    An all-electric single-molecule hybridisation detector for short DNA fragments

    In combining DNA nanotechnology and high-bandwidth single-molecule detection in nanopipettes, we demonstrate an all-electric, label-free hybridisation sensor for short DNA sequences (< 100 nt). Such short fragments are known to occur as circulating cell-free DNA in various bodily fluids, such as blood plasma and saliva, and have been identified as disease markers for cancer and infectious diseases. To this end, we use as a model system a 88-mer target from the RV1910c gene in Mycobacterium tuberculosis that is associated with antibiotic (isoniazid) resistance in TB. Upon binding to short probes attached to long carrier DNA, we show that resistive pulse sensing in nanopipettes is capable of identifying rather subtle structural differences, such as the hybridisation state of the probes, in a statistically robust manner. With significant potential towards multiplexing and high-throughput analysis, our study points towards a new, single-molecule DNA assay technology that is fast, easy to use and compatible with point of care environments

    Reverse micelles with spines: L-2 phases of surfactant ion-polyion complex salts, n-alcohols and water investigated by rheology, NMR diffusion and SAXS measurements

    Fundação de Amparo à Pesquisa do Estado de São Paulo (FAPESP)Homogeneous alcoholic isotropic solutions (L-2 phases) of "complex salts'' of hexadecyltrimethylammonium neutralized by polyacrylate CTAPA(n) (n = 30 or 6000), in the presence of water were examined at 25 degrees C by means of small-angle X-ray scattering (SAXS), rheology and NMR self-diffusion measurements for different n-alcohols (octanol, hexanol and butanol) and at varying water contents. The greater water solubility and the slower water self-diffusion coefficients in these L-2 phases, when compared to results with surfactant-free alcohols, suggested that these phases are composed of reverse aggregates with water in their cores. A comparatively rapid self-diffusion of the surfactant ion in butanol suggested a significant fraction of "free'' surfactant ions, dissociated from the reverse aggregates. Rheological data confirmed that the obtained viscoelastic properties were consistent with a system containing entangled elongated micelles, whose properties were controlled by the polyion chain length. NMR diffusion measurements for complex salt solutions with the shorter counter-polyion (CTAPA(30)) produced estimates of the aggregate lengths that were close to the extended length of one PA(30) chain.     Chitosan/virgin-coconut-oil-based system enriched with cubosomes: a 3D drug-delivery approach

    Emulsion-based systems that combine natural polymers with vegetable oils have been identified as a promising research avenue for developing structures with potential for biomedical applications. Herein, chitosan (CHT), a natural polymer, and virgin coconut oil (VCO), a resource obtained from coconut kernels, were combined to create an emulsion system. Phytantriol-based cubosomes encapsulating sodium diclofenac, an anti-inflammatory drug, were further dispersed into CHT/VCO- based emulsion. Then, the emulsions were frozen and freeze-dried to produce scaffolds. The scaffolds had a porous structure ranging from 20.4 to 73.4 µm, a high swelling ability (up to 900%) in PBS, and adequate stiffness, notably in the presence of cubosomes. Moreover, a well-sustained release of the entrapped diclofenac in the cubosomes into the CHT/VCO-based system, with an accumulated release of 45 ± 2%, was confirmed in PBS, compared to free diclofenac dispersed (80 ± 4%) into CHT/VCO-based structures.     Self-paced reaching after stroke: a quantitative assessment of longitudinal and directional sensitivity using the H-Man planar robot for upper limb neurorehabilitation

    Technology aided measures offer a sensitive, accurate and time-efficient approach for the assessment of sensorimotor function after neurological insult compared to standard clinical assessments. This study investigated the sensitivity of robotic measures to capture differences in planar reaching movements as a function of neurological status (stroke, healthy), direction (front, ipsilateral, contralateral), movement segment (outbound, inbound), and time (baseline, post-training, 2-week follow-up) using a planar, two-degrees of freedom, robotic-manipulator (H-Man). Twelve chronic stroke (age: 55 ± 10.0 years, 5 female, 7 male, time since stroke: 11.2 ± 6.0 months) and nine aged-matched healthy participants (age: 53 ± 4.3 years, 5 female, 4 male) participated in this study. Both healthy and stroke participants performed planar reaching movements in contralateral, ipsilateral and front directions with the H-Man, and the robotic measures, spectral arc length (SAL), normalized time to peak velocities (TpeakN), and root-mean square error (RMSE) were evaluated. Healthy participants went through a one-off session of assessment to investigate the baseline. Stroke participants completed a 2-week intensive robotic training plus standard arm therapy (8 × 90 min sessions). Motor function for stroke participants was evaluated prior to training (baseline, week-0), immediately following training (post-training, week-2), and 2-weeks after training (follow-up, week-4) using robotic assessment and the clinical measures Fugl-Meyer Assessment (FMA), Activity-Research-Arm Test (ARAT), and grip-strength. Robotic assessments were able to capture differences due to neurological status, movement direction, and movement segment. Movements performed by stroke participants were less-smooth, featured longer TpeakN, and larger RMSE values, compared to healthy controls. Significant movement direction differences were observed, with improved reaching performance for the front, compared to ipsilateral and contralateral movement directions. There were group differences depending on movement segment. Outbound reaching movements were smoother and featured longer TpeakN values than inbound movements for control participants, whereas SAL, TpeakN, and RMSE values were similar regardless of movement segment for stroke patients. Significant change in performance was observed between initial and post-assessments using H-Man in stroke participants, compared to conventional scales which showed no significant difference. Results of the study indicate the potential of H-Man as a sensitive tool for tracking changes in performance compared to ordinal scales (i.e., FM, ARAT)

