    CD1d expression demarcates CDX4+ hemogenic mesoderm with definitive hematopoietic potential

    To achieve efficient, reproducible differentiation of human pluripotent stem cells (hPSCs) towards specific hematopoietic cell-types, a comprehensive understanding of the necessary cell signaling and developmental trajectories involved is required. Previous studies have identified the mesodermal progenitors of extra-embryonic-like and intra-embryonic-like hemogenic endothelium (HE), via stage-specific WNT and ACTIVIN/NODAL, with GYPA/GYPB (CD235a/b) expression serving as a positive selection marker for mesoderm harboring exclusively extra-embryonic-like hemogenic potential. However, a positive mesodermal cell-surface marker with exclusively intra-embryonic-like hemogenic potential has not been identified. Recently, we reported that early mesodermal expression of CDX4 critically regulates definitive HE specification, suggesting that CDX4 may act in a cell-autonomous manner during hematopoietic development.     SAF-A mutants disrupt chromatin structure through dominant negative effects on RNAs associated with chromatin

    Here we provide a brief review of relevant background before presenting results of our investigation into the interplay between scaffold attachment factor A (SAF-A), chromatin-associated RNAs, and DNA condensation. SAF-A, also termed heterogenous nuclear protein U (hnRNP U), is a ubiquitous nuclear scaffold protein that was implicated in XIST RNA localization to the inactive X-chromosome (Xi) but also reported to maintain open DNA packaging in euchromatin. Here we use several means to perturb SAF-A and examine potential impacts on the broad association of RNAs on euchromatin, and on chromatin compaction. SAF-A has an N-terminal DNA binding domain and C-terminal RNA binding domain, and a prominent model has been that the protein provides a single-molecule bridge between XIST RNA and chromatin. Here analysis of the impact of SAF-A on broad RNA-chromatin interactions indicate greater biological complexity. We focus on SAF-A\u27s role with repeat-rich C0T-1 hnRNA (repeat-rich heterogeneous nuclear RNA), shown recently to comprise mostly intronic sequences of pre-mRNAs and diverse long non-coding RNAs (lncRNAs). Our results show that SAF-A mutants cause dramatic changes to cytological chromatin condensation through dominant negative effects on C0T-1 RNA\u27s association with euchromatin, and likely other nuclear scaffold factors. In contrast, depletion of SAF-A by RNA interference (RNAi) had no discernible impact on C0T-1 RNA, nor did it cause similarly marked chromatin changes as did three different SAF-A mutations. Overall results support the concept that repeat-rich, chromatin-associated RNAs interact with multiple RNA binding proteins (RBPs) in a complex dynamic meshwork that is integral to larger-scale chromatin architecture and collectively influences cytological-scale DNA condensation

    Tunable control of the hydrophilicity and wettability of conjugated polymers by a postpolymerization modification approach.

    A facile method to prepare hydrophilic polymers by a postpolymerization nucleophillic aromatic substitution reaction of fluoride on an emissive conjugated polymer (CP) backbone is reported. Quantitative functionalization by a series of monofunctionalized ethylene glycol oligomers, from dimer to hexamer, as well as with high molecular weight polyethylene glycol is demonstrated. The length of the ethylene glycol sidechains is shown to have a direct impact on the surface wettability of the polymer, as well as its solubility in polar solvents. However, the energetics and band gap of the CPs remain essentially constant. This method therefore allows an easy way to modulate the wettability and solubility of CP materials for a diverse series of applications

    Psychological health of Australian veterans of the 1991 Gulf War: an assessment using the SF-12, GHQ-12 and PCL-S

