    Regional differences in store-operated Ca2+Ā entry in the epithelium of the intact human lens

    An elevated level of Ca2+ is an important factor in cataract, yet precisely how Ca2+ enters the lens is unknown. Lens epithelial cells contain a range of G-proteinā€“coupled receptors and receptor tyrosine kinases that induce increases in intracellular Ca2+. Receptor-associated Ca2+ influx is, therefore, likely to be an important route for Ca2+ influx to the lens. The authors investigated stimulated and passive Ca2+ influx in in situ human lens epithelium. Ca2+ changes in equatorial (E) and central anterior (CA) epithelial cells were monitored with the use of a Ca2+ indicator (Fluo4) and confocal microscopy. Gene expression was monitored by RT-PCR and immunoblotting. Adenosine triphosphate (ATP) induced Ca2+ responses that were smaller in CA than E. Ca2+ store depletion, using ATP (100 ĀµM) or thapsigargin (1 ĀµM), revealed greater relative store capacity and Ca2+ influx in E. Ca2+ influx was blocked by La3+ (0.5 ĀµM) in both regions. Unstimulated Ca2+ influx was greater in E than CA. Greater expression of Orai1 and STIM1 was detected in E than in CA. Greater Ca2+ store capacity and Ca2+ influx in E compared with CA reflects underlying differences in proliferation and differentiation between the regions. The relatively small resting Ca2+ influx in CA epithelium suggests that store-operated Ca2+ entry (SOCE) is the main route of Ca2+ influx in these cells. Greater resting influx and SOCE in E cells suggests that these are a major route for Ca2+ influx into the lens. Increased expression of Orai1 and STIM1 in E could account for the differences in Ca2+ entry. Receptor activation will modulate Ca2+ influx, and inappropriate activity may contribute to cortical cataract

    Noncanonical spike-related BOLD responses in focal epilepsy

    Till now, most studies of the Blood Oxygen Level-Dependent (BOLD) response to interictal epileptic discharges (IED) have assumed that its time course matches closely to that of brief physiological stimuli, commonly called the canonical event-related haemodynamic response function (canonical HRF). Analyses based on that assumption have produced significant response patterns that are generally concordant with prior electroclinical data. In this work, we used a more flexible model of the event-related response, a Fourier basis set, to investigate the presence of other responses in relation to individual IED in 30 experiments in patients with focal epilepsy. We found significant responses that had a noncanonical time course in 37% of cases, compared with 40% for the conventional, canonical HRF-based approach. In two cases, the Fourier analysis suggested activations where the conventional model did not. The noncanonical activations were almost always remote from the presumed generator of epileptiform activity. In the majority of cases with noncanonical responses, the noncanonical responses in single-voxel clusters were suggestive of artifacts. We did not find evidence for IED-related noncanonical HRFs arising from areas of pathology, suggesting that the BOLD response to IED is primarily canonical. Noncanonical responses may represent a number of phenomena, including artefacts and propagated epileptiform activity

    Implementation and evaluation of simultaneous video-electroencephalography and functional magnetic resonance imaging

    The objective of this study was to demonstrate that the addition of simultaneous and synchronised video to electroencephalography (EEG)-correlated functional magnetic resonance imaging (fMRI) could increase recorded information without data quality reduction. We investigated the effect of placing EEG, video equipment and their required power supplies inside the scanner room, on EEG, video and MRI data quality, and evaluated video-EEG-fMRI by modelling a hand motor task. Gradient-echo, echo-planner images (EPI) were acquired on a 3-T MRI scanner at variable camera positions in a test object [with and without radiofrequency (RF) excitation], and human subjects. EEG was recorded using a commercial MR-compatible 64-channel cap and amplifiers. Video recording was performed using a two-camera custom-made system with EEG synchronization. An in-house script was used to calculate signal to fluctuation noise ratio (SFNR) from EPI in test object with variable camera positions and in human subjects with and without concurrent video recording. Five subjects were investigated with video-EEG-fMRI while performing hand motor task. The fMRI time series data was analysed using statistical parametric mapping, by building block design general linear models which were paradigm prescribed and video based. Introduction of the cameras did not alter the SFNR significantly, nor did it show any signs of spike noise during RF off conditions. Video and EEG quality also did not show any significant artefact. The Statistical Parametric Mapping{T} maps from video based design revealed additional blood oxygen level-dependent responses in the expected locations for non-compliant subjects compared to the paradigm prescribed design. We conclude that video-EEG-fMRI set up can be implemented without affecting the data quality significantly and may provide valuable information on behaviour to enhance the analysis of fMRI data

