Requirement of the NPXY motif in the integrin beta 3 subunit cytoplasmic tail for melanoma cell migration in vitro and in vivo.
The NPXY sequence is highly conserved among integrin beta subunit cytoplasmic tails, suggesting that it plays a fundamental role in regulating integrin-mediated function. Evidence is provided that the NPXY structural motif within the beta 3 subunit, comprising residues 744-747, is essential for cell morphological and migratory responses mediated by integrin alpha v beta 3 in vitro and in vivo. Transfection of CS-1 melanoma cells with a cDNA encoding the wild-type integrin beta 3 subunit, results in de novo alpha v beta 3 expression and cell attachment, spreading, and migration on vitronectin. CS-1 cells expressing alpha v beta 3 with mutations that disrupt the NPXY sequence interact with soluble vitronectin or an RGD peptide, yet fail to attach, spread, or migrate on immobilized ligand. The biological consequences of these observations are underscored by the finding that CS-1 cells expressing wild-type alpha v beta 3 acquire the capacity to form spontaneous pulmonary metastases in the chick embryo when grown on the chorioallantoic membrane. However, migration-deficient CS-1 cells expressing alpha v beta 3 with mutations in the NPXY sequence lose this ability to metastasize. These findings demonstrate that the NPXY motif within the integrin beta 3 cytoplasmic tail is essential for alpha v beta 3-dependent post-ligand binding events involved in cell migration and the metastatic phenotype of melanoma cells
Effect of insulin-like growth factor I on HIV type 1 long terminal repeat-driven chloramphenicol acetyltransferase expression
In this study, we have investigated the ability of insulin-like growth factor I (IGF-I) to inhibit HIV long terminal repeat (LTR)-driven gene expression. Using COS 7 cells cotransfected with tat and an HIV LTR linked to a chloramphenicol acetyltransferase (CAT) reporter, we observed that physiological levels of IGF-I (10-9 M) significantly inhibited CAT expression in a concentration- and time-dependent manner. IGF-I did not inhibit C AT expression in COS 7 cells transfected with pSVCAT, and did not affect CAT expression in the absence of cotransfection with tat . Transfection of HIV-1 proviral DNA into COS 7 cells +/- IGF-I resulted in a significant decrease ( p 0.05) in infectious virion production. Both IGF-I and Ro24-7429 inhibited LTR-driven C AT expression, while TNF- alpha -enhanced CAT expression was not affected by IGF-I. On the other hand, a plasmid encoding parathyroid hormone-related peptide exhibited dramatic additivity of inhibition of CAT expression in COS 7 cells. Finally, we show that in Jurkat or U937 cells cotransfected with HIVLTRCAT/tat, IGF-I significantly inhibited CAT expression. Further, interleukin 4 showed in U937 cells inhibition of CAT expression that was not additive to IGF-I induced inhibition. Our data demonstrate that IGF-I can specifically inhibit HIVLTRCAT expression. This inhibition may occur at the level of the tat /TAR interaction. Finally, this IGF-I effect is seen in target cell lines and similar paths of inhibition may be involved in the various cell types employed. <br /
Emission Characteristics of the Projectile Fragments at Relativistic Energy
A projectile (84^Kr_36) having kinetic energy around 1 A GeV was used to
expose NIKFI BR-2 emulsion target. A total of 700 inelastic events are used in
the present studies on projectile fragments. The emission angle of the
projectile fragments are strongly affected by charge of the other projectile
fragments emitted at same time with different emission angle is observed. The
angular distribution studies show symmetrical nature for lighter charge
projectile fragments. The symmetrical nature decreased with the charge of
projectile fragments. At ~4o of emission angle for double charge projectile
fragments, the momentum transfer during interaction is similar for various
target species of emulsion were observed. We also observed a small but
significant amplitude peaks on both side of the big peak for almost all light
charge projectile fragments having different delta angle values. It reflects
that there are few percent of projectile fragments that are coming from the
decay of heavy projectile fragments or any other process.Comment: 32 pages, 17 Figure
Vaginal hydrolytic enzymes, immunoglobulin A against Gardnerella vaginalis toxin, and risk of early preterm birth among women in preterm labor with bacterial vaginosis or intermediate flora
Objective: The purpose of this study was to determine whether the microbial hydrolytic enzymes, sialidase and prolidase, and immunoglobulin A against the Gardnerella vaginalis cytolysin (anti-Gvh IgA) increase the risk for early preterm birth ( 6434 weeks of gestation) among women with bacterial vaginosis or intermediate flora. Study Design: Two hundred eighteen afebrile women in preterm labor with intact membranes had a vaginal Gram stain performed, and sialidase, prolidase, and anti-Gvh IgA concentrations were determined. Results: Women with bacterial vaginosis or intermediate flora had significantly higher sialidase and prolidase concentrations than women with normal flora. Among women with bacterial vaginosis or intermediate flora, the women with sialidase had a higher rate of early preterm birth (P =.05). Sialidase had a sensitivity of 43% and specificity of 77% for early preterm birth. Prolidase and anti-Gvh IgA did not predict early preterm birth. Conclusion: Women in preterm labor with bacterial vaginosis or intermediate flora and detectable sialidase are at increased risk of early preterm birth
Are oxygen and sulfur atoms structurally equivalent in organic crystals?
