    Quantitative Assessment of Desensitizing Agents in Occluding Dentine Tubules using Image Analysis

    Previous in vitro studies assessing the tubule occluding properties of various desensitizing agents in the dentine disc model appear to provide only qualitative data. The aim of this study was to establish a reliable and reproducible system to evaluate the in vitro effectiveness of three desensitizing agents. Six selected fields from SEM negatives (magnification x1000; working distance 10mm) of test and control dentine disc specimens treated with the desensitizing agents (Butler Protect, Colgate FluoriGard (GelKam), Macleans Sensitive) were evaluated. Fields were assessed using a Quantimet 520 Image analysis system (Leica UK) and the data recorded included patent tubule area, mean tubule diameter, mean patent/field area and number of tubules per unit area. Comparison of test and control specimens indicated that differences in the number of tubules, patent area, width of tubules and percentage of patent areas can be assessed quantitatively Furthermore, this methodology also demonstrated differences between the tubule occluding properties of the selected desensitizing agents

    The Strutjet Rocket Based Combined Cycle Engine

    The multi stage chemical rocket has been established over many years as the propulsion System for space transportation vehicles, while, at the same time, there is increasing concern about its continued affordability and rather involved reusability. Two broad approaches to addressing this overall launch cost problem consist in one, the further development of the rocket motor, and two, the use of airbreathing propulsion to the maximum extent possible as a complement to the limited use of a conventional rocket. In both cases, a single-stage-to-orbit (SSTO) vehicle is considered a desirable goal. However, neither the "all-rocket" nor the "all-airbreathing" approach seems realizable and workable in practice without appreciable advances in materials and manufacturing. An affordable system must be reusable with minimal refurbishing on-ground, and large mean time between overhauls, and thus with high margins in design. It has been suggested that one may use different engine cycles, some rocket and others airbreathing, in a combination over a flight trajectory, but this approach does not lead to a converged solution with thrust-to-mass, specific impulse, and other performance and operational characteristics that can be obtained in the different engines. The reason is this type of engine is simply a combination of different engines with no commonality of gas flowpath or components, and therefore tends to have the deficiencies of each of the combined engines. A further development in this approach is a truly combined cycle that incorporates a series of cycles for different modes of propulsion along a flight path with multiple use of a set of components and an essentially single gas flowpath through the engine. This integrated approach is based on realizing the benefits of both a rocket engine and airbreathing engine in various combinations by a systematic functional integration of components in an engine class usually referred to as a rocket-based combined cycle (RBCC) engine. RBCC engines exhibit a high potential for lowering the operating cost of launching payloads into orbit. Two sources of cost reductions can be identified. First, RBCC powered vehicles require only 20% takeoff thrust compared to conventional rockets, thereby lowering the thrust requirements and the replacement cost of the engines. Second, due to the higher structural and thermal margins achievable with RBCC engines coupled with a higher degree of subsystem redundance lower maintenance and operating cost are obtainable

    The prevalence of non-alcoholic fatty liver disease (NAFLD) and its progressive form, non-alcoholic steatohepatitis (NASH), is increasing in parallel with the rising rates of obesity and type 2 diabetes. Approximately one in four adults are diagnosed with liver steatosis globally. NAFLD is associated with insulin resistance, hypertension, obesity, visceral adiposity, and dyslipidaemia. These risk factors are often accompanied by inflammation and oxidative stress, which also play a role in extrahepatic diseases, including conditions related to the central nervous system, such as mild cognitive impairment and Alzheimer’s disease. The number of people living with dementia is approximately 55 million and is estimated to increase to approximately 2 billion people by 2050. Recent studies have found that NAFLD is associated with poorer cognition. The aim of this review was to summarise the findings of hitherto studies that have linked NAFLD with cognition and dementia, as well as to discuss the potential liver–brain pathways

    Can the microbiome drive the suppression of grapevine trunk diseases?

    Grapevine trunk diseases (GTDs), caused by several fungal species, are among the most destructive grapevine diseases in New Zealand and other grape-growing countries. The control of the diseases is problematic, and there is currently no approved fungicide for their eradication. This has necessitated seeking alternative strategies, including a sustainable biological control approach, to manage the diseases. Therefore, this study aimed to identify taxa in the grapevine microbiome that contribute to plant health. In some New Zealand vineyards, observations have revealed vines that remain healthy within a background of trunk diseases. These grapevines were termed ‘disease-escape’ to represent their apparent health under heavy disease pressure. Recent research on the grapevine microbiome has shown that microorganisms from these ‘disease-escape’ plants could contribute to disease suppression. Putative disease escape vines were identified in vineyards in two grape-growing regions in New Zealand: Hawke’s Bay and Canterbury. The vines were selected based on their presence in a diseased area, maturity, and absence of trunk disease symptoms. Trunk core samples were taken from the disease-escape vines and neighbouring symptomatic vines. Subsequently, the samples’ total fungal and bacterial communities were identified and compared using culture-independent DNA metabarcoding and culture-dependent approaches. After analysing the metabarcoding and culturing results, microbial taxa that were differentially more abundant in disease-escape grapevines and the ones that correlated negatively with GTD pathogens were identified. The next stage of the study is to design a synthetic community using members of the taxa of interest from the disease-escape grapevines. This SynCom will be introduced into young grapevines and monitored for their ability to suppress the development and severity of GTDs. The research results will provide information on the roles (if any) that the grapevine trunk’s microbiome plays in suppressing GTDs

