    Defining Early Human NK Cell Developmental Stages in Primary and Secondary Lymphoid Tissues

    A better understanding of human NK cell development in vivo is crucial to exploit NK cells for immunotherapy. Here, we identified seven distinctive NK cell developmental stages in bone marrow of single donors using 10-color flow cytometry and found that NK cell development is accompanied by early expression of stimulatory co-receptor CD244 in vivo. Further analysis of cord blood (CB), peripheral blood (PB), inguinal lymph node (inLN), liver lymph node (liLN) and spleen (SPL) samples showed diverse distributions of the NK cell developmental stages. In addition, distinctive expression profiles of early development marker CD33 and C-type lectin receptor NKG2A between the tissues, suggest that differential NK cell differentiation may take place at different anatomical locations. Differential expression of NKG2A and stimulatory receptors (e.g. NCR, NKG2D) within the different subsets of committed NK cells demonstrated the heterogeneity of the CD56brightCD16+/− and CD56dimCD16+ subsets within the different compartments and suggests that microenvironment may play a role in differential in situ development of the NK cell receptor repertoire of committed NK cells. Overall, differential in situ NK cell development and trafficking towards multiple tissues may give rise to a broad spectrum of mature NK cell subsets found within the human body

    Outcome of paediatric intensive care survivors

    The development of paediatric intensive care has contributed to the improved survival of critically ill children. Physical and psychological sequelae and consequences for quality of life (QoL) in survivors might be significant, as has been determined in adult intensive care unit (ICU) survivors. Awareness of sequelae due to the original illness and its treatment may result in changes in treatment and support during and after the acute phase. To determine the current knowledge on physical and psychological sequelae and the quality of life in survivors of paediatric intensive care, we undertook a computerised comprehensive search of online databases for studies reporting sequelae in survivors of paediatric intensive care. Studies reporting sequelae in paediatric survivors of cardiothoracic surgery and trauma were excluded, as were studies reporting only mortality. All other studies reporting aspects of physical and psychological sequelae were analysed. Twenty-seven studies consisting of 3,444 survivors met the selection criteria. Distinct physical and psychological sequelae in patients have been determined and seemed to interfere with quality of life. Psychological sequelae in parents seem to be common. Small numbers, methodological limitations and quantitative and qualitative heterogeneity hamper the interpretation of data. We conclude that paediatric intensive care survivors and their parents have physical and psychological sequelae affecting quality of life. Further well-designed prospective studies evaluating sequelae of the original illness and its treatment are warranted

    A 'snip' in time: what is the best age to circumcise?

    <p>Abstract</p> <p>Background</p> <p>Circumcision is a common procedure, but regional and societal attitudes differ on whether there is a need for a male to be circumcised and, if so, at what age. This is an important issue for many parents, but also pediatricians, other doctors, policy makers, public health authorities, medical bodies, and males themselves.</p> <p>Discussion</p> <p>We show here that infancy is an optimal time for clinical circumcision because an infant's low mobility facilitates the use of local anesthesia, sutures are not required, healing is quick, cosmetic outcome is usually excellent, costs are minimal, and complications are uncommon. The benefits of infant circumcision include prevention of urinary tract infections (a cause of renal scarring), reduction in risk of inflammatory foreskin conditions such as balanoposthitis, foreskin injuries, phimosis and paraphimosis. When the boy later becomes sexually active he has substantial protection against risk of HIV and other viral sexually transmitted infections such as genital herpes and oncogenic human papillomavirus, as well as penile cancer. The risk of cervical cancer in his female partner(s) is also reduced. Circumcision in adolescence or adulthood may evoke a fear of pain, penile damage or reduced sexual pleasure, even though unfounded. Time off work or school will be needed, cost is much greater, as are risks of complications, healing is slower, and stitches or tissue glue must be used.</p> <p>Summary</p> <p>Infant circumcision is safe, simple, convenient and cost-effective. The available evidence strongly supports infancy as the optimal time for circumcision.</p

    The Views of Engineering Students on Creativity, In Contributions to Higher Engineering Education

    Creativity plays a growing role in education, from elementary school to higher education. Nowadays, both employers and universities develop research and are committed to the development of the twenty-first-century interpersonal, applied skills—creativity included—foreseen as fundamental to all professionals, engineers added. Generally, engineering degrees focus on the content of their scientific areas. In some higher education degrees, creativity still plays a small role. In order to reinforce the importance of creativity in the engineering degrees in a Portuguese northeastern university, it was pertinent to study the conceptions of engineering students about creativity. This study presents the conceptions of creativity of the first-year students of higher education, in the engineering area in two school years. The answers of 128 first-year students from two academic years (61 from 2014/15 and 67 from 2016/17) and four different degrees to the open question—“What is creativity?” were analyzed. It was a mixed study, qualitative to deepen students’ conceptions and quantitative to study some proportions differences and variables crossing. The results show low personal involvement even in the use of the first person plural in either school year, although the students’ most used sentence was “for me.” In both academic years, students’ definitions mentioned more the creation of the implicit category in the content analysis. The words “new” and “way” were common to all the word clouds produced, and creativity and innovation appear somehow connected. In general, proportion differences were not statistically significant and degree crossed with categories showed no dependency

    Transport of Snow by the Wind: A Comparison Between Observations in Adélie Land, Antarctica, and Simulations Made with the Regional Climate Model MAR

    For the first time a simulation of blowing snow events was validated in detail using one-month long observations (January 2010) made in Adélie Land, Antarctica. A regional climate model featuring a coupled atmosphere/blowing snow/snowpack model is forced laterally by meteorological re-analyses. The vertical grid spacing was 2 m from 2 to 20 m above the surface and the horizontal grid spacing was 5 km. The simulation was validated by comparing the occurrence of blowing snow events and other meteorological parameters at two automatic weather stations. The Nash test allowed us to compute effi- ciencies of the simulation. The regional climate model simulated the observed wind speed with a positive efficiency (0.69). Wind speeds higher than 12 m s−1 were underestimated. Positive efficiency of the simulated wind speed was a prerequisite for validating the blowing snow model. Temperatures were simulated with a slightly negative efficiency (−0.16) due to overestimation of the amplitude of the diurnal cycle during one week, probably because the cloud cover was underestimated at that location during the period concerned. Snowfall events were correctly simulated by our model, as confirmed by field reports. Because observations suggested that our instrument (an acoustic sounder) tends to overestimate the blowing snow flux, data were not sufficiently accurate to allow the complete validation of snow drift val- ues. However, the simulation of blowing snow occurrence was in good agreement with the observations made during the first 20 days of January 2010, despite the fact that the blowing snow flux may be underestimated by the regional climate model during pure blowing snow events. We found that blowing snow occurs in Adélie Land only when the 30-min wind speed value at 2 m a.g.l. is >10 m s−1. The validation for the last 10 days of January 2010 was less satisfactory because of complications introduced by surface melting and refreezing

    The complement system and age-related macular degeneration.

    PURPOSE: Age-related macular degeneration (AMD) is the leading cause of blindness in the developed world. There are increasing evidences to suggest the complement system may play a significant role on the pathogenesis of AMD. In this review, we summarise the current research in this area. METHODS: Review of literature. RESULTS: The complement system is a complex system with several activation pathways. Complement factor H (CFH) polymorphisms has been associated with increase risk of AMD. CFH is an inhibitor protein; the polymorphisms might cause uncontrolled activation by initiation events. CONCLUSION: Further studies on the molecular basis of the complement-mediated pathogenesis of AMD may offer novel therapy to AMD
