    MicroRNA-197 Promotes Metastasis of Hepatocellular Carcinoma by Activating Wnt/β-Catenin Signaling

    Background/Aims: MicroRNA-197 (miR-197) has been shown to play roles in epithelialmesenchymal transition (EMT) and metastasis. The Wnt/β-catenin pathway is associated with EMT, but whether miR-197 regulatesWnt/β-catenin remains unclear. This study was to demonstrate the role of miR-197 on the Wnt/β-catenin pathway in hepatocellular carcinoma (HCC). Methods: Quantitative reverse transcription polymerase chain reaction (qRT-PCR) was used to detect the expression of miR-197 in 105 HCC specimens and 15 HCC cell lines. We tested the predicted target gene of miR-197 using a genetic report system. The role of miR-197 in HCC cell invasion and migration (wound healingand cell invasion and migrationby Transwell assays) and in an HCC xenograft modelwas analyzed. Results: Using a miRNA microarray analysis of HCC specimens and compared with non-metastatic HCC, miR-197 was identified as one of the most upregulated miRNAs in metastatic HCC. miR-197 expression was positively associated with the invasiveness of HCC cell lines. Metastatic HCC cells with high miR-197 expression had Wnt/β-catenin signaling activation. High levels of miR-197 expression also promoted EMT and invasionHCC cells in vitro and in vivo. miR-197 directly targeted Axin-2, Naked cuticle 1 (NKD1), and Dickkopf-related protein 2 (DKK2), leading to inhibition of Wnt/β-catenin signaling. High miR-197 expression was found in HCC specimens from patients with portal vein metastasis;high miR-197 expression correlated to the expression of Axin2, NKD1, and DKK2. Conclusion: miR-197 promotes HCC invasion and metastasis by activating Wnt/β-catenin signaling. miR-197 could possibly be used as a prognostic marker and therapeutic target for HCC

    The ongoing outbreak of COVID-19 that began in Wuhan, China, has constituted a Public Health Emergency of International Concern, with cases confirmed in multiple countries. Currently, patients are the primary source of infection. We report a confirmed case of COVID-19 whose oropharyngeal swab test of SARS-CoV-2 RNA turned positive in convalescence. This case highlights the importance of active surveillance of SARS-CoV-2 RNA for infectivity assessment

    Effects of Fe concentration on microstructure and corrosion of Mg-6Al-1Zn-xFe alloys for fracturing balls applications

    Mg-6Al-1Zn-xFe (x = 0, 1, 3, 5 and 7 wt%) alloys were prepared by powder metallurgy and followed by hot extrusion. Majority of Fe element exists as insoluble particles in the alloys. The as-extruded alloys showed higher degradable rates but less stable mechanical properties than as-annealed alloys. Corrosion rate of all the alloys increased with increasing Fe concentration, reaching 2.4 mL cm h. 0.2% yield strength of all the alloys was higher than 150 MPa. In short, Mg-6Al-1Zn-xFe alloys have an attractive combination of corrosion and mechanical properties, which holds a bright future for fracturing balls applications

    Microstructure evolution, mechanical properties and deformation characteristics of ultrafine-grained annealed pure aluminum

    © 2022In this paper, ultrafine-grained pure aluminum was first fabricated by heavy extrusion. Subsequently, the prepared samples were annealed at 350 oC for different times to tailor heterostructures with the aim of achieving strength-ductility balance. The microstructure characterization revealed that the annealed samples formed heterostructures composed of recrystallized coarse grains, retained ultrafine grains, and coarsening banded structure. Typical rolling texture components prevailed as annealing time was below 8 min. When annealing time exceeded 8 min, these texture components were gradually replaced by the Cube-oriented and random-oriented grains, and at the same time, abnormal grain growth occurred with a faster kinetic (growth exponent ñ3.4). Tensile tests indicated that 5–8 min annealed samples obtained a reasonable strength-ductility balance, which was higher than their counterparts predicted by the rule-of-mixture based on the recrystallization model. In addition, the macro-deformation characteristics of the heterostructured sample were comparatively studied using digital image correlation, while the micro-deformation behaviors were revealed by in-situ tension. The results showed that the heterostructured sample caused inhomogeneous deformation and significant geometrically necessary dislocations gradient at the hetero-boundaries. The loading-unloading-reloading tests found that the interaction of heterostructures induced a noteworthy hetero-deformation-induced (HDI) stress, thereby contributing to HDI strengthening.N

    Association between the nasopharyngeal microbiome and metabolome in patients with COVID-19

    SARS-CoV-2, the causative agent for COVID-19, infect human mainly via respiratory tract, which is heavily inhabited by local microbiota. However, the interaction between SARS-CoV-2 and nasopharyngeal microbiota, and the association with metabolome has not been well characterized. Here, metabolomic analysis of blood, urine, and nasopharyngeal swabs from a group of COVID-19 and non-COVID-19 patients, and metagenomic analysis of pharyngeal samples were used to identify the key features of COVID-19. Results showed lactic acid, l-proline, and chlorogenic acid methyl ester (CME) were significantly reduced in the sera of COVID-19 patients compared with non-COVID-19 ones. Nasopharyngeal commensal bacteria including Gemella morbillorum, Gemella haemolysans and Leptotrichia hofstadii were notably depleted in the pharynges of COVID-19 patients, while Prevotella histicola, Streptococcus sanguinis, and Veillonella dispar were relatively increased. The abundance of G. haemolysans and L. hofstadii were significantly positively associated with serum CME, which might be an anti-SARS-CoV-2 bacterial metabolite. This study provides important information to explore the linkage between nasopharyngeal microbiota and disease susceptibility. The findings were based on a very limited number of patients enrolled in this study; a larger size of cohort will be appreciated for further investigation
