    Antibacterial and phytochemical screening of Anethum graveolens, Foeniculum vulgare and Trachyspermum ammi

    <p>Abstract</p> <p>Background</p> <p><it>Anethum graveolens </it>Linn., <it>Foeniculum vulgare </it>Mill. and <it>Trachyspermum ammi </it>L. are widely used traditional medicinal plants to treat various ailments. To provide a scientific basis to traditional uses of these plants, their aqueous and organic seed extracts, as well as isolated phytoconstituents were evaluated for their antibacterial potential.</p> <p>Methods</p> <p>Antibacterial activity of aqueous and organic seed extracts was assessed using agar diffusion assay, minimum inhibitory concentration and viable cell count studies; and their antibacterial effect was compared with some standard antibiotics. The presence of major phytoconstituents was detected qualitatively and quantitatively. The isolated phytoconstituents were subjected to disc diffusion assay to ascertain their antibacterial effect.</p> <p>Results</p> <p>Hot water and acetone seed extracts showed considerably good antibacterial activity against all the bacteria except <it>Klebsiella pneumoniae </it>and one strain of <it>Pseudomonas aeruginosa</it>. Minimum inhibitory concentration for aqueous and acetone seed extracts ranged from 20–80 mg/ml and 5–15 mg/ml respectively. Viable cell count studies revealed the bactericidal nature of the seed extracts. Statistical analysis proved the better/equal efficacy of some of these seed extracts as compared to standard antibiotics. Phytochemical analysis showed the presence of 2.80 – 4.23% alkaloids, 8.58 – 15.06% flavonoids, 19.71 – 27.77% tannins, 0.55–0.70% saponins and cardiac glycosides.</p> <p>Conclusion</p> <p>Antibacterial efficacy shown by these plants provides a scientific basis and thus, validates their traditional uses as homemade remedies. Isolation and purification of different phytochemicals may further yield significant antibacterial agents.</p

    Developing an effective forest therapy program to manage academic stress in conservative societies: a multi-disciplinary approach

    We conducted a multi-disciplinary research to develop a forest therapy program that could manage academic stress in students. The first part of the study comprised of a survey to develop a forest therapy program based on the expectations of students, and involved 412 students aged 19–24 years (21.73 ± 2.33 years). The second part was a field study to determine the sustained effects the forest therapy program had on the blood pressure of students, involving twenty-nine students aged 21–23 years (21.83 ± 0.711). The survey determined that students were suffering from academic stress but their fear of societal stigma prevented them from seeking assistance. The majority (57.26%) expressed interest in forest therapy, and wanted a half-day forest therapy program during the weekend. Systolic (SBP) and diastolic (DBP) blood pressure were used as measurement indices for the field study, and was conducted before breakfast, lunch and dinner. Readings were taken 3 days before (baseline values), during, 3, 5 and 7 days after the forest therapy program. The blood pressure reading 3 days prior to the program served as a representation of the participants’ every blood pressure levels. When compared to the everyday blood pressure levels (124/81 mmHg), both the SBP and DBP significantly decreased during the forest therapy (117/77 mmHg), and the decrement maintained 3 days (114/77 mmHg) and 5 days after (118/79 mmHg). There was no significant decrease in blood pressure between the everyday levels and 7 days after. In conclusion, a half-day forest therapy program is able to decrease students’ SBP and DBP, and the decrements were maintained for 5 days. The exit survey reaffirmed the blood pressure results, whereby the participants believed that forest therapy had reduced their stress

    Antistaphylococcal and biofilm inhibitory activities of acetyl-11-keto-β-boswellic acid from Boswellia serrata