    Background. Elevated rates of psychological morbidity and symptomatology have been widely reported in 1991 Gulf War veterans. The present study used brief self-report instruments to compare the psychological health of Australian Gulf War veterans with that of a randomly sampled military comparison group. Method. The 12-item Short Form Health Survey (SF-12), 12-item General Health Questionnaire (GHQ-12), Posttraumatic Stress Disorder Checklist – Specific (PCL-S) and Military Service Experience (MSE) questionnaire were administered to 1424 male Australian Gulf War veterans and 1548 male Australian Defence Force members who were operational at the time of the Gulf War conflict, but were not deployed there. Results. The Gulf War veterans exhibited poorer psychological health, as measured by the above three instruments, than the comparison group members. For Gulf War veterans, the number of stressful experiences, as measured by the MSE questionnaire, was correlated with scores on the three instruments. SF-12 mental health component summary scores and PCL-S caseness, but not GHQ-12 caseness, differed significantly between Gulf War veterans and comparison group members who had been on at least one active deployment. Conclusions. More than a decade after the 1991 Gulf War, Australian Gulf War veterans are exhibiting higher levels of current (past month) psychological ill-health, as measured using the GHQ-12 and PCL-S, as well as lower mental health status, as measured by the SF-12, than the comparison group.     Universal correlations of trapped one-dimensional impenetrable bosons

    We calculate the asymptotic behaviour of the one body density matrix of one-dimensional impenetrable bosons in finite size geometries. Our approach is based on a modification of the Replica Method from the theory of disordered systems. We obtain explicit expressions for oscillating terms, similar to fermionic Friedel oscillations.     Diagnostic Utility of the Impact of Event Scale-Revised in Two Samples of Survivors of War

    The study aimed at examining the diagnostic utility of the Impact of Event Scale-Revised (IES-R) as a screening tool for post-traumatic stress disorder (PTSD) in survivors of war. The IES-R was completed by two independent samples that had survived the war in the Balkans: a sample of randomly selected people who had stayed in the area of former conflict (n = 3,313) and a sample of refugees to Western European countries (n = 854). PTSD was diagnosed using the MINI International Neuropsychiatric Interview. Prevalence of PTSD was 20.1% in the Balkan sample and 33.1% in the refugee sample. Results revealed that when considering a minimum value of specificity of 0.80, the optimally sensitive cut-off score for screening for PTSD in the Balkan sample was 34. In both the Balkan sample and the refugee sample, this cut-off score provided good values on sensitivity (0.86 and 0.89, respectively) and overall efficiency (0.81 and 0.79, respectively). Further, the kappa coefficients for sensitivity for the cut-off of 34 were 0.80 in both samples. Findings of this study support the clinical utility of the IES-R as a screening tool for PTSD in large-scale research studies and intervention studies if structured diagnostic interviews are regarded as too labor-intensive and too costly

    Biochar Reduced Nitrous Oxide and Carbon Dioxide Emissions from Soil with Different Water and Temperature Cycles

    Interactions among biochar, respiration, nitrification, and soils can result in biochar increasing, decreasing, or not impacting greenhouse gas (GHG) emissions. This experiment determined the impact of water-filled porosity (WFP) and corn (Zea mays L.) stover biochar on CO2 and N2O emissions in May (spring) and August (summer). The May experiment contained two N rates [0 and 224 kg Ca(NO3)2–N ha–1], whereas the August had three N rates [0, 224 kg Ca(NO3)2–N ha–1, and 224 kg (NH4)2SO4–N ha–1]. The average temperatures in the May and Augusts 2014 experiments were 14 and 24°C, respectively. Biochar reduced CO2–C emissions in the high WFP Ca(NO3)2 treatment in the May and August experiments 15.4 and 16.3 kg ha–1, respectively. Associated with the CO2–C decrease was a 15.7% reduction in the soil solution dissolved organic C. In addition, N2O–N and CO2–C emissions were not correlated in the May Ca(NO3)2 ha–1 treatment, whereas in the August experiment, N2O–N and CO2–C emissions were correlated (r2 = 0.98, P \u3c 0.01). In August, biochar increased the apparent nitrification from 16 to 25 kg NH4–N (ha × d)–1 in the low WFP (NH4)2SO4treatment, and it did not influence the nitrification rate in the high WFP (NH4)2SO4 treatment. In general, N2O–N emissions increased with WFP and N rate and were reduced 21.7% by biochar. The findings suggest that multiple mechanisms contributed to N2O emissions and seasonal differences in soil temperature could result in biochar having a mixed impact on GHG emissions

