The Association of Semen Factors with the Recovery of \u3ci\u3eUreaplasma urealyticum\u3c/i\u3e
Fifty-six semen samples from males under examination for suspected subfertility were examined to determine if alterations in specific semen parameters could be associated with the presence or absence or Ureaplasma urealvticum. Twenty-seven (48.2%) of the specimens cultured positive for U· urealyticum in U9B urease broth and A-7 differential agar. Both positive and negative specimens have a normal average motility rating. Overall sperm counts (millions per ml) were slightly lower and ejaculate volumes higher in U. urealvticum positive samples. Morphological abnormalicies were not found more frequently in U. urealyticum positive samples. No statistically significant difference between seminal parameters of U. urealyticum positive and U. urealyticum negative samples was noted with one way analysis of variance test. Non parametric analysis produced similar non-significant results
Meta-analysis of genome-wide association studies of asthma in ethnically diverse North American populations.
Asthma is a common disease with a complex risk architecture including both genetic and environmental factors. We performed a meta-analysis of North American genome-wide association studies of asthma in 5,416 individuals with asthma (cases) including individuals of European American, African American or African Caribbean, and Latino ancestry, with replication in an additional 12,649 individuals from the same ethnic groups. We identified five susceptibility loci. Four were at previously reported loci on 17q21, near IL1RL1, TSLP and IL33, but we report for the first time, to our knowledge, that these loci are associated with asthma risk in three ethnic groups. In addition, we identified a new asthma susceptibility locus at PYHIN1, with the association being specific to individuals of African descent (P = 3.9 × 10(-9)). These results suggest that some asthma susceptibility loci are robust to differences in ancestry when sufficiently large samples sizes are investigated, and that ancestry-specific associations also contribute to the complex genetic architecture of asthma

Social asymmetry and physical health in the United States and Japan
OBJECTIVE: Social relationships are increasingly recognized as crucial determinants of health, but cultural variations in the health implications of social disconnection remain understudied. This study examines how nationality, reflecting cultural differences in social norms, moderates the relationship between social asymmetry and physical health in Japanese and U.S. adults. We hypothesized that the association between greater social asymmetry and poorer health would be attenuated in Japan compared to the United States. METHOD: The sample comprised adults aged 23-84 years from Japan (N = 1,027) and the United States (N = 6,650) participating in the Midlife in Japan and Midlife in the United States longitudinal studies. Social asymmetry was quantified as the residual score from regressing loneliness on social isolation, with positive residuals indicating higher loneliness than expected based on isolation levels. Physical health was a latent variable indicated by chronic conditions, symptoms, activities of daily living, and physical activity. Structural equation modeling examined the moderating effect of nationality on the social asymmetry-health link. RESULTS: Across both cultural contexts, greater social asymmetry (i.e., higher loneliness than predicted by isolation) was associated with worse physical health. However, as hypothesized, this relationship was significantly weaker in Japan compared to the United States, highlighting the role of cultural context in shaping the health implications of discrepant social experiences. CONCLUSIONS: Findings contribute to understanding cultural variations in the health consequences of social disconnection and emphasize the need to consider sociocultural factors when examining social determinants of health across diverse populations. Yellow sea mine hunting using the Navy's CASS/GRAB model
The purpose of this work is to determine the necessity of a near real time ocean modeling capability such as the Naval Oceanographic Office's (NAVOCENANO) Modular Ocean Data Assimilation System (MODAS) model in shallow water (such as the Yellow Sea) mine hunting applications using the Navy's Comprehensive Acoustic Simulation System/Gaussian Ray Bundle (CASS/GRAB) Mode. Sound speed profiles inputted into CASS/GRAB were calculated from observational (MOODS) and climatological (GDEM) data sets for different seasons and regions of four different bottom types (sand, gravel, mud and rock). The CASS/GRAB model outputs were compared to the outputs from corresponding MODAS data sets. The results of the comparisons domonstrated in many cases a significant acoustic difference between the alternate profiles. These results demonstrated that there is a need for a predictive modeling capability such as MODAS to address the mine warfare (MIW) needs in the Yellow Sea region. District administrator perspectives of current and ideal approaches to identifying and supporting student social, emotional, and behavioral needs
IntroductionLeadership support has been identified as a key facilitator to successful implementation of school-based initiatives. School leadership contributions to student academic success and school reform have been documented, but less work has focused on the effects of leadership on school mental health initiatives such as social, emotional, and behavioral (SEB) screening. Few studies have investigated administrator knowledge and support for SEB screening or compared their current and ideal approaches to SEB screening, both of which are important to informing directions for implementation supports.MethodsUsing a nationally representative sample of U.S. public school districts, we investigated tensions between school district administrators’ (n = 1,330) current and ideal approaches to SEB screening within the current study. We fit binomial and multinomial logistic regression models to determine predictors of positive, negative, or no tensions based on the administrators’ current approach to SEB screening, administrators’ beliefs about the role of schools in student SEB concerns, and district demographics.ResultsNearly half of administrators (46.7%) reported using an approach to SEB screening that was not their ideal approach. Higher beliefs scores commonly predicted administrator desire for more proactive approaches, whereas district characteristics yielded varied results.DiscussionResults call for targeted next steps, including implementation supports, funding, and knowledge building aligned with administrator and district characteristics

