish strain able to survive and grow fast in a saline environment. To implement the selection program, we first collected males and females from different provinces in the Mekong delta. We next performed a factorial cross of these breeders to produce half- and full-sib families. When fish reached fry stage (47 dph), we put them in a saline environment (10 ppt) and subsequently kept 50 % of the fastest-growing fish after 143 days post hatching (dph). We repeated this mass selection procedure after 237 dph and 340 dph. We maintained in parallel a randomly selected group in saline conditions and a group of fish reared in freshwater to serve as controls. After crossing the selected individuals, we performed several tests on the next generation of fish to evaluate the effectiveness of selection after one generation in saline conditions. Average direct responses to selection were 18.0 % for growth and 11.4 % for survival rate after one generation of selection. We estimated a moderate realized heritability (0.29) for body weight. The genetic gains obtained in our study for body weight and survival rate after one generation of selection under saline conditions suggest that selection can be effective to improve ability of striped catfish to cope with saline stress. We conclude that our selection program has succeeded in developing a productive strain of striped catfish with better tolerance to salinity. © 2022 The Author

    Habenaria diphylla (Nimmo) Dalzell is reported for the first time as a new discovery for the flora of Vietnam based on the specimens collected in Binh Chau-Phuoc Buu Nature Reserve, Ba Ria-Vung Tau Province. The present study provided the detailed characteristics of the species including detailed photographs of the morphological characteristics, the cross section of the leaf, inflorescence axis and root. Furthermore, the information about the species, including distribution, habitat, ecology and conservation status were also provided

    Analysis of Antimicrobial Resistance in Non-typhoidal Salmonella Collected From Pork Retail Outlets and Slaughterhouses in Vietnam Using Whole Genome Sequencing.

    Non-typhoidal salmonella (TS) remains a significant health burden worldwide. In Vietnam, pork accounts for 70% of the total meat consumed, and contamination with Salmonella is high. High levels of antimicrobial resistance (AMR) have emerged among porcine NTS and of particular concern is the emergence of colistin resistance, a "last defense" antibioic against multi-drug resistant (MDR) Gram-negative pathogens. This study aimed to investigate the antibiotic susceptibility of 69 NTS isolates collected from the pork retail outlets and slaughterhouses in Vietnam during 2014 a nd 2018/19. Phenotypic testing and whole genome sequencing was used to assess the serotype and AMR gene profiles of the 69 NTS isolates. Seventeen different serotypes were identified, of which S. enterica subsp enterica serotype Typhimurium was the most common followed by S. ser. Rissen, S. ser. London, S. ser. Anatum, and S. ser. Derby. Phenotype AMR was common with 41 (59.4%) isolates deemed MDR. MDR strains were most common in slaughterhouses (83%) and supermarkets (75%) and lowest in traditional markets (38%) and convenience stores (40%). Colistin resistance was identified in 18 strains (15 resistant, three intermediate) with mcr-1 identified in seven isolates (S. ser. Meleagridis, S. Rissen, S. Derby) and mcr-3 in two isolates (S. Typhimurium). This includes the first mcr positive S. Meleagridis to our knowledge. Surprisingly, boutique stores had high levels (60%) of MDR isolates including 5/20 isolates with mcr-1. This study demonstrates that pork from modern retail stores classed as supermarkets or boutique (with pork claiming to be high quality, traceable, environmentally friendly marketed toward higher income consumers) still contained NTS with high levels of AMR

    Chemical profiles and antibacterial activity of acetone extract of two Curcuma species from Vietnam

    Curcuma thorelii Gagnep. and Curcuma cotuana Luu, Škorni?k. & H.?.Tr?n are the rare species only found in Southeast Asia. The present study was the first to explore the chemical compositions and antibacterial effects of the whole plant acetone extracts of these 2 species. Altogether 41 and 31 compounds have been identified in C. thorelii and C. cotuana extracts by gas chromatography/mass spectrometry. Accordingly, the C. thorelii extract contained (E)-labda-8(17),12-diene-15,16-dial (33.37%), vitamin E (12.33%), phytol (9.83%) as the major compounds while C. cotuana extract contained predominantly (E)-labda-8(17),12-diene-15,16-dial (14.58%), n-hexadecanoic acid (10.96%), 3,7,11,15-tetramethylhexadec-2-en-1-yl acetate (8.13%), ?-sitosterol (7.97%). In addition, results from disc diffusion assay have shown that C. thorelii acetone extract had inhibitory effects on 5 out of 10 pathogenic bacterial strains such as Bacillus cereus (ATCC 11778), Listeria monocytogenes (ATCC 19111), Staphylococcus aureus (ATCC 25923), S. aureus (ATCC 29213) and S. saprophyticus (BAA750) while C. cotuana acetone extract was found to be effective only against B. cereus. The obtained results showed that the acetone extracts of C. thorelii and C. cotuana possessed several valuable bioactive compounds as well as promising antibacterial activity, which place a good foundation for future pharmaceutical product development

    Short Tandem Repeats Used in Preimplantation Genetic Testing of Î’-Thalassemia: Genetic Polymorphisms For 15 Linked Loci in the Vietnamese Population

