    Community Ideas Factory

    The Community Ideas Factory is a community-college partnership exploring social innovations within the charitable sector of the Halton Region. It is a collaborative research project between Sheridan College and the Oakville Community Foundation. The goal of the project is to change the philanthropic granting process in Oakville so that it is more bottom-up, participatory, and evidenced-based. The principle community partner on the project is The Foundation (OCF). A community organization tasked with managing and disbursing donor contributions for philanthropic projects in the Town of Oakville. The Community Ideas Factory is made possible by the College-Community Social Innovation Fund of the Social Sciences and Humanities Research Council of Canada (SSHRC)

    The Use of Imagery to Manipulate Challenge and Threat Appraisal States in Athletes

    The present study investigated whether imagery could manipulate athletes’ appraisal of stress-evoking situations (i.e., challenge or threat) and whether psychological and cardiovascular responses and interpretations varied according to cognitive appraisal of three imagery scripts: challenge, neutral, and threat. Twenty athletes (Mage = 20.85; SD = 1.76; 10 female, 10 male) imaged each script while heart rate, stroke volume, and cardiac output were obtained using Doppler echocardiography. State anxiety and self-confidence were assessed following each script using the Immediate Anxiety Measures Scale. During the imagery, a significant increase in heart rate, stroke volume, and cardiac output occurred for the challenge and threat scripts (p &lt; .05). Although there were no differences in physiological response intensities for both stress-evoking scripts, these responses, along with anxiety symptoms, were interpreted as facilitative during the challenge script and debilitative during the threat script. Results support using imagery to facilitate adaptive stress appraisal.</jats:p

    Do the physical and environment PETTLEP elements predict sport imagery ability?

    The present study aimed to examine whether physical and environment elements of PETTLEP imagery relate to the ability to image five types of sport imagery (i.e. skill, strategy, goal, affect and mastery). Two hundred and ninety participants (152 males, 148 females; Mage = 20.24 years, SD = 4.36) from various sports completed the Sport Imagery Ability Questionnaire (SIAQ), and a set of items designed specifically for the study to assess how frequently participants incorporate physical (e.g. 'I make small movements or gestures during the imagery') and environment (e.g. 'I image in the real training/competition environment') elements of PETTLEP imagery. Structural equation modelling tested a hypothesised model in which imagery priming (i.e. the best fitting physical and environment elements) significantly and positively predicted imagery ability of the different imagery types (skill, β = 0.38; strategy, β = 0.23; goal, β = 0.21; affect, β = 0.25; mastery, β = 0.22). The model was a good fit to the data: χ2 (174) = 263.87, p < .001, CFI = .96, TLI = .95, SRMR = .09, RMSEA = 0.05 (90% CI = 0.03-0.05). Findings displayed that priming imagery with physical and environment elements is associated with better skill, strategy, goal, affect and mastery imagery ability. The findings extend models of imagery use by indicating that how athletes images may influence their imagery ability

    CRISPR/Cas9-induced (CTGâ‹…CAG)n repeat instability in the myotonic dystrophy type 1 locus: implications for therapeutic genome editing

    Myotonic dystrophy type 1 (DM1) is caused by (CTG⋅CAG)n-repeat expansion within the DMPK gene and thought to be mediated by a toxic RNA gain of function. Current attempts to develop therapy for this disease mainly aim at destroying or blocking abnormal properties of mutant DMPK (CUG)n RNA. Here, we explored a DNA-directed strategy and demonstrate that single clustered regularly interspaced short palindromic repeats (CRISPR)/Cas9-cleavage in either its 5′ or 3′ unique flank promotes uncontrollable deletion of large segments from the expanded trinucleotide repeat, rather than formation of short indels usually seen after double-strand break repair. Complete and precise excision of the repeat tract from normal and large expanded DMPK alleles in myoblasts from unaffected individuals, DM1 patients, and a DM1 mouse model could be achieved at high frequency by dual CRISPR/Cas9-cleavage at either side of the (CTG⋅CAG)n sequence. Importantly, removal of the repeat appeared to have no detrimental effects on the expression of genes in the DM1 locus. Moreover, myogenic capacity, nucleocytoplasmic distribution, and abnormal RNP-binding behavior of transcripts from the edited DMPK gene were normalized. Dual sgRNA-guided excision of the (CTG⋅CAG)n tract by CRISPR/Cas9 technology is applicable for developing isogenic cell lines for research and may provide new therapeutic opportunities for patients with DM1

    Disease burden of myotonic dystrophy type 1

    Objective: The objective of this cross-sectional, observational study was to investigate the disease burden of myotonic dystrophy type 1 (DM1), a disabling muscle disorder. Methods: Adults with DM1 were recruited as part of the PhenoDM1 study from Newcastle University (Newcastle upon Tyne, UK). Disease burden data were recorded through the Individualized Neuromuscular Quality of Life (INQoL) questionnaire. Results were examined by sex and clinical variables [e.g. the six-minute walk test (6MWT), the Mini Mental State Examination, and estimated progenitor and modal allele CTG repeat length]. Results: Our sample consisted of 60 patients with DM1 (mean age: 45 years; 45% female). Muscle weakness and fatigue constituted the two most common disease manifestations, reported by 93% and 90% of patients, respectively, followed by muscle locking (73%). Most patients (&gt; 55%) reported feeling anxious/worried, depressed, frustrated, and/or having low confidence/self-esteem, 23% and 33% indicated substantial impairment of daily and leisure activities, respectively, and 47% did not work as a consequence of the disease. Estimated progenitor CTG length corrected by age correlated surprisingly well with INQoL scores. Differences by sex were generally minor. Conclusion: We show that DM1 is associated with a substantial disease burden resulting in impairment across many different domains of patients’ lives, emphasizing the need for a holistic approach to medical management. Our results also show that the INQoL records relevant information about patients with DM1, but that further investigation of the psychometric properties of the scale is needed for meaningful interpretation of instrument scores

    Risk factors for child food contamination in low-income neighbourhoods of Maputo, Mozambique: An exploratory, cross-sectional study.

    In low- and middle-income countries, food may be a critical transmission route for pathogens causing childhood diarrhoea, but basic food hygiene is often overlooked in public health strategies. Characterising child food contamination and its risk factors could help prioritise interventions to reduce foodborne diarrhoeal disease, especially in low-income urban areas where the diarrhoeal disease burden is often high. This cross-sectional study comprised a caregiver questionnaire coupled with food sampling, and food preparation observations, among the study population of an ongoing sanitation trial in Maputo. The aim was to determine the prevalence of child food contamination and associated risk factors. The prevalence of Enterococcus spp., as an indicator of faecal contamination, was estimated in food samples. Risk factor analyses were performed through zero-inflated negative binomial regression on colony counts. A modified hazard analysis and critical control point approach was used to determine critical control points (CCPs) that might effectively reduce risk. Fifty-eight linked caregiver questionnaires and food samples were collected, and 59 food preparation observations were conducted. The prevalence of enterococci in child foods exceeding 10 colony forming units per gram was 53% (95% confidence interval [40%, 67%]). Risk factors for child food contamination were identified, including type of food, food preparation practices, and hygiene behaviours. CCPs included cooking/reheating of food and food storage and handling. This exploratory study highlights the need for more research into diarrhoeagenic pathogens and foodborne risks for children living in these challenging urban environments
