    An analysis of atmospheric entry trajectories for manned and unmanned missions to the planet Venus

    A unified atmospheric density model is presented for the planet Venus, and this model is compatible with Russian and American data obtained during the 1967 Venus launch opportunity. Trajectory characteristics involved in atmospheric entry are discussed with reference to roll-control modulation and entry corridor. Subsequent to a trajectory-oriented discussion of potential Venus mission characteristics, a parametric analysis of manned and unmanned vehicle entry trajectories into the Venusian atmosphere is presented. A sensitivity analysis with reference to atmospheric density deviations is included to show the dependence of corridor depth on the atmospheric density profile

    Lin28A induces energetic switching to glycolytic metabolism in human embryonic kidney cells

    Background: Loss of a cell’s capacity to generate sufficient energy for cellular functions is a key hallmark of the ageing process and ultimately leads to a variety of important age-related pathologies such as cancer, Parkinson’s disease and atherosclerosis. Regenerative medicine has sought to reverse these pathologies by reprogramming somatic cells to a more juvenile energetic state using a variety of stem cell factors. One of these factors, Lin28, is considered a candidate for modification in the reprogramming of cellular energetics to ameliorate the ageing process while retaining cell phenotype. Results: Over-expression of Lin28A resulted in key changes to cellular metabolism not observed in wild-type controls. Extracellular pH flux analysis indicated that Lin28A over expression significantly increased the rate of glycolysis, whilst high resolution oxygen respirometry demonstrated a reduced oxygen consumption. Western blot and real-time PCR analysis identified Hexokinase II as one of the key modulators of glycolysis in these cells which was further confirmed by increased glucose transport. A metabolic switching effect was further emphasised by Western blot analysis where the oxygen consuming mitochondrial complex IV was significantly reduced after Lin28A over expression. Conclusions: Results from this study confirm that Lin28A expression promotes metabolic switching to a phenotype that relies predominantly on glycolysis as an energy source, while compromising oxidative phosphorylation. Mechanisms to augment regulated Lin28A in age related pathologies that are characterised by mitochondria dysfunction or in differentiated and aged post-mitotic cells is the future goal of this work

    Getting on the E List: E-Mail Use in a Community of Service Provider

    This case examines how a community of organizations providing service to people experiencing homelessness made use of an electronic mail list. Current economic conditions have encouraged organizations in various sectors—including nonprofits—that might normally compete for scarce resources to collaborate with one another to increase their chances of survival. One set of tools likely to be of value in such relationships includes various online discussion technologies. An examination of this community’s email list use over a three-year period suggests a somewhat complex picture regarding technology use. More specifically, some issues both constrain and enable use. Additionally, seemingly basic and minimal uses of the list provided not only the greatest functionality for the users, but also led to several unanticipated consequences for those involved

    An Energy-Based Body Temperature Threshold between Torpor and Normothermia for Small Mammals

    Field studies of use of torpor by heterothermic endotherms suffer from the lack of a standardized threshold differentiating torpid body temperatures (T_b) from normothermic T_b's. This threshold can be more readily observed if metabolic rate (MR) is measured in the laboratory. I digitized figures from the literature that depicted simultaneous traces of MR and T_b from 32 respirometry runs for 14 mammal species. For each graph, I quantified the T_b measured when MR first began to drop at the onset of torpor (T_b-onset). I used a general linear model to quantify the effect of ambient temperature (T_a) and body mass (BM) on T_b-onset. For species lighter than 70 g, the model was highly significant and was described by the equation T_b-onset = (0.055 ± 0.014)BM + (0.071 ± 0.031)T_a + (31.823 ± 0.740). To be conservative, I recommend use of these model parameters minus 1 standard error, which modifies the equation to T_b-onset - 1 SE = (0.041)BM + (0.040)T_a + 31.083. This approach provides a standardized threshold for differentiating torpor from normothermia that is based on use of energy, the actual currency of interest for studies of torpor in the wild. Few laboratory studies have presented the time-course data required to quantify T_b-onset, so more data are needed to validate this relationship.https://www.journals.uchicago.edu/doi/10.1086/52108

    The ASCA Spectrum of the Vela Pulsar Jet

    ROSAT observations of the Vela pulsar and its surroundings revealed a collimated X-ray feature almost 45' in length (Markwardt & Ogelman 1995), interpreted as the signature ``cocoon'' of a one-sided jet from the Vela pulsar. We report on a new ASCA observation of the Vela pulsar jet at its head, the point where the jet is believed to interact with the supernova remnant. The head is clearly detected, and its X-ray spectrum is remarkably similar to the surrounding supernova remnant spectrum, extending to X-ray energies of at least 7 keV. A ROSAT+ASCA spectrum can be fit by two-component emission models but not standard one-component models. The lower energy component is thermal and has a temperature of 0.29+/-0.03 keV (1 sigma); the higher energy component can be fit by either a thermal component of temperature ~4 keV or a power law with photon index ~2.0. Compared to the ROSAT-only results, the mechanical properties of the jet and its cocoon do not change much. If the observed spectrum is that of a hot jet cocoon, then the speed of the jet is at least 800 km s^-1, depending on the angle of inclination. The mechanical power driving the jet is >10^36 erg s^-1, and the mass flow rate at the head is > 10^-6 M_sun yr^-1. We conclude that the jet must be entraining material all along its length in order to generate such a large mass flow rate.     Groundtruthing next-gen sequencing for microbial ecology-biases and errors in community structure estimates from PCR amplicon pyrosequencing

    Analysis of microbial communities by high-throughput pyrosequencing of SSU rRNA gene PCR amplicons has transformed microbial ecology research and led to the observation that many communities contain a diverse assortment of rare taxa-a phenomenon termed the Rare Biosphere. Multiple studies have investigated the effect of pyrosequencing read quality on operational taxonomic unit (OTU) richness for contrived communities, yet there is limited information on the fidelity of community structure estimates obtained through this approach. Given that PCR biases are widely recognized, and further unknown biases may arise from the sequencing process itself, a priori assumptions about the neutrality of the data generation process are at best unvalidated. Furthermore, post-sequencing quality control algorithms have not been explicitly evaluated for the accuracy of recovered representative sequences and its impact on downstream analyses, reducing useful discussion on pyrosequencing reads to their diversity and abundances. Here we report on community structures and sequences recovered for in vitro-simulated communities consisting of twenty 16S rRNA gene clones tiered at known proportions. PCR amplicon libraries of the V3-V4 and V6 hypervariable regions from the in vitro-simulated communities were sequenced using the Roche 454 GS FLX Titanium platform. Commonly used quality control protocols resulted in the formation of OTUs with >1% abundance composed entirely of erroneous sequences, while over-aggressive clustering approaches obfuscated real, expected OTUs. The pyrosequencing process itself did not appear to impose significant biases on overall community structure estimates, although the detection limit for rare taxa may be affected by PCR amplicon size and quality control approach employed. Meanwhile, PCR biases associated with the initial amplicon generation may impose greater distortions in the observed community structure
