    Performance of AAOmega: the AAT multi-purpose fibre-fed spectrograph

    AAOmega is the new spectrograph for the 2dF fibre-positioning system on the Anglo-Australian Telescope. It is a bench-mounted, double-beamed design, using volume phase holographic (VPH) gratings and articulating cameras. It is fed by 392 fibres from either of the two 2dF field plates, or by the 512 fibre SPIRAL integral field unit (IFU) at Cassegrain focus. Wavelength coverage is 370 to 950nm and spectral resolution 1,000-8,000 in multi-Object mode, or 1,500-10,000 in IFU mode. Multi-object mode was commissioned in January 2006 and the IFU system will be commissioned in June 2006. The spectrograph is located off the telescope in a thermally isolated room and the 2dF fibres have been replaced by new 38m broadband fibres. Despite the increased fibre length, we have achieved a large increase in throughput by use of VPH gratings, more efficient coatings and new detectors - amounting to a factor of at least 2 in the red. The number of spectral resolution elements and the maximum resolution are both more than doubled, and the stability is an order of magnitude better. The spectrograph comprises: an f/3.15 Schmidt collimator, incorporating a dichroic beam-splitter; interchangeable VPH gratings; and articulating red and blue f/1.3 Schmidt cameras. Pupil size is 190mm, determined by the competing demands of cost, obstruction losses, and maximum resolution.     Traits, habitats, and clades: Identifying traits of potential importance to environmental filtering

    Environmental filtering is a fundamental process in the ecological assembly of communities. Recently developed phylogenetic tools identify patterns associated with environmental filtering across whole communities. Here we introduce a novel method that allows the detection of traits involved in the environmental filtering of species from specific clades in specific habitat types. Our approach identifies nonindependent trait/habitat/clade (THC) associations and also provides a framework for detecting clearly defined two‐way trait/clade, trait/habitat, and clade/habitat associations. The THC method relies on exact binomial tests and differentiates THC associations resulting from a three‐way interaction from those that are generated by one or more underlying significant two‐way interactions. It can also detect THC associations for which there are no significant two‐way associations (trait/habitat, trait/clade, clade/habitat). To illustrate the THC method, we examine plant pollination and dispersal traits from six habitat types in a fragmented Costa Rican landscape. Results suggest that these traits are not widely important for the environmental filtering of most clades in this landscape, but animal dispersal and insect pollination are involved in the filtering of monocots and the Piperaceae in rain forest understory

    Effects of habitat and livestock on nest productivity of the Asian houbara Chlamydotis macqueenii in Bukhara Province, Uzbekistan

    To inform population support measures for the unsustainably hunted Asian houbara Chlamydotis macqueenii (IUCN Vulnerable) we examined potential habitat and land-use effects on nest productivity in the Kyzylkum Desert, Uzbekistan. We monitored 177 nests across different semi-arid shrub assemblages (clay-sand and salinity gradients) and a range of livestock densities (0–80 km-2). Nest success (mean 51.4%, 95% CI 42.4–60.4%) was similar across four years; predation caused 85% of those failures for which the cause was known, and only three nests were trampled by livestock. Nesting begins within a few weeks of arrival when food appears scarce, but later nests were more likely to fail owing to the emergence of a key predator, suggesting foraging conditions on wintering and passage sites may be important for nest productivity. Nest success was similar across three shrub assemblages and was unrelated to landscape rugosity, shrub frequency or livestock density, but was greater with taller mean shrub height (range 13–67 cm) within 50 m. Clutch size (mean = 3.2 eggs) and per-egg hatchability in successful nests (87.5%) did not differ with laying date, shrub assemblage or livestock density. We therefore found no evidence that livestock density reduced nest productivity across the range examined, while differing shrub assemblages appeared to offer similar habitat quality. Asian houbara appear well-adapted to a range of semi-desert habitats and tolerate moderate disturbance by pastoralism. No obvious in situ mitigation measures arise from these findings, leaving regulation and control as the key requirement to render hunting sustainable

