    PENGARUH BERBAGAI DOSIS PUPUK KIMIA DAN PUPUK ORGANIK CAIR TERHADAP PERTUMBUHAN DAN PRODUKSI TANAMAN TEMBAKAU PAYAKUMBUH (Nicotiana tabacum L.)

    Tembakau merupakan salah satu komoditi perkebunan yang diusahakan di Indonesia. Diversifikasi produk tanaman daun tembakau dapat dimanfaatkan menjadi bahan kimia dasar sebagai bahan baku pestisida, produk kosmetik dan industri farmasi. Selain dari pada itu minyak atsiri dan pemanfaatannya untuk campuran parfum (tabac perfume) dan debu tembakau untuk pupuk kompos. Untuk mendapatkan nilai ekonomis yang baik maka perlu peningkatan produksi dan kualitas hasil tanaman tembakau. Sehingga dalam aspek budidaya perlu untuk dilakukan perbaikan.  Pemupukan merupakan salah satu upaya pemeliharaan tanaman dengan tujuan memperbaiki kesuburan tanah melalui cara penambahan unsur hara, baik makro maupun mikro yang berguna bagi pertumbuhan dan perkembangan tanaman tembakau. Salah satu produk yang bisa dipakai adalah Pupuk organik cair Hantu. Penelitian ini dilaksanakan di kebun percobaan Politeknik Pertanian Negeri Payakumbuh pada bulan Februari sampai Juli 2022. Rancangan yang digunakana Rancangan Acak Kelompok yang terdiri atas lima taraf perlakuan dan lima ulangan yang dianalisis dengan Uji F dan uji lanjut Duncan Multiple Range Test (DMRT) taraf 5%. Perlakuan adalah P0 = Tanpa Pupuk Kimia dan POC 2 ml/l air,P1 = Pupuk Kimia 25% dosis anjuran dan POC 2 ml/l air, P2= Pupuk Kimia 50% dosis anjuran dan POC 2 ml/l air, P3 = Pupuk Kimia 75% dosis anjuran dan POC 2 ml/l air, dan P4 = Pupuk Kimia 100% dosis anjuran dan POC Hantu 2 ml/l air. Parameter yang diamati adalah tinggi tanaman (cm), diameter batang (mm), panjang daun terpanjang (cm), lebar daun terlebar (cm), jumlah daun (helai), panen (gram), berat basah (gram), dan berat kering (gram). Hasil penelitian pemberian pupuk kimia 25%, 50% 75% dan 100% dosis anjuran dengan pemberian POC Hantu 2 ml/l air memberikan pengaruh terhadap pertumbuhan dan produksi tanaman tembakau Payakumbuh. Pemberian perlakuan pupuk buatan 75% dosis anjuran dengan POC 2 ml/l air merupakan perlakuan terbaik yang memberikan pengaruh terhadap dibandingkan terhadap semua parameter pengamatan dan mampu menghemat penggunaan pupuk buatan 25%.

    Response to the Growth of Local Payakumbuh Tobacco (Nicotiana Tabacum L) on Giving Kiambang Compose (Pistia Stratiotes)

    Tobacco plants are one of plants that are often cultivated by farmers in the Limapuluh Kota district. While the productivity of tobacco plants ranges from 1.13 tons/ha – 1.83 tons/ha. The productivity of tobacco plants cannot be achieved by farmers. many things that could be the cause. one of them, is poor soil health due to a lack of organic matter contained in the soil. Various sources of organic matter can be used, one of which is organic fertilizer derived from kiambang plants (Pistia stratiotes). Kiambang compost contains Nitrogen, Phosphorus and Potassium nutrients. The aim of this study was to obtain the best basic fertilizer dose for tobacco plants from kiambang plant compost as an alternative. The time for conducting the research is from April 2022 to July 2022. The implementation of this research activity is in the experimental garden of the Payakumbuh State Agricultural Polytechnic, Tanjung Pati, Harau District, Limapuluh Kota Regency. The study used a Non-Factorial Randomized Block Design (RBD). with kiambang fertilizer doses of 0 gramss, 100 gramss, 200 gramss, 300 gramss and 400 gramss. Parameters observed in the study include plant height, number of leaves, leaf length Longest, Widest Leaf Width, Leaf Wet Weight, and Leaf Dry Weight. The results of the study Application of kiambang compost had an effect on plant height, number of leaves, leaf width, and leaf length. An additional dose of kiambang compost at a dose of 400 gramss per plant is a good treatment for the development of growth and yield of the Rudaau Gadang tobacco variety

    Role of Chaperone Mediated Autophagy (CMA) in the Degradation of Misfolded N-CoR Protein in Non-Small Cell Lung Cancer (NSCLC) Cells

