    Determination of the thermodynamic temperature between 236 K and 430 K from speed of sound measurements in helium

    We report speed of sound measurements in helium at 273.16 K and at eight temperatures in the range between 236 K and 430 K. These results determine the difference (T  −  T 90) between the thermodynamic temperature T and its approximation T 90 by the International Temperature Scale of 1990 (ITS-90). The uncertainty of our measurements of (T  −  T 90) spans between a minimum of 0.25 mK near 247 K and a maximum of 0.89 mK at the freezing point of indium (429.75 K) with comparable contributions from the uncertainty of our acoustic determination of T and from the uncertainty of our laboratory realization of ITS-90. On the overlapping temperature ranges these results are consistent with other recent acoustic determinations of (T  −  T 90). We also present evidence that (T  −  T 90) can be determined with comparably small uncertainties by the alternative, time-saving procedure of measuring the speed-of-sound in helium using only a single, judiciously-chosen, pressure on each isotherm

    A determination of the molar gas constant R by acoustic thermometry in helium

    We have determined the acoustic and microwave frequencies of a misaligned spherical resonator maintained near the temperature of the triple point of water and filled with helium with carefully characterized molar mass M = (4.002 6032 ± 0.000 0015) g mol-1, with a relative standard uncertainty ur(M) = 0.37×10-6. From these data and traceable thermometry we estimate the speed of sound in our sample of helium at TTPW = 273.16 K and zero pressure to be u0 2 = (945 710.45 ± 0.85) m2 s-2 and correspondingly deduce the value R = (8.314 4743 ± 0.000 0088) J mol-1 K-1 for the molar gas constant. We estimate the value k = R/NA = (1.380 6508 ± 0.000 0015) × 10-23 J K-1 for the Boltzmann constant using the currently accepted value of the Avogadro constant NA. These estimates of R and k, with a relative standard uncertainty of 1.06 × 10-6, are 1.47 parts in 106 above the values recommended by CODATA in 2010

    Calcineurin controls expression of EAAT1/GLAST in mouse and human cultured astrocytes through dynamic regulation of protein synthesis and degradation

    Alterations in the expression of glutamate/aspartate transporter (GLAST) have been associated with several neuropathological conditions including Alzheimer\u2019s disease and epilepsy. However, the mechanisms by which GLAST expression is altered are poorly understood. Here we used a combination of pharmacological and genetic approaches coupled with quantitative PCR and Western blot to investigate the mechanism of the regulation of GLAST expression by a Ca2+ /calmodulin-activated phosphatase calcineurin (CaN). We show that treatment of cultured hippocampal mouse and fetal human astrocytes with a CaN inhibitor FK506 resulted in a dynamic modulation of GLAST protein expression, being downregulated after 24\u201348 h, but upregulated after 7 days of continuous FK506 (200 nM) treatment. Protein synthesis, as assessed by puromycin incorporation in neo-synthesized polypeptides, was inhibited already after 1 h of FK506 treatment, while the use of a proteasome inhibitor MG132 (1 \ub5M) shows that GLAST protein degradation was only suppressed after 7 days of FK506 treatment. In astrocytes with constitutive genetic ablation of CaN both protein synthesis and degradation were significantly inhibited. Taken together, our data suggest that, in cultured astrocytes, CaN controls GLAST expression at a posttranscriptional level through regulation of GLAST protein synthesis and degradation

    Quantification of the Chemical Chaperone 4-Phenylbutyric Acid (4-PBA) in Cell Culture Media via LC-HRMS: Applications in Fields of Neurodegeneration and Cancer

