    Personalised aesthetics with residual adapters

    The use of computational methods to evaluate aesthetics in photography has gained interest in recent years due to the popularization of convolutional neural networks and the availability of new annotated datasets. Most studies in this area have focused on designing models that do not take into account individual preferences for the prediction of the aesthetic value of pictures. We propose a model based on residual learning that is capable of learning subjective, user specific preferences over aesthetics in photography, while surpassing the state-of-the-art methods and keeping a limited number of user-specific parameters in the model. Our model can also be used for picture enhancement, and it is suitable for content-based or hybrid recommender systems in which the amount of computational resources is limited.Comment: 12 pages, 4 figures.     Kinetic modelling of competition and depletion of shared miRNAs by competing endogenous RNAs

    Non-conding RNAs play a key role in the post-transcriptional regulation of mRNA translation and turnover in eukaryotes. miRNAs, in particular, interact with their target RNAs through protein-mediated, sequence-specific binding, giving rise to extended and highly heterogeneous miRNA-RNA interaction networks. Within such networks, competition to bind miRNAs can generate an effective positive coupling between their targets. Competing endogenous RNAs (ceRNAs) can in turn regulate each other through miRNA-mediated crosstalk. Albeit potentially weak, ceRNA interactions can occur both dynamically, affecting e.g. the regulatory clock, and at stationarity, in which case ceRNA networks as a whole can be implicated in the composition of the cell's proteome. Many features of ceRNA interactions, including the conditions under which they become significant, can be unraveled by mathematical and in silico models. We review the understanding of the ceRNA effect obtained within such frameworks, focusing on the methods employed to quantify it, its role in the processing of gene expression noise, and how network topology can determine its reach.Comment: review article, 29 pages, 7 figure

    Absorption Enhancement in Lossy Transition Metal Elements of Plasmonic Nanosandwiches

    Combination of catalytically active transition metals and surface plasmons offers a promising way to drive chemical reactions by converting incident visible light into energetic electron-hole pairs acting as a mediator. In such a reaction enhancement scheme, the conversion efficiency is dependent on light absorption in the metal. Hence, increasing absorption in the plasmonic structure is expected to increase generation of electron-hole pairs and, consequently, the reaction rate. Furthermore, the abundance of energetic electrons might facilitate new reaction pathways. In this work we discuss optical properties of homo- and heterometallic plasmonic nanosandwiches consisting of two parallel disks made of gold and palladium. We show how near-field coupling between the sandwich elements can be used to enhance absorption in one of them. The limits of this enhancement are investigated using finite-difference time-domain simulations. Physical insight is gained through a simple coupled dipole analysis of the nanostructure. For small palladium disks (compared to the gold disk), total absorption enhancement integrated over the near visible solar AM 1.5 spectrum is 8-fold, while for large palladium disks, similar in size to the gold one, it exceeds three

    Globally optimal geodesic active contours

    An approach to optimal object segmentation in the geodesic active contour framework is presented with application to automated image segmentation. The new segmentation scheme seeks the geodesic active contour of globally minimal energy under the sole restriction that it contains a specified internal point p_int. This internal point selects the object of interest and may be used as the only input parameter to yield a highly automated segmentation scheme. The image to be segmented is represented as a Riemannian space S with an associated metric induced by the image. The metric is an isotropic and decreasing function of the local image gradient at each point in the image, encoding the local homogeneity of image features. Optimal segmentations are then the closed geodesics which partition the object from the background with minimal similarity across the partitioning. An efficient algorithm is presented for the computation of globally optimal segmentations and applied to cell microscopy, x-ray, magnetic resonance and cDNA microarray images

    Multiple mechanisms disrupt the let-7 microRNA family in neuroblastoma

    Poor prognosis in neuroblastoma is associated with genetic amplification of MYCN. MYCN is itself a target of let-7, a tumour suppressor family of microRNAs implicated in numerous cancers. LIN28B, an inhibitor of let-7 biogenesis, is overexpressed in neuroblastoma and has been reported to regulate MYCN. Here we show, however, that LIN28B is dispensable in MYCN-amplified neuroblastoma cell lines, despite de-repression of let-7. We further demonstrate that MYCN messenger RNA levels in amplified disease are exceptionally high and sufficient to sponge let-7, which reconciles the dispensability of LIN28B. We found that genetic loss of let-7 is common in neuroblastoma, inversely associated with MYCN amplification, and independently associated with poor outcomes, providing a rationale for chromosomal loss patterns in neuroblastoma. We propose that let-7 disruption by LIN28B, MYCN sponging, or genetic loss is a unifying mechanism of neuroblastoma development with broad implications for cancer pathogenesis.United States.     Effects of Astragalus Polysaccharide on Immune Responses of Porcine PBMC Stimulated with PRRSV or CSFV

