    Correlation between Enzymatic and Non-Enzymatic Antioxidants in Several Edible Mushrooms Species

    Characterization of several wild growing and cultivated mushrooms from geographical area of Dambovita County, Romania, in terms of enzymatic and non-enzymatic antioxidants, through a chemometrics approach, was the aim of this study. Related to the authors’ previous studies, the novelty of this paper consists in deepening research toward the complete characterization of the regional mushroom species through emphasizing their potential as food resources. In the context in which species showed their content in biological active compounds, future practical applications in the area of functional food will be developed by integrating the data concerning their lack of the toxicity and nutritional value too. Lack of data focused on the characterization of mushroom species investigated in the paper supports the significance of this research. The statistical analysis of data highlights the relationship between compounds showing antioxidant activity of autochthonous mushrooms (both cap and stipe)

    EVALUATION OF HEAVY METALS CONTENT IN EDIBLE MUSHROOMS BY MICROWAVE DIGESTION AND FLAME ATOMIC ABSORPTION SPECTROMETRY

    The aim of this work was to determine the heavy metal (Cd, Cr, Ni, Pb, Mn, Zn, Fe and Cu) content of the fruiting bodies (cap and stipe) of four species (Amanita caesarea, Pleurotus ostreatus, Fistulina hepatica and Armillariella mellea) and their substrate, collected from forest sites in Dâmboviţa County, Romania. The elements were determined by Flame Atomic Absorption Spectrometry (FAAS) after microwave assisted digestion. From the same collecting point were taken n = 5 samples of young and mature fruiting bodies of mushrooms and their substrate. The high concentrations of lead, chrome and cadmium (Pb: 0.25 – 1.89 mg.kg-1, Cr: 0.36 – 1.94 mg.kg-1, Cd: 0.23 – 1.13 mg.kg-1) for all collected wild edible mushrooms, were determined. These data were compared with maximum level for certain contaminants in foodstuffs established by the commission of the European Committees (EC No 466/2001). A quantitative evaluation of the relationship of element uptake by mushrooms from substrate was made by calculating the accumulation coefficient (Ka). The moderately acid pH value of soil influenced the accumulation of Zn and Cd inside of the studied species. The variation of heavy metals content between edible mushrooms species is dependent upon the ability of the species to extract elements from the substrate and on the selective uptake and deposition of metals in tissue

Though much less accurate than the other methods recommended in monographs, the thinlayer chromatography -TLC -finds its well deserved place among the analysis methods, when the relatively costly equipments involved by other methods are not available

    Grape by-Products: Potential Sources of Phenolic Compounds for Novel Functional Foods

    Grapes occupy an important position in the human diet, providing both macronutrients and micronutrients indispensable for growth and development. Grapes contain enzymatic and non-enzymatic (nutritional) antioxidants, such as ascorbic acid and bioflavones. High amounts of these valuable compounds are removed through processing, when the skin and the seeds are separated from the pulp. During the vinification process, a large amount of grape pomace is generated that constitutes an important source of value-added products such as phenolic compounds, mainly flavonoids, phenolic acids, and stilbenes. Valorization of wine industry by-products like grape pomace is a suitable strategy for recovering bioactive compounds (mainly polyphenols) and reducing the environmental impact of this industrial waste. The circular bioeconomy refers to maintaining the value of the biological resources in economy for as long as possible, minimizing the waste production. Recovery and utilization of pomace from grape process favors closing the loop to ensure the abovementioned circularity. The experimental screening performed was designed to assess several indices of the polyphenolic composition of several grape byproducts (pomace, steams, and skin and seed mixture), such as total polyphenolic content, total flavonoid content, and their antioxidant activity, for two white grape Romanian varieties (i.e., Fetească Albă and Tămâioasă Românească)

    Romanian Organic and Conventional Red Grapes Vineyards as Potential Sources of High Value-Added Products, in a Circular Economy Approach

    The use of natural ingredients with active functions has been intensively studied in the last years, as a consequence to consumer preferences for organic products. Application of circular economy principles determined a significant research activity in the viticulture field. The use or re-use of vines parts for so-called nutraceuticals or other consumer-goods applications, are basically centered on their phytochemical and microbiological characterization. Eurostat updates ranks Romania fifth among the EU member states, with a total area under vines of 183,717 hectares. Characterization of four Vitis vinifera L. varieties, out of which one pure Romanian variety (Feteasca Neagra), cultivated in organic and conventional vineyards, together with pedoclimatic conditions have been provided. Data on phytochemical parameters and antimicrobial activity of extracts obtained from different anatomic parts of grapes were included. Analytical protocols and techniques applied were presented, together with data and results interpretation. Several chemometric algorithms have been used as complementary tools for interpretation of the instrumental analytical data