    In recent years, 4-phenylbutyric acid (4-PBA), an FDA-approved drug, has increasingly been used as a nonspecific chemical chaperone in vitro and in vitro, but its pharmacodynamics is still not clear. In this context, we developed and validated a Liquid Chromatography–High Resolution Mass Spectrometry (LC-HRMS) method to quantify 4-PBA in NeuroBasal-A and Dulbecco’s Modified Eagle widely used cell culture media. Samples were injected on a Luna® 3 µm PFP(2) 100 Å (100 × 2.0 mm) column maintained at 40 °C. Water and methanol both with 0.1% formic acid served as mobile phases in a step gradient mode. The mass acquisition was performed by selected ion monitoring (SIM) in negative mode for a total run time of 10.5 min at a flow rate of 0.300 mL/min. The analogue 4-(4-Nitrophenyl)-Butyric Acid served as internal standard. Validation parameters were verified according to FDA and EMA guidelines. The quantification ranges from 0.38–24 µM. Inter and intraday RSDs (Relative Standard Deviations) were within 15%. The developed LC-HRMS method allowed the estimation of 4-PBA absorption and adsorption kinetics in vitro in two experimental systems: (i) 4-PBA improvement of protein synthesis in an Alzheimer’s disease astrocytic cell model; and (ii) 4-PBA reduction of endoplasmic reticulum stress in thapsigargin-treated melanoma cell lines. © 2023 by the authors

    Equilibrium and nonequilibrium description of negative temperature states in a one-dimensional lattice using a wave kinetic approach

    We predict negative temperature states in the discrete nonlinear Schödinger (DNLS) equation as exact solutions of the associated wave kinetic equation. Within the wave kinetic approach, we define an entropy that results monotonic in time and reaches a stationary state, that is consistent with classical equilibrium statistical mechanics. We also perform a detailed analysis of the fluctuations of the actions at fixed wave numbers around their mean values. We give evidence that such fluctuations relax to their equilibrium behavior on a shorter timescale than the one needed for the spectrum to reach the equilibrium state. Numerical simulations of the DNLS equation are shown to be in agreement with our theoretical results. The key ingredient for observing negative temperatures in lattices characterized by two invariants is the boundedness of the dispersion relation

    Chromosome studies in Orchidaceae from Argentina

    The center of diversity of Argentinean orchids is in the northeast region of the country. Chromosome numbers and karyotype features of 43 species belonging to 28 genera are presented here. Five chromosome records are the first ones at the genus level; these taxa are Aspidogyne kuckzinskii (2n = 42), Eurystyles actinosophila (2n = 56), Skeptrostachys paraguayensis (2n = 46), Stigmatosema polyaden (2n = 40) and Zygostates alleniana (2n = 54). In addition, a chromosome number is presented for the first time for 15 species: Corymborkis flava (2n = 56), Cyclopogon callophyllus (2n = 28), C. oliganthus (2n = 64), Cyrtopodium hatschbachii (2n = 46), C. palmifrons (2n = 46), Galeandra beyrichii (2n = 54), Habenaria bractescens (2n = 44), Oncidium edwallii (2n = 42), O. fimbriatum (2n = 56), O. pubes (2n = 84), O. riograndense (2n = 56), Pelexia ekmanii (2n = 46), P. lindmanii (2n = 46) and Warrea warreana (2n = 48). For Oncidium longicornu (2n = 42), O. divaricatum (2n = 56) and Sarcoglottis fasciculata (2n = 46+1B?, 46+3B?), a new cytotype was found. Chromosome data support phylogenetic relationships proposed by previous cytological, morphologic and molecular analyses, and in all the cases cover some gaps in the South American literature on orchid chromosomes

    Very often the concept of smart city is strongly related to the flourishing of mobile applications, stressing the technological aspects and a top-down approach of high-tech centralized control systems capable of resolving all the urban issues, completely forgetting the essence of a city with its connected problems. The real challenge in future years will be a huge increase in the urban population and the changes this will produce in energy and resource consumption. It is fundamental to manage this phenomenon with clever approaches in order to guarantee a better management of resources and their sustainable access to present and future generations. This chapter develops some considerations on these aspects, trying to insert the technological issues within a framework closer to planning and with attention to the social impact