    Nuclear receptor co-repressor (N-CoR) plays important role in transcriptional control mediated by several tumor suppressor proteins. Recently, we reported a role of misfolded-conformation dependent loss (MCDL) of N-CoR in the activation of oncogenic survival pathway in acute promyelocytic leukemia (APL). Since N-CoR plays important role in cellular homeostasis in various tissues, therefore, we hypothesized that an APL like MCDL of N-CoR might also be involved in other malignancy. Indeed, our initial screening of N-CoR status in various leukemia and solid tumor cells revealed an APL like MCDL of N-CoR in primary and secondary tumor cells derived from non-small cell lung cancer (NSCLC). The NSCLC cell specific N-CoR loss could be blocked by Kaletra, a clinical grade protease inhibitor and by genistein, an inhibitor of N-CoR misfolding previously characterized by us. The misfolded N-CoR presented in NSCLC cells was linked to the amplification of ER stress and was subjected to degradation by NSCLC cell specific aberrant protease activity. In NSCLC cells, misfolded N-CoR was found to be associated with Hsc70, a molecular chaperone involved in chaperone mediated autophagy (CMA). Genetic and chemical inhibition of Lamp2A, a rate limiting factor of CMA, significantly blocked the loss of N-CoR in NSCLC cells, suggesting a crucial role of CMA in N-CoR degradation. These findings identify an important role of CMA-induced degradation of misfolded N-CoR in the neutralization of ER stress and suggest a possible role of misfolded N-CoR protein in the activation of oncogenic survival pathway in NSCLC cells

    Sepsis Enhances Epithelial Permeability with Stretch in an Actin Dependent Manner

    Ventilation of septic patients often leads to the development of edema and impaired gas exchange. We hypothesized that septic alveolar epithelial monolayers would experience stretch-induced barrier dysfunction at a lower magnitude of stretch than healthy alveolar epithelial monolayers. Alveolar epithelial cells were isolated from rats 24 hours after cecal ligation and double puncture (2CLP) or sham surgery. Following a 5-day culture period, monolayers were cyclically stretched for 0, 10, or 60 minutes to a magnitude of 12% or 25% change in surface area (ΔSA). Barrier function, MAPk and myosin light chain (MLC) phosphorylation, tight junction (TJ) protein expression and actin cytoskeletal organization were examined after stretch. Significant increases in epithelial permeability were observed only in 2CLP monolayers at the 12% ΔSA stretch level, and in both 2CLP and sham monolayers at the 25% ΔSA stretch level. Increased permeability in 2CLP monolayers was not associated with MAPk signaling or alterations in expression of TJ proteins. 2CLP monolayers had fewer actin stress fibers before stretch, a more robust stretch-induced actin redistribution, and reduced phosphorylated MLCK than sham monolayers. Jasplakinolide stabilization of the actin cytoskeleton in 2CLP monolayers prevented significant increases in permeability following 60 minutes of stretch to 12% ΔSA. We concluded that septic alveolar epithelial monolayers are more susceptible to stretch-induced barrier dysfunction than healthy monolayers due to actin reorganization

    Genome-Wide Association Study in Asian Populations Identifies Variants in ETS1 and WDFY4 Associated with Systemic Lupus Erythematosus

    Systemic lupus erythematosus is a complex and potentially fatal autoimmune disease, characterized by autoantibody production and multi-organ damage. By a genome-wide association study (320 patients and 1,500 controls) and subsequent replication altogether involving a total of 3,300 Asian SLE patients from Hong Kong, Mainland China, and Thailand, as well as 4,200 ethnically and geographically matched controls, genetic variants in ETS1 and WDFY4 were found to be associated with SLE (ETS1: rs1128334, P = 2.33×10−11, OR = 1.29; WDFY4: rs7097397, P = 8.15×10−12, OR = 1.30). ETS1 encodes for a transcription factor known to be involved in a wide range of immune functions, including Th17 cell development and terminal differentiation of B lymphocytes. SNP rs1128334 is located in the 3′-UTR of ETS1, and allelic expression analysis from peripheral blood mononuclear cells showed significantly lower expression level from the risk allele. WDFY4 is a conserved protein with unknown function, but is predominantly expressed in primary and secondary immune tissues, and rs7097397 in WDFY4 changes an arginine residue to glutamine (R1816Q) in this protein. Our study also confirmed association of the HLA locus, STAT4, TNFSF4, BLK, BANK1, IRF5, and TNFAIP3 with SLE in Asians. These new genetic findings may help us to gain a better understanding of the disease and the functions of the genes involved