    Partisan Asymmetries in Online Political Activity

    We examine partisan differences in the behavior, communication patterns and social interactions of more than 18,000 politically-active Twitter users to produce evidence that points to changing levels of partisan engagement with the American online political landscape. Analysis of a network defined by the communication activity of these users in proximity to the 2010 midterm congressional elections reveals a highly segregated, well clustered partisan community structure. Using cluster membership as a high-fidelity (87% accuracy) proxy for political affiliation, we characterize a wide range of differences in the behavior, communication and social connectivity of left- and right-leaning Twitter users. We find that in contrast to the online political dynamics of the 2008 campaign, right-leaning Twitter users exhibit greater levels of political activity, a more tightly interconnected social structure, and a communication network topology that facilitates the rapid and broad dissemination of political information.Comment: 17 pages, 10 figures, 6 table

    Technical report: Modeling nitrate leaching risk from specialty crop fields during on-farm managed floodwater recharge in the Kings Groundwater Basin and the potential for its management

    This project has focused on better understanding the potential impact of On-Farm Flood Capture and Recharge (OFFCR) on groundwater quality pertaining to salts and nitrate and on assessing potential management opportunities. To achieve these goals, we used a combination of field and modeling studies. For the field study, soil cores were taken to a depth of 30 feet in replicate across fields with three different specialty crops identified as important to the San Joaquin Valley (tomatoes, almonds, vineyards) and with potential suitability for OFFCR. A prime goal of the field study was to provide data for parameterizing two models developed to assess nitrate, salt and water transport through the vadose zone, prior to percolating into the groundwater aquifer. However, the field study also resulted in key findings that show its value as a stand-alone study: 1) Nitrate concentrations are highest in the upper vadose zone and affected by texture. Those effects are not evident in the deeper vadose zone. 2) Vadose zone nitrate concentrations are affected by the crop grown. These results suggest an opportunity for lower legacy mass transport for grapes and higher legacy mass transport for both tomatoes and almonds. 3) Variability in individual farmers’ past and present fertilizer and water management practices contributes to different legacy salt and nitrate loads in the vadose zone. Data from the field study and other related and concurrent OFFCR field efforts were used during model development. The overall modeling approach was designed to model nitrate and salt transport for lands under OFFCR operation for different crop types, vadose zone characteristics and groundwater characteristics. The defined goals of this design and modeling approach were to: 1) model nitrate and salt movement through the vadose zone and into groundwater; 2) test the model against scenarios that consider different recharge rates, cultural practices, soil types, and depths to groundwater, assessing the timing and magnitude of loading through the vadose zone and the effects on underlying groundwater; and 3) recommend management practices to mitigate potential groundwater impacts. To achieve these goals, two models were integrated to simulate nitrate and salt transport through the vadose zone to groundwater under different scenarios: a 1D Hydrus model and an analytical groundwater model (AGM)

    Supersymmetric QCD: Exact Results and Strong Coupling

    We revisit two longstanding puzzles in supersymmetric gauge theories. The first concerns the question of the holomorphy of the coupling, and related to this the possible definition of an exact (NSVZ) beta function. The second concerns instantons in pure gluodynamics, which appear to give sensible, exact results for certain correlation functions, which nonetheless differ from those obtained using systematic weak coupling expansions. For the first question, we extend an earlier proposal of Arkani-Hamed and Murayama, showing that if their regulated action is written suitably, the holomorphy of the couplings is manifest, and it is easy to determine the renormalization scheme for which the NSVZ formula holds. This scheme, however, is seen to be one of an infinite class of schemes, each leading to an exact beta function; the NSVZ scheme, while simple, is not selected by any compelling physical consideration. For the second question, we explain why the instanton computation in the pure supersymmetric gauge theory is not reliable, even at short distances. The semiclassical expansion about the instanton is purely formal; if infrared divergences appear, they spoil arguments based on holomorphy. We demonstrate that infrared divergences do not occur in the perturbation expansion about the instanton, but explain that there is no reason to think this captures all contributions from the sector with unit topological charge. That one expects additional contributions is illustrated by dilute gas corrections. These are infrared divergent, and so difficult to define, but if non-zero give order one, holomorphic, corrections to the leading result. Exploiting an earlier analysis of Davies et al, we demonstrate that in the theory compactified on a circle of radius beta, due to infrared effects, finite contributions indeed arise which are not visible in the formal limit that beta goes to infinity.Comment: 28 pages, two references added, one typo correcte