New guidelines for the design of structurally equivalent molecular crystals were derived from structural analyses of new cocrystals and polymorphs of saccharin and thiosaccharin, aided by a computational study. The study shows that isostructural crystals may be obtained through an exchange of >C?O with >C?S in the molecular components of the solids, but only if the exchanged atom is not involved in hydrogen bonding
The relationship between web enjoyment and student perceptions and learning using a web-based tutorial
Web enjoyment has been regarded as a component of system experience. However, there has been little targeted research considering the role of web enjoyment alone in student learning using web-based systems. To address this gap, this study aims to examine the influence of web enjoyment on learning performance and perceptions by controlling system experience as a variable in the study. 74 students participated in the study, using a web-based tutorial covering subject matter in the area of 'Computation and algorithms'. Their learning performance was assessed with a pre-test and a post-test and their learning perceptions were evaluated with a questionnaire. The results indicated that there are positive relationships between the levels of web enjoyment and perceived usefulness and non-linear navigation for users with similar, significant levels of system experience. The implications of these findings in relation to web-based learning are explored and ways in which the needs of students who report different levels of web enjoyment might be met are discussed
A Suborbital Payload for Soft X-ray Spectroscopy of Extended Sources
We present a suborbital rocket payload capable of performing soft X-ray
spectroscopy on extended sources. The payload can reach resolutions of
~100(lambda/dlambda) over sources as large as 3.25 degrees in diameter in the
17-107 angstrom bandpass. This permits analysis of the overall energy balance
of nearby supernova remnants and the detailed nature of the diffuse soft X-ray
background. The main components of the instrument are: wire grid collimators,
off-plane grating arrays and gaseous electron multiplier detectors. This
payload is adaptable to longer duration orbital rockets given its comparatively
simple pointing and telemetry requirements and an abundance of potential

Field Theory for a Deuteron Quantum Liquid
Based on general symmetry principles we study an effective Lagrangian for a
neutral system of condensed spin-1 deuteron nuclei and electrons, at
greater-than-atomic but less-than-nuclear densities. We expect such matter to
be present in thin layers within certain low-mass brown dwarfs. It may also be
produced in future shock-wave-compression experiments as an effective fuel for
laser induced nuclear fusion. We find a background solution of the effective
theory describing a net spin zero condensate of deuterons with their spins
aligned and anti-aligned in a certain spontaneously emerged preferred
direction. The spectrum of low energy collective excitations contains two spin
waves with linear dispersions -- like in antiferromagnets -- as well as gapped
longitudinal and transverse modes related to the Meissner effect -- like in
superconductors. We show that counting of the Nambu-Goldstone modes of
spontaneously broken internal and space-time symmetries obeys, in a nontrivial
way, the rules of the Goldstone theorem for Lorentz non-invariant systems. We
discuss thermodynamic properties of the condensate, and its potential

Joint actions with large partners and small-firm ambidexterity in asymmetric alliances:The mediating role of relational identification
This study investigates the role of relational identification in the relation between joint actions and small-firm ambidexterity in asymmetric alliances. Using survey data on Chinese high-technology firms, we find that joint problem-solving and joint sensemaking are both positively associated with small firm's relational identification. We also find a positive relationship between small firm's relational identification and knowledge exploration and exploitation. More importantly, we show that relational identification mediates the relationships between joint actions (i.e., joint problem-solving and joint sensemaking) and small-firm ambidexterity, except for the relationship between joint sensemaking and small-firm knowledge exploitation. This study advances our understanding of the association between joint actions and ambidexterity by providing a social identification explanation
