    Dose distribution in the thyroid gland following radiation therapy of breast cancer-a retrospective study

    <p>Abstract</p> <p>Purpose</p> <p>To relate the development of post-treatment hypothyroidism with the dose distribution within the thyroid gland in breast cancer (BC) patients treated with loco-regional radiotherapy (RT).</p> <p>Methods and materials</p> <p>In two groups of BC patients postoperatively irradiated by computer tomography (CT)-based RT, the individual dose distributions in the thyroid gland were compared with each other; Cases developed post-treatment hypothyroidism after multimodal treatment including 4-field RT technique. Matched patients in Controls remained free for hypothyroidism. Based on each patient's dose volume histogram (DVH) the volume percentages of the thyroid absorbing respectively 20, 30, 40 and 50 Gy were then estimated (V20, V30, V40 and V50) together with the individual mean thyroid dose over the whole gland (MeanTotGy). The mean and median thyroid dose for the included patients was about 30 Gy, subsequently the total volume of the thyroid gland (VolTotGy) and the absolute volumes (cm<sup>3</sup>) receiving respectively < 30 Gy and ≄ 30 Gy were calculated (Vol < 30 and Vol ≄ 30) and analyzed.</p> <p>Results</p> <p>No statistically significant inter-group differences were found between V20, V30, V40 and V50Gy or the median of MeanTotGy. The median VolTotGy in Controls was 2.3 times above VolTotGy in Cases (ρ = 0.003), with large inter-individual variations in both groups. The volume of the thyroid gland receiving < 30 Gy in Controls was almost 2.5 times greater than the comparable figure in Cases.</p> <p>Conclusions</p> <p>We concluded that in patients with small thyroid glands after loco-radiotherapy of BC, the risk of post-treatment hypothyroidism depends on the volume of the thyroid gland.</p

    Epigenetic expansion of VHL-HIF signal output drives multiorgan metastasis in renal cancer.

    Inactivation of the von Hippel-Lindau tumor suppressor gene, VHL, is an archetypical tumor-initiating event in clear cell renal carcinoma (ccRCC) that leads to the activation of hypoxia-inducible transcription factors (HIFs). However, VHL mutation status in ccRCC is not correlated with clinical outcome. Here we show that during ccRCC progression, cancer cells exploit diverse epigenetic alterations to empower a branch of the VHL-HIF pathway for metastasis, and the strength of this activation is associated with poor clinical outcome. By analyzing metastatic subpopulations of VHL-deficient ccRCC cells, we discovered an epigenetically altered VHL-HIF response that is specific to metastatic ccRCC. Focusing on the two most prominent pro-metastatic VHL-HIF target genes, we show that loss of Polycomb repressive complex 2 (PRC2)-dependent histone H3 Lys27 trimethylation (H3K27me3) activates HIF-driven chemokine (C-X-C motif) receptor 4 (CXCR4) expression in support of chemotactic cell invasion, whereas loss of DNA methylation enables HIF-driven cytohesin 1 interacting protein (CYTIP) expression to protect cancer cells from death cytokine signals.     Sarcoptic mange in three alpacas treated successfully with amitraz

    Sarcoptic mange is a serious skin disease in alpacas that can result in high morbidity and even mortality. Three alpacas were presented with sarcoptic mange that had previously failed to respond to repeated topical applications of eprinomectin, and an injection of doramectin. They were moderately to severely pruritic, had extensive lesions of alopecia, erythema, scaling and crusting, and had lost weight. As no drug is currently licensed for the treatment of sarcoptic mange in alpacas in the UK, they were treated with a topical solution of amitraz (50 mL in 10 L) after initial bathing with antibacterial or keratolytic shampoos. The clinical signs completely resolved with no relapse over a 10-month follow-up period. In this small group of alpacas, amitraz was an effective and well-tolerated treatment for sarcoptic mange.Peri Lau, Peter B. Hill, Jan Rybníček and Lynne Stee

    Space invaders: effects of invasive alien Pallas\u2019s squirrel on home range and body mass of native red squirrel

    Alien species can affect native species through several ecological processes such as competition. Here we tested the hypothesis of interspecific competition for space and food resources between the native Eurasian red squirrel and the invasive Pallas\u2019s squirrel introduced in Italy. We used an experimental study design comparing space and habitat use and body condition parameters of red squirrels between areas of co-occurrence with the Pallas\u2019s squirrel and areas without it. There were no differences in mean home range size of red squirrels between red-only areas and red-Pallas. However, when Pallas\u2019s squirrels were removed, the red squirrels increased their home ranges. Moreover, in the area of syntopy, red squirrels had a higher degree of intraspecific home range overlap than in the red-only area. We also found indirect evidence for competition for food with red squirrels having a poorer body condition when co-occurring with the alien species. We analyzed the body mass and size of red squirrels in the two areas and our results showed that red squirrels had a reduced body mass and size when in syntopy, confirming that the interspecific competition does not allow red squirrels to reach the optimum body condition that they would have if the competitor was not present. Moreover, tree-species niche overlap was very high and both species fed primarily on the same tree seeds. Differences in vegetation cover between areas are discussed. This is the first study that confirms the invasiveness of the Pallas\u2019s squirrel also in terms of capability to compete with native species