    <p>Abstract</p> <p>Background</p> <p>Boswellic acids are pentacyclic triterpenes, which are produced in plants belonging to the genus <it>Boswellia</it>. Boswellic acids appear in the resin exudates of the plant and it makes up 25-35% of the resin. β-boswellic acid, 11-keto-β-boswellic acid and acetyl-11-keto-β-boswellic acid have been implicated in apoptosis of cancer cells, particularly that of brain tumors and cells affected by leukemia or colon cancer. These molecules are also associated with potent antimicrobial activities. The present study describes the antimicrobial activities of boswellic acid molecules against 112 pathogenic bacterial isolates including ATCC strains. Acetyl-11-keto-β-boswellic acid (AKBA), which exhibited the most potent antibacterial activity, was further evaluated in time kill studies, postantibiotic effect (PAE) and biofilm susceptibility assay. The mechanism of action of AKBA was investigated by propidium iodide uptake, leakage of 260 and 280 nm absorbing material assays.</p> <p>Results</p> <p>AKBA was found to be the most active compound showing an MIC range of 2-8 μg/ml against the entire gram positive bacterial pathogens tested. It exhibited concentration dependent killing of <it>Staphylococcus aureus </it>ATCC 29213 up to 8 × MIC and also demonstrated postantibiotic effect (PAE) of 4.8 h at 2 × MIC. Furthermore, AKBA inhibited the formation of biofilms generated by <it>S. aureus </it>and <it>Staphylococcus epidermidis </it>and also reduced the preformed biofilms by these bacteria. Increased uptake of propidium iodide and leakage of 260 and 280 nm absorbing material by AKBA treated cells of <it>S aureus </it>indicating that the antibacterial mode of action of AKBA probably occurred via disruption of microbial membrane structure.</p> <p>Conclusions</p> <p>This study supported the potential use of AKBA in treating <it>S. aureus </it>infections. AKBA can be further exploited to evolve potential lead compounds in the discovery of new anti-Gram-positive and anti-biofilm agents.</p

    A cell based high-throughput screening approach for the discovery of new inhibitors of respiratory syncytial virus

    Background: Human respiratory syncytial virus (hRSV) is a highly contagious pathogen and is the most common cause of bronchiolitis and pneumonia for infants and children under one year of age. Worldwide, greater than 33 million children under five years of age are affected by hRSV resulting in three million hospitalizations and 200,000 deaths. However, severe lower respiratory tract disease may occur at any age, especially among the elderly or those with compromised cardiac, pulmonary, or immune systems. There is no vaccine commercially available. Existing therapies for the acute infection are ribavirin and the prophylactic humanized monoclonal antibody (Synagis® from MedImmune) that is limited to use in high risk pediatric patients. Thus, the discovery of new inhibitors for hRSV would be clinically beneficial. Results: We have developed and validated a 384-well cell-based, high-throughput assay that measures the cytopathic effect of hRSV (strain Long) in HEp-2 cells using a luminescent-based detection system for signal endpoint (Cell Titer Glo®). The assay is sensitive and robust, with Z factors greater than 0.8, signal to background greater than 35, and signal to noise greater than 24. Utilizing this assay, 313,816 compounds from the Molecular Libraries Small Molecule Repository were screened at 10 μM. We identified 7,583 compounds that showed greater than 22% CPE inhibition in the primary screen. The top 2,500 compounds were selected for confirmation screening and 409 compounds showed at least 50% inhibition of CPE and were considered active. We selected fifty-one compounds, based on potency, selectivity and chemical tractability, for further evaluation in dose response and secondary assays Several compounds had SI50 values greater than 3, while the most active compound displayed an SI50 value of 58.9. Conclusions: A robust automated luminescent-based high throughput screen that measures the inhibition of hRSV-induced cytopathic effect in HEp-2 cells for the rapid identification of potential inhibitors from large compound libraries has been developed, optimized and validated. The active compounds identified in the screen represent different classes of molecules, including aryl sulfonylpyrrolidines which have not been previously identified as having anti-hRSV activity

    A high-throughput, cell-based screen was used to identify chemotypes as inhibitors for human respiratory syncytial virus (hRSV). Optimization of a sulfonylpyrrolidine scaffold resulted in compound 5o that inhibited a virus-induced cytopathic effect in the entry stage of infection (EC50 = 2.3 ± 0.8 µM) with marginal cytotoxicity (CC50 = 30.9 ± 1.1 µM) and reduced viral titer by 100-fold.     Evaluation of ground-level and space-borne sensor as tools in monitoring nitrogen nutrition status in immature and mature oil palm