Whole-Genome Sequencing of Individuals from a Founder Population Identifies Candidate Genes for Asthma
Asthma is a complex genetic disease caused by a combination of genetic and environmental risk factors. We sought to test classes of genetic variants largely missed by genome-wide association studies (GWAS), including copy number variants (CNVs) and low-frequency variants, by performing whole-genome sequencing (WGS) on 16 individuals from asthma-enriched and asthma-depleted families. The samples were obtained from an extended 13-generation Hutterite pedigree with reduced genetic heterogeneity due to a small founding gene pool and reduced environmental heterogeneity as a result of a communal lifestyle. We sequenced each individual to an average depth of 13-fold, generated a comprehensive catalog of genetic variants, and tested the most severe mutations for association with asthma. We identified and validated 1960 CNVs, 19 nonsense or splice-site single nucleotide variants (SNVs), and 18 insertions or deletions that were out of frame. As follow-up, we performed targeted sequencing of 16 genes in 837 cases and 540 controls of Puerto Rican ancestry and found that controls carry a significantly higher burden of mutations in IL27RA (2.0% of controls; 0.23% of cases; nominal p = 0.004; Bonferroni p = 0.21). We also genotyped 593 CNVs in 1199 Hutterite individuals. We identified a nominally significant association (p = 0.03; Odds ratio (OR) = 3.13) between a 6 kbp deletion in an intron of NEDD4L and increased risk of asthma. We genotyped this deletion in an additional 4787 non-Hutterite individuals (nominal p = 0.056; OR = 1.69). NEDD4L is expressed in bronchial epithelial cells, and conditional knockout of this gene in the lung in mice leads to severe inflammation and mucus accumulation. Our study represents one of the early instances of applying WGS to complex disease with a large environmental component and demonstrates how WGS can identify risk variants, including CNVs and low-frequency variants, largely untested in GWAS
Environmental and Occupational Factors; Contribution and Perspectives on Difficult to Treat Asthma
There are multiple well-recognized environmental factors that contribute to asthma exacerbation. Exposures to many of them will get unrecognized and most of the time will remain constant without knowing it is the causative agent. For an early identification of exposures and causative agents, a systematic approach needs to be taken in consideration by the encountering physician. Multiple questionnaires had been implementing and discussing organic and inorganic factors as well intrinsic and extrinsic factors. It is well-recognized that environmental exposures can cause worsening of asthma, other allergic conditions and even more severe pulmonary diseases. Asthma is a very prevalent disease with increased incidence nowadays. In the last decade, multiple new medications had been discovered for the treatment of moderate-to-severe persistent asthma, which most of them target the cellular component of the disease such as eosinophils and specific Immunoglobins. In the era of personalized medicine, environmental and occupational factors in asthma are key players that need to be recognized early in this patient population. In this chapter will go over model of effects, mechanism of action of these environmental factors, recognition, course of action and management of this patient population
Factors associated with degree of atopy in Latino children in a nationwide pediatric sample: The Genes-environments and Admixture in Latino Asthmatics (GALA II) study
BackgroundAtopy varies by ethnicity, even within Latino groups. This variation might be due to environmental, sociocultural, or genetic factors.ObjectiveWe sought to examine risk factors for atopy within a nationwide study of US Latino children with and without asthma.MethodsAeroallergen skin test responses were analyzed in 1830 US Latino subjects. Key determinants of atopy included country/region of origin, generation in the United States, acculturation, genetic ancestry, and site to which subjects migrated. Serial multivariate zero-inflated negative binomial regressions stratified by asthma status examined the association of each key determinant variable with the number of positive skin test responses. In addition, the independent effect of each key variable was determined by including all key variables in the final models.ResultsIn baseline analyses African ancestry was associated with 3 times (95% CI, 1.62-5.57) as many positive skin test responses in asthmatic participants and 3.26 times (95% CI, 1.02-10.39) as many positive skin test responses in control participants. Generation and recruitment site were also associated with atopy in crude models. In final models adjusted for key variables, asthmatic patients of Puerto Rican (exp[β] [95% CI], 1.31 [1.02-1.69]) and mixed (exp[β] [95% CI], 1.27 [1.03-1.56]) ethnicity had a greater probability of positive skin test responses compared with Mexican asthmatic patients. Ancestry associations were abrogated by recruitment site but not region of origin.ConclusionsPuerto Rican ethnicity and mixed origin were associated with degree of atopy within US Latino children with asthma. African ancestry was not associated with degree of atopy after adjusting for recruitment site. Local environment variation, represented by site, was associated with degree of sensitization
