    Positively charged amino acids are essential for electron transfer and protein-protein interactions in the soluble methane monooxygenase complex from methylococcus capsulatus (Bath)

    The soluble methane monooxygenase (sMMO) complex from Methylococcus capsulatus (Bath) catalyses oxygen- and NAD(P)H-dependent oxygenation of methane, propene and other substrates. Whole-complex sMMO oxygenase activity requires all three sMMO components: the hydroxylase, the reductase and protein B. Also, in the presence of hydrogen peroxide, the hydroxylase alone catalyses substrate oxygenation via the peroxide shunt reaction. We investigated the effect of amine cross-linking on hydroxylase activity in order to probe the role of a gross conformational change that occurs in the hydroxylase upon binding of the other protein components. The cross-linker inhibited hydroxylase activity in the whole complex but this effect was due to covalent modification of primary amine groups rather than cross-linking. Covalent modification of arginine side-chains on the hydroxylase had a similar effect but, most remarkably, neither form of modification affected the activity of the hydroxylase via the peroxide shunt reaction. It was shown that covalent modification of positively charged groups on the hydroxylase, which occurred at multiple sites, interfered with its physical and functional interactions with protein B and with the passage of electrons from the reductase. These results indicate that protein B and the reductase of the sMMO complex interact via positively charged groups on the surface of the hydroxylase to induce a conformational change that is necessary for delivery of electrons into the active site of the hydroxylase. Modification of positively charged groups on protein B had no effect on its function, consistent with the hypothesis that positively charged groups on the hydroxylase interact with negative charges on protein B. Thus, we have discovered a means of specifically inactivating the interactions between the sMMO complex while preserving the catalytic activity of the hydroxylase active site which provides a new method of studying intercomponent interactions within sMMO.</p

    Theory of Pseudomodes in Quantum Optical Processes

    This paper deals with non-Markovian behaviour in atomic systems coupled to a structured reservoir of quantum EM field modes, with particular relevance to atoms interacting with the field in high Q cavities or photonic band gap materials. In cases such as the former, we show that the pseudo mode theory for single quantum reservoir excitations can be obtained by applying the Fano diagonalisation method to a system in which the atomic transitions are coupled to a discrete set of (cavity) quasimodes, which in turn are coupled to a continuum set of (external) quasimodes with slowly varying coupling constants and continuum mode density. Each pseudomode can be identified with a discrete quasimode, which gives structure to the actual reservoir of true modes via the expressions for the equivalent atom-true mode coupling constants. The quasimode theory enables cases of multiple excitation of the reservoir to now be treated via Markovian master equations for the atom-discrete quasimode system. Applications of the theory to one, two and many discrete quasimodes are made. For a simple photonic band gap model, where the reservoir structure is associated with the true mode density rather than the coupling constants, the single quantum excitation case appears to be equivalent to a case with two discrete quasimodes

    Comparison of attraction patterns of Hermetia illucens (Diptera, Stratiomyidae) associated to buried Rattus norvergicus carcasses with steroid hormones treatment

