    Psychometrics of health-related quality of life questionnaires in bronchiectasis:A systematic review and meta-analysis

    INTRODUCTION: Understanding the psychometric properties of health related quality of life (HRQoL) questionnaires can help inform selection in clinical trials. OBJECTIVE: To assess the psychometric properties of HRQoL questionnaires in bronchiectasis. METHODS: A literature search was conducted. HRQoL questionnaires were assessed for psychometric properties (reliability, validity, minimal clinically important difference (MCID), floor/ceiling effects). Meta-analyses assessed the associations of HRQoL with clinical measures and responsiveness of HRQoL in clinical trials. RESULTS: 166 studies and 12 HRQoL questionnaires were included. The BHQ, LCQ, CAT and SF-36 had good internal consistency in all domains reported (Cronbach's α≥0.7) across all studies and the QoL-B, SGRQ, CRDQ and SOLQ had good internal consistency in all domains in the majority of (but not all) studies. The BHQ, SGRQ, LCQ and CAT had good test-retest reliability in all domains reported ((intraclass correlation coefficient) ICC ≥0.7) across all studies and the QoL-B, CRDQ and SOLQ had good test-retest reliability in all domains in the majority of (but not all) studies. HRQoL questionnaires were able to discriminate between demographics, important markers of clinical status, disease severity, exacerbations and bacteriology. For HRQoL responsiveness, there was a difference between the treatment and placebo effect. CONCLUSION: SGRQ was the most widely used HRQoL questionnaire in bronchiectasis studies and it had good psychometric properties, however good psychometric data are growing on bronchiectasis specific HRQoL questionnaires, QoL-B and BHQ. Future studies should focus on the medium-long term test-retest reliability, responsiveness and MCID in these HRQoL questionnaires which show potential in bronchiectasis

    Impact of water hardness on oxytetracycline oral bioavailability in fed and fasted piglets

    Water hardness is a critical factor that affects oxytetracycline dissolution by chelation with cations. These interactions may lead to impaired dosing and consequently decrease absorption. Moreover, feed present in gastrointestinal tract may interact with antibiotic and alter pharmacokinetic parameters. In the present study, dissolution profiles of an oxytetracycline veterinary formulation were assessed in purified, soft and hard water. Furthermore, oxytetracycline absolute bioavailability, after oral administration of the drug dissolved in soft or hard water, was evaluated in fed and fasted piglets. A maximum dissolution of 86% and 80% was obtained in soft and hard water, respectively, while in purified water dissolution was complete. Results from in vivo study reconfirmed oxytetracycline´s very low oral bioavailability. The greatest values were attained when antibiotic was dissolved in soft water and in fasted animals. Statistically significant lower absolute bioavailability was achieved when hard water was used and/or animals were fed. Moreover, Cmax attained in all treatments was lower than MIC90 of most important swine pathogens. For these reasons, the oral use of OTC formulations, that have demonstrated low oral bioavailability, should be avoided to treat systemic diseases in pigs.Fil: Decundo, Julieta María.     Isolation and characterization of a novel 2-sec-butylphenol-degrading bacterium Pseudomonas sp. strain MS-1

    A novel bacterium capable of utilizing 2-sec-butylphenol as the sole carbon and energy source, Pseudomonas sp. strain MS-1, was isolated from freshwater sediment. Within 30 h, strain MS-1 completely degraded 1.5 mM 2-sec-butylphenol in basal salt medium, with concomitant cell growth. A pathway for the metabolism of 2-sec-butylphenol by strain MS-1 was proposed on the basis of the identification of 3 internal metabolites—3-sec-butylcatechol, 2-hydroxy-6-oxo-7-methylnona-2,4-dienoic acid, and 2-methylbutyric acid—by gas chromatography-mass spectrometry analysis. Strain MS-1 degraded 2-sec-butylphenol through 3-sec-butylcatechol along a meta-cleavage pathway. Degradation experiments with various alkylphenols showed that the degradability of alkylphenols by strain MS-1 depended strongly on the position (ortho ≫ meta = para) of the alkyl substitute, and that strain MS-1 could degrade 2-alkylphenols with various sized and branched alkyl chain (o-cresol, 2-ethylphenol, 2-n-propylphenol, 2-isopropylphenol, 2-sec-butylphenol, and 2-tert-butylphenol), as well as a dialkylphenol (namely, 6-tert-butyl-m-cresol)