    BACKGROUND: β-thalassemia is one of the most common monogenic diseases worldwide. Preimplantation genetic testing (PGT) of β-thalassemia is performed to avoid affected pregnancies has become increasingly popular worldwide. In which, the indirect analysis using short tandem repeat (STRs) linking with HBB gene to detect different β-globin (HBB) gene mutation is a simple, accurate, economical and also provides additional control of contamination and allele-drop-out ADO. AIM: This study established microsatellite markers for PGT of Vietnamese β-thalassemia patient. METHODS: Fifteen (15) STRs gathered from 5 populations were identified by in silico tools within 1 Mb flanking the HBB gene. The multiplex PCR reaction was optimized and performed on 106 DNA samples from at-risk families. RESULTS: After estimating, PIC values were ≥ 0.7 for all markers, with expected heterozygosity and observed heterozygosity values ranged from 0.81 to 0.92 and 0.53 to 0.86, respectively. One hundred percent of individuals had at least seven heterozygous markers and were found to be heterozygous for at least two markers on either side of the HBB gene. The STRs panel was successfully performed on one at-risk family. CONCLUSION: In general, a pentadecaplex marker (all < 1 Mb from the HBB gene) assay was constituted for β-thalassemia PGT on Vietnamese population

    The Role of Serial NT-ProBNP Level in Prognosis and Follow-Up Treatment of Acute Heart Failure after Coronary Artery Bypass Graft Surgery

    BACKGROUND: After coronary artery bypass graft (CABG) surgery, heart failure is still major problem. The valuable marker for it is needed. AIM: Evaluating the role of serial NT-proBNP level in prognosis and follow-up treatment of acute heart failure after CABG surgery. METHODS: The prospective, analytic study evaluated 107 patients undergoing CABG surgery at Ho Chi Minh Heart Institute from October 2012 to June 2014. Collecting data was done at pre- and post-operative days with measuring NT-proBNP levels on the day before operation, 2 hours after surgery, every next 24 h until the 5th day, and in case of acute heart failure occurred after surgery. RESULTS: On the first postoperative day (POD1), the NT-proBNP level demonstrated significant value for AHF with the cut-off point = 817.8 pg/mL and AUC = 0.806. On the second and third postoperative day, the AUC value of NT- was 0.753 and 0.751. It was statistically significant in acute heart failure group almost at POD 1 and POD 2 when analyzed by the doses of dobutamine, noradrenaline, and adrenaline (both low doses and normal doses). CONCLUSION: Serial measurement of NT-proBNP level provides useful prognostic and follow-up treatment information in acute heart failure after CABG surgery

    Bidens pilosa is used in traditional medicine in Vietnam. The antioxidant potential of the ethanol extract and fractions from the flowers of Bidens pilosa was evaluated through DPPH and ABTS radical scavenging and the total antioxidant capacity method. The ethyl acetate fraction exhibits the highest activity with the lowest IC50 value (IC50 = 31.54 μg·mL–1 and IC50 = 35.33 μg·mL–1 for DPPH and ABTS radical scavenging capacity), and the total antioxidant capacity was 85.05 ± 0.28 mg GA·g–1. The composition of Bidens pilosa flowers: the total phenolic, total flavonoid, polysaccharides, and triterpenoid, was examined by using the colorimetric method, and their quantities are equivalent to 59.35 ± 0.83 mg GAE·g–1, 42.35 ± 1.50 mg QE·g–1, 4.44 ± 0.02%, and 32.88 ± 0.66 mg acid oleanolic·g–1, respectively. Specifically, the polysaccharide and total triterpenoid content of Bidens pilosa flowers was reported for the first time.Xuyến chi đã được sử dụng trong các bài thuốc cổ truyền Việt Nam. Khả năng chống oxy hóa của cao toàn phần và các cao phân đoạn từ hoa cây Xuyến chi được đánh giá thông qua ba mô hình: tổng khả năng chống oxy hoá, khả năng bắt gốc tự do DPPH và khả năng bắt gốc ABTS. Kết quả cho thấy cao ethyl acetate có khả năng chống oxy hóa tốt nhất với IC50 nhỏ nhất (IC50 = 31,54 μg·mL–1 và           IC50 = 35,33 μg·mL–1 tương ứng với khả năng bắt gốc DPPH và ABTS) và hàm lượng các chất chống oxy hóa cao nhất (85,05 ± 0,28 mg·g–1 acid gallic). Hàm lượng các hợp chất có hoạt tính sinh học (tổng các hợp chất phenol, tổng flavonoid, tổng triterpenoid và polysaccharide) trong dịch chiết hoa cây Xuyến chi được xác định bằng phương pháp trắc quang. Hàm lượng tổng các hợp chất phenol và flavonoid là 59,35 ± 0,83 mg GAE·g–1 và 42,35 ± 1,50 mg QE·g–1; hàm lượng polysacharide và triterpenoid là 4,44 ± 0,02% và 32,88 ± 0,66 mg acid oleanolic·g–1. Lần đầu tiên, tổng hàm lượng triterpenoid và polysacharide trong hoa Xuyến chi được công bố