    BACKGROUND: Astragalus polysaccharide (APS) has been used as an immunomodulator that can enhance immune responses, whereas the immunomodulatory effects of APS on porcine peripheral blood mononuclear cells (PBMCs) exposed to porcine reproductive and respiratory syndrome virus (PRRSV) and classical swine fever virus (CSFV) have not been investigated. METHODOLOGY/PRINCIPAL FINDINGS: Porcine PBMCs were cultured in complete RPMI media in the presence of the R98-strain of PRRSV (5×10(4) TCID(50)/ml) or C-strain of CSFV (10(3) TCID(50)/ml) with or without APS. The expression of mRNA for CD28, cytotoxic T-lymphocyte antigen 4 (CTLA-4), transforming growth factor-β (TGF-β), interleukin 2 (IL-2) and IL-10 was assayed by TaqMan real-time RT-PCR. The expression of mRNA for CD28 and CTLA-4 increased at 24 h after stimulation of PBMCs with CSFV and the increased production of CTLA-4 was confirmed by western blot analysis, whereas the increases were inhibited by the addition of APS. In addition, APS alone upregulated IL-2 and TGF-β mRNA expression in PBMCs and the addition of APS had the capacity to prevent a further increase in IL-2 mRNA expression in PBMCs during CSFV or PRRSV infection, but had no effect on TGF-β mRNA expression. The production of tumor necrosis factor-alpha (TNF-α) increased at 12 h after stimulation with PRRSV or CSFV, but not with PRRSV plus APS or CSFV plus APS, whereas the addition of APS to PBMCs infected with PRRSV or CSFV promoted IL-10 mRNA expression. CONCLUSIONS: We suggested that APS had immunomodulatory effects on cells exposed to PRRSV or CSFV. It might be that APS via different mechanisms affects the activities of immune cells during either PRRSV or CSFV infection. This possibility warrants further studies to evaluate whether APS would be an effective adjuvant in vaccines against PRRSV or CSFV

    Huangqi Injection (a Traditional Chinese Patent Medicine) for Chronic Heart Failure: A Systematic Review

    Chronic heart failure (CHF) is a global public health problem. Therefore, novel and effective drugs that show few side-effects are needed. Early literature studies indicated that Huangqi injection is one of the most commonly used traditional Chinese patent medicines for CHF in China. As a large number of clinical studies has been carried out and published, it is essential to evaluate the effectiveness and safety of Huangqi injection. Therefore, we carried out this systematic review under the support of the framework of the Joint Sino-Italian Laboratory (JoSIL).To evaluate the efficacy and safety of Huangqi injection for CHF according to the available scientific knowledge.An extensive search including PubMed, EMBASE, CBM, the Cochrane Library and Chinese literature databases was performed up to July 2008. Clinical trials regarding Huangqi injection for the treatment of CHF were searched for, irrespective of languages. The quality of each trial was assessed according to the Cochrane Reviewers' Handbook 5.0, and RevMan 5.0 provided by the Cochrane Collaboration and STATA 9.2 were used for data analysis.After selection of 1,205 articles, 62 RCTs and quasi-RCTs conducted in China and published in Chinese journals were included in the review. The methodological quality of the trials was low. In most trials inclusion and exclusion criteria were not specified. Furthermore, only one study evaluated the outcomes for drug efficacy after an adequate period of time. For these reasons and because of the different baseline characteristics we did not conduct a meta-analysis.Although available studies are not adequate to draw a conclusion on the efficacy and safety of Huangqi injection (a traditional Chinese patent medicine), we hope that our work could provide useful experience on further studies on Huangqi injections. The overall level of TCM clinical research needs to be improved so that the efficacy of TCM can be evaluated by the international community and possibly some TCM can enter into the international market