    Correlated radial velocity and X-ray variations in HD 154791/4U 1700+24

    We present evidence for approximately 400-d variations in the radial velocity of HD 154791 (V934 Her), the suggested optical counterpart of 4U 1700+24. The variations are correlated with the previously reported approximately 400 d variations in the X-ray flux of 4U 1700+24, which supports the association of these two objects, as well as the identification of this system as the second known X-ray binary in which a neutron star accretes from the wind of a red giant. The HD 154791 radial velocity variations can be fit with an eccentric orbit with period 404 +/- 3 d, amplitude K=0.75 +/- 0.12 km/s and eccentricity e=0.26 +/- 0.15. There are also indications of variations on longer time scales >~ 2000 d. We have re-examined all available ASM data following an unusually large X-ray outburst in 1997-98, and confirm that the 1-d averaged 2-10 keV X-ray flux from 4U 1700+24 is modulated with a period of 400 +/- 20 d. The mean profile of the persistent X-ray variations was approximately sinusoidal, with an amplitude of 0.108 +/- 0.012 ASM count/s (corresponding to 31% rms). The epoch of X-ray maximum was approximately 40 d after the time of periastron according to the eccentric orbital fit. If the 400 d oscillations from HD 154791/4U 1700+24 are due to orbital motion, then the system parameters are probably close to those of the only other neutron-star symbiotic-like binary, GX 1+4.     Measurement of the SOC State Specific Heat in ^4He

    When a heat flux Q is applied downward through a sample of liquid 4He near the lambda transition, the helium self organizes such that the gradient in temperature matches the gravity induced gradient in Tlambda. All the helium in the sample is then at the same reduced temperature tSOC = ((T[sub SOC] - T[sub lambda])/T[sub lambda]) and the helium is said to be in the Self-Organized Critical (SOC) state. We have made preliminary measurements of the 4He SOC state specific heat, C[del]T(T(Q)). Despite having a cell height of 2.54 cm, our results show no difference between C[del]T and the zero-gravity 4He specific heat results of the Lambda Point Experiment (LPE) [J.A. Lipa et al., Phys. Rev. B, 68, 174518 (2003)] over the range 250 to 450 nK below the transition. There is no gravity rounding because the entire sample is at the same reduced temperature tSOC(Q). Closer to Tlambda the SOC specific heat falls slightly below LPE, reaching a maximum at approximately 50 nK below Tlambda, in agreement with theoretical predictions [R. Haussmann, Phys. Rev.     Risk factors for failure of outpatient parenteral antibiotic therapy (OPAT) in infective endocarditis

    Objectives: To identify risk factors for failure of outpatient antibiotic therapy (OPAT) in infective endocarditis (IE). Patients and methods: We identified IE cases managed at a single centre over 12 years from a prospectively maintained database. ā€˜OPAT failureā€™ was defined as unplanned readmission or antibiotic switch due to adverse drug reaction or antibiotic resistance. We analysed patient and disease-related risk factors for OPAT failure by univariate and multivariate logistic regression. We also retrospectively collected follow-up data on adverse disease outcome (defined as IE-related death or relapse) and performed Kaplanā€“Meier survival analysis up to 36 months following OPAT. Results: We identified 80 episodes of OPAT in IE. Failure occurred in 25/80 episodes (31.3%). On multivariate analysis, cardiac or renal failure [pooled OR 7.39 (95% CI 1.84ā€“29.66), Pā€Š=ā€Š0.005] and teicoplanin therapy [OR 8.69 (95% CI 2.01ā€“37.47), Pā€Š=ā€Š0.004] were independently associated with increased OPAT failure. OPAT failure with teicoplanin occurred despite therapeutic plasma levels. OPAT failure predicted adverse disease outcome up to 36 months (Pā€Š=ā€Š0.016 log-rank test). Conclusions: These data caution against selecting patients with endocarditis for OPAT in the presence of cardiac or renal failure and suggest teicoplanin therapy may be associated with suboptimal OPAT outcomes. Alternative regimens to teicoplanin in the OPAT setting should be further investigated

    We study a three-dimensional analogue to the well-known graph visualization approach known as arc diagrams. We provide several algorithms that achieve good angular resolution for 3D arc diagrams, even for cases when the arcs must project to a given 2D straight-line drawing of the input graph.     An X-Ray Jet from a White Dwarf - Detection of the Collimated Outflow from CH Cygni with Chandra

    Most symbiotic stars consist of a white dwarf accreting material from the wind of a red giant. An increasing number of these objects have been found to produce jets. Analysis of archival Chandra data of the symbiotic system CH Cygni reveals faint extended emission to the south, aligned with the optical and radio jets seen in earlier HST and VLA observations. CH Cygni thus contains only the second known white dwarf with an X-ray jet, after R Aquarii.     Effect of Inhomogeneous Heat Flow on the Enhancement of Heat Capacity in Helium-II by Counterflow near TĪ»

    In 2000 Harter et al. reported the first measurements of the enhancement of the heat capacity Ī”CQ[equivalent]C(Q)-C(Q=0) of helium-II transporting a heat flux density Q near TĪ». Surprisingly, their measured Ī”CQ was ~7ā€“12 times larger than predicted, depending on which theory was assumed. In this report we present a candidate explanation for this discrepancy: unintended heat flux inhomogeneity. Because C(Q) should diverge at a critical heat flux density Qc, homogeneous heat flow is required for an accurate measurement. We present results from numerical analysis of the heat flow in the Harter et al. cell indicating that substantial inhomogeneity occurred. We determine the effect of the inhomogeneity on Ī”CQ and find rough agreement with the observed disparity between prediction and measurement