    Semidiurnal temperature changes caused by tidal front movements in the warm season in seabed habitats on the Georges Bank northern margin and their ecological implications

Previous studies demonstrated a strong tidal-mixing front during the warm season on the northern bank margin between thermally stratified water in the Gulf of Maine and mixed water on the bank. Tides transport warm water off the bank during flood tide and cool gulf water onto the bank during ebb tide. During 10 days in August 2009, we mapped frontal temperatures in five study areas along ~100 km of the bank margin. The seabed “frontal zone”, where temperature changed with frontal movment, experienced semidiurnal temperature maxima and minima. The tidal excursion of the frontal boundary between stratified and mixed water ranged 6 to 10 km. This “frontal boundary zone” was narrower than the frontal zone. Along transects perpendicular to the bank margin, seabed temperature change at individual sites ranged from 7.0°C in the frontal zone to 0.0°C in mixed bank water. At time series in frontal zone stations, changes during tidal cycles ranged from 1.2 to 6.1°C. The greatest rate of change (−2.48°C hr−1) occurred at mid-ebb. Geographic plots of seabed temperature change allowed the mapping of up to 8 subareas in each study area. The magnitude of temperature change in a subarea depended on its location in the frontal zone. Frontal movement had the greatest effect on seabed temperature in the 40 to 80 m depth interval. Subareas experiencing maximum temperature change in the frontal zone were not in the frontal boundary zone, but rather several km gulfward (off-bank) of the frontal boundary zone.     The development of an in-situ geochronology capability for Mars and other planetary surfaces has the potential to fundamentally change our understanding of the evolution of terrestrial bodies in the Solar System. For Mars specifically, many of our most basic scientific questions about the geologic history of the planet require accurate knowledge of the absolute time at which an event or process took place. For instance, what was the age and rate of early Martian climate change faithfully recorded in the mineralogy and morphology of surface lithologies (e.g., [1])? Currently, our only means of assessing the absolute age of a surface on a planetary body is through the use of crater counting statistics. This technique is fraught with uncertainty for planets with active geologic surfaces, on the order of billions of years in some cases (e.g., [2]). Accordingly, there is much room for improvement in our understanding of the absolute chronology of the surfaces of rocky planetary bodies

    Why bluefin tuna have warm tummies: temperature effect on trypsin and chymotrypsin

    abstract: Giant bluefin tuna (Thunnus thynnus) warm their viscera during and after a meal. The caecum of a 500-kg bluefin weighs about 9 kg and contains about 20 000 pyloric caeca, each about 10 cm long and 1.5 mm diameter. Trypsin was assayed with α-benzoyl-DL-arginine-p-nitroanilide HCl and chymotrypsin with glutaryl-L-phenylalanine-p-nitroaniline. The effects of pH on specific activity over the range 7.5 to 9.5 were negligible relative to temperature effects. Specific activity and maximal reaction velocity extrapolated from a Lineweaver-Burke plot (Vmax) increased with an increase in temperature in a similar fashion, whereas Km was constant over the same temperature range. The advantage of the warm caecum is that protein is digested in about one-third the time, so that these tuna can process about three times as much food per day

    Do suspended sediments modulate the effects of octylphenol on rainbow trout?

    A system was devised which allows particles to remain in suspension in a conventional 60 L aquarium without undue disturbance to resident fish. Using this system, juvenile rainbow trout were exposed for one week to 4-tert-octylphenol (OP, 10-1000 µg/L) with or without the presence of suspended sediments (10-20 mg/L; collected from the River Calder, UK). About 8 % of the added OP partitioned to the solid phase. Vitellogenin levels were determined in the plasma of the exposed rainbow trout and showed a dose-dependent increase with regards to OP exposure concentration. Considerable variation in the vitellogenin response was observed between separate runs with the same OP concentration. There was no statistically significant (at P<0.05) difference in plasma VTG levels between the OP treatments with or without suspended sediments. This suggests that the dissolved concentration is the key factor and natural suspended sediment neither protects against, nor exacerbates, the endocrine disrupting effect of OP on fish
