    Lying or Believing? Measuring Preference Falsification from a Political Purge in China 1

    Abstract Despite its wide usage in explaining some nontrivial regime dynamics in nondemocracies, preference falsification remains an empirical myth for students of authoritarian politics. We offer the first quantitative study of this phenomenon in an authoritarian setting using a rare coincidence between a major political purge in Shanghai, China, and the administration of a nationwide survey in 2006. We construct two synthetic measures for expressed and actual political support from a set of survey questions and track their changes over time. We find that after the purge there was a dramatic increase in expressed support among Shanghai respondents, yet the increase was paralleled by an equally evident decline in actual support. We interpret this divergence as evidence for the presence of preference falsification. We further show that falsification was most intense among groups that have access to alternative information but are vulnerable to state sanctions, and use two additional surveys to explore the intertemporal dynamics of falsifying behaviors

    Observation of TeV gamma-rays from the unidentified source HESS J1841-055 with the ARGO-YBJ experiment

    We report the observation of a very high energy \gamma-ray source, whose position is coincident with HESS J1841-055. This source has been observed for 4.5 years by the ARGO-YBJ experiment from November 2007 to July 2012. Its emission is detected with a statistical significance of 5.3 standard deviations. Parameterizing the source shape with a two-dimensional Gaussian function we estimate an extension \sigma=(0.40(+0.32,-0.22}) degree, consistent with the HESS measurement. The observed energy spectrum is dN/dE =(9.0-+1.6) x 10^{-13}(E/5 TeV)^{-2.32-+0.23} photons cm^{-2} s^{-1} TeV^{-1}, in the energy range 0.9-50 TeV. The integral \gamma-ray flux above 1 TeV is 1.3-+0.4 Crab units, which is 3.2-+1.0 times the flux derived by HESS.     Managing digital contention in China

    This paper explores new developments in cyber content management strategies in China by highlighting the rise of participatory, peer-to-peer censoring practices, and examining how the People's Daily have responded to the contentious events in the top 20 public opinion incidents of 2016, to illustrate how official media uses different types of management strategies to mediate and demobilise contention, on top of information containment strategies such as censorship. I also discuss briefly the creation of a Digital United Front which seeks to incorporate social influencers and cyber elites into mainstream political institutions such as the CPPCC. Not only do these strategies further undermine the formation of a political locus opposite the state, they continue to subsume previously oppositional narratives into grander narratives of stability and national progress. Online political participation in Chinese cyberspace must seek further paternalistic protection from Party authorities in order to legitimise their contention. Although this strengthens the Party-state's claim to legitimacy, ultimately this weakens the emergence of civil society in China as the only form of contention that can survive is those that are legitimised by the Party-state, and the political space oppositional to the state remain closed off

    Human Sirt-1: Molecular Modeling and Structure-Function Relationships of an Unordered Protein

    BACKGROUND: Sirt-1 is a NAD+-dependent nuclear deacetylase of 747 residues that in mammals is involved in various important metabolic pathways, such as glucose metabolism and insulin secretion, and often works on many different metabolic substrates as a multifunctional protein. Sirt-1 down-regulates p53 activity, rising lifespan, and cell survival; it also deacetylases peroxisome proliferator-activated receptor-gamma (PPAR-gamma) and its coactivator 1 alpha (PGC-1alpha), promoting lipid mobilization, positively regulating insulin secretion, and increasing mitochondrial dimension and number. Therefore, it has been implicated in diseases such as diabetes and the metabolic syndrome and, also, in the mechanisms of longevity induced by calorie restriction. Its whole structure is not yet experimentally determined and the structural features of its allosteric site are unknown, and no information is known about the structural changes determined by the binding of its allosteric effectors. METHODOLOGY: In this study, we modelled the whole three-dimensional structure of Sirt-1 and that of its endogenous activator, the nuclear protein AROS. Moreover, we modelled the Sirt-1/AROS complex in order to study the structural basis of its activation and regulation. CONCLUSIONS: Amazingly, the structural data show that Sirt-1 is an unordered protein with a globular core and two large unordered structural regions at both termini, which play an important role in the protein-protein interaction.     AMPK Regulates Circadian Rhythms in a Tissue- and Isoform-Specific Manner

    AMP protein kinase (AMPK) plays an important role in food intake and energy metabolism, which are synchronized to the light-dark cycle. In vitro, AMPK affects the circadian rhythm by regulating at least two clock components, CKIα and CRY1, via direct phosphorylation. However, it is not known whether the catalytic activity of AMPK actually regulates circadian rhythm in vivo.THE CATALYTIC SUBUNIT OF AMPK HAS TWO ISOFORMS: α1 and α2. We investigate the circadian rhythm of behavior, physiology and gene expression in AMPKα1-/- and AMPKα2-/- mice. We found that both α1-/- and α2-/- mice are able to maintain a circadian rhythm of activity in dark-dark (DD) cycle, but α1-/- mice have a shorter circadian period whereas α2-/- mice showed a tendency toward a slightly longer circadian period. Furthermore, the circadian rhythm of body temperature was dampened in α1-/- mice, but not in α2-/- mice. The circadian pattern of core clock gene expression was severely disrupted in fat in α1-/- mice, but it was severely disrupted in the heart and skeletal muscle of α2-/- mice. Interestingly, other genes that showed circadian pattern of expression were dysreguated in both α1-/- and α2-/- mice. The circadian rhythm of nicotinamide phosphoryl-transferase (NAMPT) activity, which converts nicotinamide (NAM) to NAD+, is an important regulator of the circadian clock. We found that the NAMPT rhythm was absent in AMPK-deficient tissues and cells.This study demonstrates that the catalytic activity of AMPK regulates circadian rhythm of behavior, energy metabolism and gene expression in isoform- and tissue-specific manners