    Taking Foreign Policy Personally: Personal Values and Foreign Policy Attitudes

    Previous research has shown that on issues of foreign policy, individuals have “general stances,” “postures,” “dispositions” or “orientations” that inform their beliefs toward more discrete issues in international relations. While these approaches delineate the proximate sources of public opinion in the foreign policy domain, they evade an even more important question: what gives rise to these foreign policy orientations in the first place? Combining an original survey on a nationally representative sample of Americans with Schwartz’s theory of values from political psychology, we show that people take foreign policy personally: the same basic values we know people use to guide choices in their daily lives also travel to the domain of foreign affairs, offering one potential explanation why people who are otherwise uninformed about world politics nonetheless express coherent foreign policy beliefs

    Impact ionization in GaAs: a screened exchange density functional approach

    Results are presented of a fully ab-initio calculation of impact ionization rates in GaAs within the density functional theory framework, using a screened-exchange formalism and the highly precise all-electron full-potential linearized augmented plane wave (FLAPW) method. The calculated impact ionization rates show a marked orientation dependence in {\bf k} space, indicating the strong restrictions imposed by the conservation of energy and momentum. This anisotropy diminishes as the impacting electron energy increases. A Keldysh type fit performed on the energy-dependent rate shows a rather soft edge and a threshold energy greater than the direct band gap. The consistency with available Monte Carlo and empirical pseudopotential calculations shows the reliability of our approach and paves the way to ab-initio calculations of pair production rates in new and more complex materials.Comment: 11 pages, 4 figures, Submitted to Phys.     Potential protein activity modifications of amino acid variants in the human transcriptome

    Background: The occurrence of widespread RNA and DNA sequence differences in the human transcriptome was reported in 2011. Similar findings were described in a second independent publication on personal omics profiling investigating the occurrence of dynamic molecular and related medical phenotypes. The suggestion that the RNA sequence variation was likely to affect disease susceptibility prompted us to investigate with a range of algorithms the amino acid variants reported to be present in the identified peptides to determine if they might be disease-causing. Results: The predictive qualities of the different algorithms were first evaluated by using nonsynonymous single-base nucleotide polymorphism (nsSNP) datasets, using independently established data on amino acid variants in several proteins as well as data obtained by mutational mapping and modelling of binding sites in the human serotonin transporter protein (hSERT). Validation of the used predictive algorithms was at a 75% level. Using the same algorithms, we found that widespread RNA and DNA sequence differences were predicted to impair the function of the peptides in over 57% of cases. Conclusions: Our findings suggest that a proportion of edited RNAs which serve as templates for protein synthesis is likely to modify protein function, possibly as an adaptive survival mechanism in response to environmental modifications

    Things change: Women’s and men’s marital disruption dynamics in Italy during a time of social transformations, 1970-2003

    We study women’s and men’s marital disruption in Italy between 1970 and 2003. By applying an event-history analysis to the 2003 Italian variant of the Generations and Gender Survey we found that the spread of marital disruption started among middle-highly educated women. Then in recent years it appears that less educated women have also been able to dissolve their unhappy unions. Overall we can see the beginning of a reversed educational gradient from positive to negative. In contrast the trend in men’s marital disruption risk appears as a change over time common to all educational groups, although with persisting educational differentials.determinants, educational differences, event history analysis, gender difference, Italy, marital disruption

    Trauma as counter-revolutionary colonisation: narratives from (post)revolutionary Egypt

    We argue that multiple levels of trauma were present in Egypt before, during and after the 2011 revolution. Individual, social and political trauma constitute a triangle of traumatisation which was strategically employed by the Egyptian counter-revolutionary forces – primarily the army and the leadership of the Muslim Brotherhood – to maintain their political and economic power over and above the social, economic and political interests of others. Through the destruction of physical bodies, the fragmentation and polarisation of social relations and the violent closure of the newly emerged political public sphere, these actors actively repressed the potential for creative and revolutionary transformation. To better understand this multi-layered notion of trauma, we turn to Habermas’ ‘colonisation of the lifeworld’ thesis which offers a critical lens through which to examine the wider political and economic structures and context in which trauma occurred as well as its effects on the personal, social and political realms. In doing so, we develop a novel conception of trauma that acknowledges individual, social and political dimensions. We apply this conceptual framing to empirical narratives of trauma in Egypt’s pre- and post-revolutionary phases, thus both developing a non-Western application of Habermas’ framework and revealing ethnographic accounts of the revolution by activists in Cairo