    Infected pancreatic necrosis: outcomes and clinical predictors of mortality. A post hoc analysis of the MANCTRA-1 international study

    : The identification of high-risk patients in the early stages of infected pancreatic necrosis (IPN) is critical, because it could help the clinicians to adopt more effective management strategies. We conducted a post hoc analysis of the MANCTRA-1 international study to assess the association between clinical risk factors and mortality among adult patients with IPN. Univariable and multivariable logistic regression models were used to identify prognostic factors of mortality. We identified 247 consecutive patients with IPN hospitalised between January 2019 and December 2020. History of uncontrolled arterial hypertension (p = 0.032; 95% CI 1.135-15.882; aOR 4.245), qSOFA (p = 0.005; 95% CI 1.359-5.879; aOR 2.828), renal failure (p = 0.022; 95% CI 1.138-5.442; aOR 2.489), and haemodynamic failure (p = 0.018; 95% CI 1.184-5.978; aOR 2.661), were identified as independent predictors of mortality in IPN patients. Cholangitis (p = 0.003; 95% CI 1.598-9.930; aOR 3.983), abdominal compartment syndrome (p = 0.032; 95% CI 1.090-6.967; aOR 2.735), and gastrointestinal/intra-abdominal bleeding (p = 0.009; 95% CI 1.286-5.712; aOR 2.710) were independently associated with the risk of mortality. Upfront open surgical necrosectomy was strongly associated with the risk of mortality (p < 0.001; 95% CI 1.912-7.442; aOR 3.772), whereas endoscopic drainage of pancreatic necrosis (p = 0.018; 95% CI 0.138-0.834; aOR 0.339) and enteral nutrition (p = 0.003; 95% CI 0.143-0.716; aOR 0.320) were found as protective factors. Organ failure, acute cholangitis, and upfront open surgical necrosectomy were the most significant predictors of mortality. Our study confirmed that, even in a subgroup of particularly ill patients such as those with IPN, upfront open surgery should be avoided as much as possible.     Food plants provide a regulated source of delivery of functional compounds, plant secondary metabolites production being also tissue specific. In grape berries, the phenolic compounds, flavonoids and non-flavonoids, are distributed in the different parts of the fruit. The aim of this study was to investigate the applicability of FTIR and Raman screening spectroscopic techniques combined with multivariate statistical tools to find patterns in red grape berry parts (skin, seeds and pulp) according to grape variety and vineyard type (organic and conventional). Spectral data were acquired and processed using the same pattern for each different berry part (skin, seeds and pulp). Multivariate analysis has allowed a separation between extracts obtained from organic and conventional vineyards for each grape variety for all grape berry parts. The innovative approach presented in this work is low-cost and feasible, being expected to have applications in studies referring to the authenticity and traceability of foods. The findings of this study are useful as well in solving a great challenge that producers are confronting, namely the consumers’ distrust of the organic origin of food products. Further analyses of the chemical composition of red grapes may enhance the capability of the method of using both vibrational spectroscopy and chemometrics for discriminating the hydroalcoholic extracts according to grape varieties

    Phytochemical profiles, antioxidant and antibacterial activities of grape (Vitis vinifera L.) seeds and skin from organic and conventional vineyards

    The therapeutic benefits of extracts obtained from different red grape fractions were thoroughly studied, however, data regarding the comparison of phytochemical extracts prepared from the same varieties coming from organic versus conventional management systems are rather lacking. The present study aimed at comparing some of the phytochemical characteristics and antimicrobial activity of hydroalcoholic (50% v/v) extracts obtained from four varieties of red grapes cultivated respectively in organic and conventional vineyards. Total flavonoid content, total phenolic compounds, and antioxidant activity were determined by molecular absorption spectroscopy. Antimicrobial activity of the studied extracts was evaluated against common bacterial strains isolated from different habitats according to specific lab procedures. The analyses were performed in solid broths by applying the disk diffusion method, which allowed for the simultaneous determination of the spectrum of the sensitivity of the tested bacteria as well as the values of the minimum inhibition concentration (MIC).