    Monitoring nitrogen (N) in oil palm is crucial for the production sustainability. The objective of this study is to examine the capability of visible (Vis), near infrared (NIR) and a combination of Vis and NIR (Vis + NIR) spectral indices acquired from different sensors for predicting foliar N content of different palm age groups. The N treatments varied from 0 to 2 kg per palm, subjected according to immature, young mature and prime mature classes. The Vis + NIR indices from the ground level-sensor that is green + red + NIR (G + R + NIR) was the best index for predicting N for immature palms (R2 = 0.91), while Vis indices blue + red (B + R) and Green Red Index from the space-borne sensor were significantly useful for N assessment of young and prime mature palms (R2 = 0.70 and 0.50), respectively. The application of vegetation indices for monitoring N status of oil palm is beneficial to examine extensive plantation areas

    <p>Abstract</p> <p>Background</p> <p>Boswellic acids mixture of triterpenic acids obtained from the oleo gum resin of <it>Boswellia serrata </it>and known for its effectiveness in the treatment of chronic inflammatory disease including peritumor edema. Boswellic acids have been extensively studied for a number of activities including anti inflammatory, antitumor, immunomodulatory, and inflammatory bowel diseases. The present study describes the antimicrobial activities of boswellic acid molecules against oral cavity pathogens. Acetyl-11-keto-β-boswellic acid (AKBA), which exhibited the most potent antibacterial activity, was further evaluated in time kill studies, mutation prevention frequency, postantibiotic effect (PAE) and biofilm susceptibility assay against oral cavity pathogens.</p> <p>Findings</p> <p>AKBA exhibited an inhibitory effect on all the oral cavity pathogens tested (MIC of 2-4 μg/ml). It exhibited concentration dependent killing of S<it>treptococcus mutans </it>ATCC 25175 up to 8 × MIC and also prevented the emergence of mutants of <it>S.mutans </it>ATCC 25175 at 8× MIC. AKBA demonstrated postantibiotic effect (PAE) of 5.7 ± 0.1 h at 2 × MIC. Furthermore, AKBA inhibited the formation of biofilms generated by <it>S.mutans </it>and <it>Actinomyces viscosus </it>and also reduced the preformed biofilms by these bacteria.</p> <p>Conclusions</p> <p>AKBA can be useful compound for the development of antibacterial agent against oral pathogens and it has great potential for use in mouthwash for preventing and treating oral infections.</p

    Obstetric professionals’ perceptions of non-invasive prenatal testing for Down syndrome: clinical usefulness compared with existing tests and ethical implications

    Background: While non-invasive prenatal testing (NIPT) for fetal aneuploidy is commercially available in many countries, little is known about how obstetric professionals in non-Western populations perceive the clinical usefulness of NIPT in comparison with existing first-trimester combined screening (FTS) for Down syndrome (DS) or invasive prenatal diagnosis (IPD), or perceptions of their ethical concerns arising from the use of NIPT. Methods: A cross-sectional survey among 327 obstetric professionals (237 midwives, 90 obstetricians) in Hong Kong. Results: Compared to FTS, NIPT was believed to: provide more psychological benefits and enable earlier consideration of termination of pregnancy. Compared to IPD, NIPT was believed to: provide less psychological stress for high-risk women and more psychological assurance for low-risk women, and offer an advantage to detect chromosomal abnormalities earlier. Significant differences in perceived clinical usefulness were found by profession and healthcare sector: (1) obstetricians reported more certain views towards the usefulness of NIPT than midwives and (2) professionals in the public sector perceived less usefulness of NIPT than the private sector. Beliefs about earlier detection of DS using NIPT were associated with ethical concerns about increasing abortion. Participants believing that NIPT provided psychological assurance among low-risk women were less likely to be concerned about ethical issues relating to informed decision-making and pre-test consultation for NIPT. Conclusions: Our findings suggest the need for political debate initially on how to ensure pregnant women accessing public services are informed about commercially available more advanced technology, but also on the potential implementation of NIPT within public services to improve access and equity to DS screening services