    Hermetia illucens (L.) (Diptera, Stratiomyidae) é uma espécie abundante em todo o continente americano. No entanto, ainda há poucos estudos no Brasil abordando aspectos em relação a sua biologia e ecologia, em particular associados a estudos forenses. Neste trabalho, nós observamos os efeitos de hormônios esteróides na decomposição corporal de Rattus norvergicus, variedade Wistar enterrados na atração de H. illucens. Além da presença de diversas espécies de larvas e adultos de insetos freqüentemente encontrados em carcaças nessas condições, observou-se uma grande quantidade de imaturos de H. illucens. Essa espécie representou aproximadamente 22% de todos os espécimes coletados, tendo aparecido somente nos animais que receberam tratamento com esteróides sexuais dos tipos testosterona, progesterona e estradiol. Dentre esses, a maior abundância foi verificada nos que haviam recebido testosterona (68%). A presença de imaturos de H. illucens, apenas nos animais que receberam o tratamento com esteróides, em experimentos realizados em dois anos consecutivos revela uma possível atração/seleção dessa espécie por corpos com quantidades aumentadas de hormônios sexuais. Esse comportamento pode ser particularmente útil para a entomologia forense no que diz respeito à estimativa do intervalo pós-morte (IPM), particularmente quando tem como base informações sobre o ciclo de vida e ecologia de insetos necrófagos.Hermetia illucens (L.) (Diptera, Stratiomyidae) is an abundant species in the entire American continent. Nevertheless, there are few studies in Brazil dealing with aspects of its biology and ecology, particularly associated with forensic studies. In this study we intended to observe the effects of steroids hormones in the corporal decomposition of buried Rattus norvergicus from the Wistar variety in the attraction of H. illucens. Besides the presence of several insect species commonly found in carcasses, a great amount of immature H. illucens was observed. This species represented approximately 22% of all collected specimens and it was only found in the animals that have received treatment with sexual steroids, such as testosterone, progesterone and estradiol. Among the animals under treatment, the greater abundance was verified in those who had received testosterone (68%). The presence of immatures of H. illucens only in the animals that received hormone treatments in experiments accomplished in two consecutive years strongly suggests a possible attraction of this species for bodies with different amounts of sexual hormones. This behavior can also be relevant for the forensic entomology in regard to the post-mortem interval (PMI) estimation, particularly when the PMI is based on information about the necrophagous insect's life cycle and ecology

    Analysis of the effects of overexpression of metallothionein-I in transgenic mice on the reproductive toxicology of cadmium.

    Exposure to low levels of cadmium reduces fertility. In male mice spermatogenesis is highly sensitive to cadmium, whereas in females the peri-implantation period of pregnancy is sensitive. To examine the potential roles of the cadmium-binding protein, metallothionein (MT), in the reproductive toxicology of cadmium, we examined a transgenic mouse strain that overexpresses metallothionein-I (MT-I). These mice had dramatically increased steady-state levels of MT-I mRNA and MT in the testes and in the female reproductive tract during the peri-implantation period of pregnancy, and this overexpression occurred in a cell-specific and temporally regulated manner similar to that of the endogenous MT-I gene. Transgenic and control males were injected with cadmium, and the histology of the testes was examined. An injection of 7.5 mumol Cd/kg had no effect on histology of the testes in either transgenic or control mice. In contrast, an injection of 10 mumol Cd/kg caused rapid changes in the histology of the testes and resulted in pronounced testicular necrosis in both control and transgenic mice. Female transgenic and control mice were mated and then injected with cadmium (30-45 mumol Cd/kg) on the day of blastocyst implantation (day 4). In both of these groups, injection of cadmium reduced pregnancy rate, and no dramatic protection was afforded by maternal and/or embryonic overexpression of MT. Thus, overexpression of MT-I does not significantly protect against either of these cadmium-induced effects on fertility

    Assessing the use of NMR chemical shifts for prediction of VLE in non-ideal binary liquid mixtures

    A method of estimating vapour liquid equilibrium (VLE) using NMR chemical shift data has been proposed by Xu et al. (2012). This method is based on the concept that the average local composition around each species is determined by the thermodynamics of the system, and also determines the screening of the NMR active groups within that molecule, and so their NMR chemical shifts. Xu et al.‘s method has been replicated and verified; results are confirmed to be accurate for alcohol + hydrocarbon mixtures, giving VLE predictions of comparable accuracy to the UNIFAC, generally considered the best predictive activity coefficient model available. However, for more strongly non-ideal mixtures, the method becomes less reliable, giving significantly less accurate predictions of total pressure than UNIFAC. Several causes for this are identified. The model proposed by Xu et al. (2012) is unable to fit minima or maxima in chemical shifts, which are observed experimentally in some binary mixtures. Different NMR resonances within the same molecule lead to different predictions of VLE, clearly an un-physical result. The thermodynamics of strongly non-ideal mixtures are determined by more complex interactions than a simple description of average local composition around each component in the mixtures, for example strong and directional hydrogen bonds. Different groups within the same molecule may have different local compositions in their immediate vicinity; for example in the case of alcohol + water mixtures, one would expect a clustering of water molecules around the hydroxyl group but not the aliphatic group. Hence, the concept of a simple local composition model is not valid for these more complex cases, and it is therefore not surprising that a model based on this simple concept is often not effective in predicting VLE.C.