    “Read and Mark in Holy Scripture”: Reassessing the Scripture Readings of "A Festival of Nine Lessons and Carols" in a Post-Shoah World

    The Christian Eve service, A Festival of Nine Lessons and Carols, broadcast yearly around the world by King’s College, Cambridge, has nine lessons (i.e., biblical passages) read during the service. Today, many churches around the world celebrate the Christmas season by reading these same nine passages: Genesis 3:8-15, 17-19; Genesis 22:15-18; Isaiah 9:2, 6-7; Isaiah 11:1-4a, 6-9; Luke 1:26-35, 38; Luke 2:1, 3-7; Luke 2:8-16; Matthew 2:1-12; John 1:1-14. This article unfolds in three sections as it critically examines the nine biblical lessons to problematize their selection and ordering within this service. First, I place these scripture readings within the literary context of the biblical canon and the liturgical context of the service to understand how these readings weave a particular theological story within the overall service. I focus on how the service frames the lessons liturgically, examining its use of an introductory interpretive sentence at the beginning of each lesson as an exercise in liturgical hermeneutics of the bible. Second, I critique the assumed theology and hermeneutics behind these biblical selections using the theological concept of the Christmas standard canonical narrative and argue that these specific readings in this particular sequence (i.e., the reading of the biblical passages in canonical order from Genesis to Isaiah to the three gospels) are ethically and theologically problematic in a post-Holocaust world. These lessons individually and collectively posit an implicit, problematic theology of the bible as a whole and of the Hebrew Bible/Old Testament in particular, and its understanding of the grand story of Scripture. Third, I suggest new guidelines for selecting lessons and examples for a reimagined service. I explore how Christian leaders might rethink the role of Scripture in this service

    Comparisons of the motivational sources of high school business students

    The purpose of this study was to determine and compare the motivational sources of students based on gender, course nature, and grade level. The respondents include a sample of the students in the Business Department at Bearden High School (Knoxville, TN) during the last nine weeks of the spring semester of 2001. Keyboarding classes during the first nine weeks were not included in the sample. Each respondent reported on his or her motivational sources using the survey instrument developed by the researcher. Major findings of the study were (a) that the respondents had an optimal combination of extrinsic and intrinsic motivational sources, which the teacher can foster and build upon within instruction and (b) difference in motivational sources was most pronounced between freshmen and seniors. Teachers who have a combination of these grade levels in the classroom should recognize and accommodate them within business courses

    Aptamer-based multiplexed proteomic technology for biomarker discovery

    Interrogation of the human proteome in a highly multiplexed and efficient manner remains a coveted and challenging goal in biology. We present a new aptamer-based proteomic technology for biomarker discovery capable of simultaneously measuring thousands of proteins from small sample volumes (15 [mu]L of serum or plasma). Our current assay allows us to measure ~800 proteins with very low limits of detection (1 pM average), 7 logs of overall dynamic range, and 5% average coefficient of variation. This technology is enabled by a new generation of aptamers that contain chemically modified nucleotides, which greatly expand the physicochemical diversity of the large randomized nucleic acid libraries from which the aptamers are selected. Proteins in complex matrices such as plasma are measured with a process that transforms a signature of protein concentrations into a corresponding DNA aptamer concentration signature, which is then quantified with a DNA microarray. In essence, our assay takes advantage of the dual nature of aptamers as both folded binding entities with defined shapes and unique sequences recognizable by specific hybridization probes. To demonstrate the utility of our proteomics biomarker discovery technology, we applied it to a clinical study of chronic kidney disease (CKD). We identified two well known CKD biomarkers as well as an additional 58 potential CKD biomarkers. These results demonstrate the potential utility of our technology to discover unique protein signatures characteristic of various disease states. More generally, we describe a versatile and powerful tool that allows large-scale comparison of proteome profiles among discrete populations. This unbiased and highly multiplexed search engine will enable the discovery of novel biomarkers in a manner that is unencumbered by our incomplete knowledge of biology, thereby helping to advance the next generation of evidence-based medicine