    Coordenação de Aperfeiçoamento de Pessoal de Nível Superior (CAPES)Microbial consortia recuperated of crude oil samples from the Pampo Sul field, Campos Basin, RJ, produced biosurfactants in mineral media containing glucose and 9,10-dihydrophenantrene, n-nonadecane, nonadecanoic acid, slightly biodegraded crude oil (P1) or heavily biodegraded crude oil (P2) as carbon sources. The production of exopolimeric substances (EPS) and petroleum biodegradation do not necessarily occur simultaneously. The EPS analyses by infrared and eletronspray ionization mass spectrometry (ESI-MS) revealed a mixture of surfactine isoforms. The biosurfactants reduced the surface tension of water and Zinder medium from 72.4 and 55.7 to 28.6 mN m(-1).     Speculation on the origin of sub-baseline excursions of CH4 at Cape Grim

    The Advanced Global Atmospheric Gases Experiment (AGAGE) program has historically measured in situ methane (CH4 ) at Cape Grim via gas chromatography with flame ionization detection (GC-FID) in 40 minutely grab samples. By adding continuous, high precision in situ measurements of CH4 (Picarro cavity ring-down spectroscopy [CRDS]) at both Cape Grim, Tasmania, and Casey, Antarctica, a new feature has become apparent in the Cape Grim CH4 record. During the austral summer (December to February), the Cape Grim CH4 record periodically drops below baseline. For example, in Figure 1, a number of sustained episodes of depressed CH4 concentration can be seen below the baseline selected data shown in red. Notably, these episodes are also seen in the GC-FID record. In this presentation, we examine these sub-baseline excursions of CH4 . In conjunction with meteorology and a variety of other chemical species measured at Cape Grim, including radon, ozone, hydrogen and ethane, we speculate on a number of possible mechanisms that might be responsible for these dips in CH4 mixing ratio

    Luminous Red Galaxy Clustering at z~0.7 - First Results using AAOmega

    We report on the AAT-AAOmega LRG Pilot observing run to establish the feasibility of a large spectroscopic survey using the new AAOmega instrument. We have selected Luminous Red Galaxies (LRGs) using single epoch SDSS riz-photometry to i<20.5 and z<20.2. We have observed in 3 fields including the COSMOS field and the COMBO-17 S11 field, obtaining a sample of ~600 redshift z>=0.5 LRGs. Exposure times varied from 1 - 4 hours to determine the minimum exposure for AAOmega to make an essentially complete LRG redshift survey in average conditions. We show that LRG redshifts to i<20.5 can measured in approximately 1.5hr exposures and present comparisons with 2SLAQ and COMBO-17 (photo-)redshifts. Crucially, the riz selection coupled with the 3-4 times improved AAOmega throughput is shown to extend the LRG mean redshift from z=0.55 for 2SLAQ to z=0.681+/- 0.005 for riz-selected LRGs. This extended range is vital for maximising the S/N for the detection of the baryon acoustic oscillations (BAOs). Furthermore, we show that the amplitude of LRG clustering is s_0 = 9.9+/-0.7 h^-1 Mpc, as high as that seen in the 2SLAQ LRG Survey. Consistent results for the real-space amplitude are found from projected and semi-projected correlation functions. This high clustering amplitude is consistent with a long-lived population whose bias evolves as predicted by a simple ``high-peaks'' model. We conclude that a redshift survey of 360 000 LRGs over 3000deg^2, with an effective volume some 4 times bigger than previously used to detect BAO with LRGs, is possible with AAOmega in 170 nights.Comment: 12 pages, 7 figures, 8 tables, minor changes, matches published versio
