    Effect of the Use of Natural Gas-Diesel Fuel Mixture on Performance, Emissions and Combustion Characteristics of a Compression-Ignition Engine

    A compression ignition engine with a mechanical fuel system was converted into common rail fuel system by means of a self-developed electronic control unit. The engine was modified to be operated with mixtures of diesel and natural gas fuels in dual-fuel mode. Then, diesel fuel was injected into the cylinder while natural gas was injected into intake manifold with both injectors controlled with the electronic control unit. Energy content of the sprayed gas fuel was varied in the amounts of 0% (only diesel fuel), 15%, 40%, and 75% of total fuel’s energy content. All tests were carried out at constant engine speed of 1500 r/min at full load. In addition to the experiments, the engine was modeled with a one-dimensional commercial software. The experimental and numerical results were compared and found to be in reasonable agreement with each other. Both NO x and soot emissions were dropped with 15% and 40%, respectively, energy content rates in gas–fuel mixture compared to only diesel fuel. However, an increase was observed in carbon monoxide emissions with 15% natural gas fuel addition compared to only diesel fuel. Although smoke emission was reduced with natural gas fuel addition, there was a dramatic increase in NO x emissions with 75% natural gas fuel addition

    Analysis of AML genes in dysregulated molecular networks

    <p>Abstract</p> <p>Background</p> <p>Identifying disease causing genes and understanding their molecular mechanisms are essential to developing effective therapeutics. Thus, several computational methods have been proposed to prioritize candidate disease genes by integrating different data types, including sequence information, biomedical literature, and pathway information. Recently, molecular interaction networks have been incorporated to predict disease genes, but most of those methods do not utilize invaluable disease-specific information available in mRNA expression profiles of patient samples.</p> <p>Results</p> <p>Through the integration of protein-protein interaction networks and gene expression profiles of acute myeloid leukemia (AML) patients, we identified subnetworks of interacting proteins dysregulated in AML and characterized known mutation genes causally implicated to AML embedded in the subnetworks. The analysis shows that the set of extracted subnetworks is a reservoir rich in AML genes reflecting key leukemogenic processes such as myeloid differentiation.</p> <p>Conclusion</p> <p>We showed that the integrative approach both utilizing gene expression profiles and molecular networks could identify AML causing genes most of which were not detectable with gene expression analysis alone due to the minor changes in mRNA level.</p

    Sarcomere Formation Occurs by the Assembly of Multiple Latent Protein Complexes

    The stereotyped striation of myofibrils is a conserved feature of muscle organization that is critical to its function. Although most components that constitute the basic myofibrils are well-characterized biochemically and are conserved across the animal kingdom, the mechanisms leading to the precise assembly of sarcomeres, the basic units of myofibrils, are poorly understood. To gain insights into this process, we investigated the functional relationships of sarcomeric protein complexes. Specifically, we systematically analyzed, using either RNAi in primary muscle cells or available genetic mutations, the organization of myofibrils in Drosophila muscles that lack one or more sarcomeric proteins. Our study reveals that the thin and thick filaments are mutually dependent on each other for striation. Further, the tension sensor complex comprised of zipper/Zasp/α-actinin is involved in stabilizing the sarcomere but not in its initial formation. Finally, integrins appear essential for the interdigitation of thin and thick filaments that occurs prior to striation. Thus, sarcomere formation occurs by the coordinated assembly of multiple latent protein complexes, as opposed to sequential assembly

    A systematic review of interventions to improve knowledge and self-management skills concerning contraception, pregnancy and breastfeeding in people with rheumatoid arthritis

    This systematic review aimed to determine the effectiveness of interventions for improving knowledge and/or self-management skills concerning contraception, pregnancy and breastfeeding in people with rheumatoid arthritis (RA). We searched four databases (MEDLINE, CINAHL, Cochrane Trials, PsycINFO) using a comprehensive search strategy. Studies were eligible if they were prospective, published in English from 2004 to 2015, included participants with RA and tested an intervention designed to improve knowledge and/or self-management skills relating to family planning, pregnancy or breastfeeding. As no studies met the latter criterion, the search strategy was expanded to include all prospective studies evaluating RA educational and/or self-management interventions. Data on study characteristics, participant characteristics and programme content were extracted to summarise the evidence base for interventions to support people with RA during their reproductive years. Expanded literature searches identified 2290 papers, of which 68 were eligible. Of these, nine papers (13 %) specifically excluded pregnant women/breastfeeding mothers or recruited only older people.Only one study (1 %) explicitly evaluated pregnancy-focused education via a motherhood decision aid, while eight studies (12 %) incorporated relevant (albeit minor) components within broader RA educational or self-management interventions. Of these, three studies provided methotrexate education in relation to conception/pregnancy/breastfeeding; three incorporated discussions on RA and relationships, impact of RA on the family or sexual advice; one provided information regarding contraception and fertility; and one issued a warning regarding use of biologic therapy in pregnancy/breastfeeding. In conclusion, information regarding family planning, pregnancy or breastfeeding represents a negligible part of published RA educational interventions, with scope to develop targeted resources

    The effect of coriander seed (Coriandrum sativum L.) as diet ingredient on the performance of Japanese quail

    This study was conducted to determine the potential of coriander seed as a natural growth promoting substance in quail nutrition. Five hundred and ninety four 3-day old Japanese quails were divided into six groups of 99 birds each and randomly assigned to six treatment diets. Four of the diets contained coriander seed at 0.5%, 1%, 2% or 4% levels of inclusion, one contained 10 mg of the antibiotic, avilamycin, per kg and a control group received no supplement. Effects on feed intake, daily live weight gain, feed conversion ratio and carcass characteristics of the birds were measured over a period of 42 days. After 42 days the highest feed intake was observed in the 4% coriander group. Birds receiving the diet containing 2% coriander seed showed the highest weight gain, followed by those receiving the diets containing 1% coriander seed, the antibiotic, the 4% and the 0.5% coriander seed. The control had the lowest weight gain. The feed conversion ratio over the experimental period was significantly better in the group receiving 2% coriander seed than in the other treatments. The highest carcass yield and liver weights were also recorded in the 2% coriander group, while the lowest abdominal fat percentages were present in the 1%, 2% and 4% coriander groups. These results suggested that coriander seeds could be considered a potential natural growth promoter for poultry, and showed the best responses at a 2% level of inclusion.     Loss of FilaminC (FLNc) Results in Severe Defects in Myogenesis and Myotube Structure

    FilaminC (FLNc) is the muscle-specific member of a family of actin binding proteins. Although it interacts with many proteins involved in muscular dystrophies, its unique role in muscle is poorly understood. To address this, two models were developed. First, FLNc expression was stably reduced in C2C12 myoblasts by RNA interference. While these cells start differentiation normally, they display defects in differentiation and fusion ability and ultimately form multinucleated “myoballs” rather than maintain elongated morphology. Second, a mouse model carrying a deletion of last 8 exons of Flnc was developed. FLNc-deficient mice die shortly after birth, due to respiratory failure, and have severely reduced birth weights, with fewer muscle fibers and primary myotubes, indicating defects in primary myogenesis. They exhibit variation in fiber size, fibers with centrally located nuclei, and some rounded fibers resembling the in vitro phenotype. The similarity of the phenotype of FLNc-deficient mice to the filamin-interacting TRIO null mice was further confirmed by comparing FLNc-deficient C2C12 cells to TRIO-deficient cells. These data provide the first evidence that FLNc has a crucial role in muscle development and maintenance of muscle structural integrity and suggest the presence of a TRIO-FLNc-dependent pathway in maintaining proper myotube structure
