    Rate and frequency of demands on children with autism

    Applied Behavior Analytic (ABA) intervention seeks to improve deficits in children with autism by providing multiple planned opportunities for learners to develop and practice skills that are useful to them, and are effective alternatives to less acceptable behaviors. Throughout a given day, teachers give instruction to children. While the rate and frequency of these instructions occur in high numbers, there is little literature on just how often they occur. This project sought to find out the frequency of demands in one-hour increments for ten children in a behavior analytic school setting, and to test for differences between male and female students. Demands were tallied for any directive that was presented to the individual in which evokes a response. Results implicated that students averaged 173.47 demands per hour. These results indicate that a high number of demands are needed to keep the students engaged and maximize their learning opportunities

    Predicting individual differences in decision-making process from signature movement styles: an illustrative study of leaders

    There has been a surge of interest in examining the utility of methods for capturing individual differences in decision-making style. We illustrate the potential offered by Movement Pattern Analysis (MPA), an observational methodology that has been used in business and by the US Department of Defense to record body movements that provide predictive insight into individual differences in decision-making motivations and actions. Twelve military officers participated in an intensive 2-h interview that permitted detailed and fine-grained observation and coding of signature movements by trained practitioners using MPA. Three months later, these subjects completed four hypothetical decision-making tasks in which the amount of information sought out before coming to a decision, as well as the time spent on the tasks, were under the partial control of the subject. A composite MPA indicator of how a person allocates decision-making actions and motivations to balance both Assertion (exertion of tangible movement effort on the environment to make something occur) and Perspective (through movements that support shaping in the body to perceive and create a suitable viewpoint for action) was highly correlated with the total number of information draws and total response time—individuals high on Assertion reached for less information and had faster response times than those high on Perspective. Discussion focuses on the utility of using movement-based observational measures to capture individual differences in decision-making style and the implications for application in applied settings geared toward investigations of experienced leaders and world statesmen where individuality rules the day

    Explain To Me: Salience-Based Explainability for Synthetic Face Detection Models

    The performance of convolutional neural networks has continued to improve over the last decade. At the same time, as model complexity grows, it becomes increasingly more difficult to explain model decisions. Such explanations may be of critical importance for reliable operation of human-machine pairing setups, or for model selection when the "best" model among many equally-accurate models must be established. Saliency maps represent one popular way of explaining model decisions by highlighting image regions models deem important when making a prediction. However, examining salience maps at scale is not practical. In this paper, we propose five novel methods of leveraging model salience to explain a model behavior at scale. These methods ask: (a) what is the average entropy for a model's salience maps, (b) how does model salience change when fed out-of-set samples, (c) how closely does model salience follow geometrical transformations, (d) what is the stability of model salience across independent training runs, and (e) how does model salience react to salience-guided image degradations. To assess the proposed measures on a concrete and topical problem, we conducted a series of experiments for the task of synthetic face detection with two types of models: those trained traditionally with cross-entropy loss, and those guided by human salience when training to increase model generalizability. These two types of models are characterized by different, interpretable properties of their salience maps, which allows for the evaluation of the correctness of the proposed measures.     Political parties and democratic consolidation in post-communist societies

    Political parties have a central role to play in democratic consolidation, yet we know comparatively little about how effectively they represent social cleavages in newly emerging democracies. Using the Lipset–Rokkan framework, this article examines the role of parties in articulating social cleavages in 14 established and 6 emerging democracies using the Comparative Study of Electoral Systems datasets. The results show that the social cleavages in the emerging democracies are similar to those of the established democracies, with religion and class predominating. Parties appear to be less effective in representing social cleavages in the emerging than in the established democracies

    Stromal cell inhibition of anti-CD20 antibody mediated killing of B-cell malignancies

    Introduction: The glycoengineered type II anti-CD20 monoclonal antibody obinutuzumab has been licensed for treatment in follicular non-Hodgkin lymphoma and B-CLL following clinical trials demonstrating superior outcomes to standard of care treatment. However, ultimately many patients still relapse, highlighting the need to understand the mechanisms behind treatment failure to improve patient care. Resistance to chemotherapy is often caused by the ability of malignant B-cells to migrate to the bone marrow and home into the stromal layer. Therefore, this study aimed to investigate whether stromal cells were also able to inhibit type II anti-CD20 antibody mechanisms of action, contributing to resistance to therapy.Methods: A stromal-tumor co-culture was established in vitro between Raji or Daudi B-cell tumor cells and M210B4 stromal cells in 24 well plates.Results: Contact with stromal cells was able to protect tumor cells from obinutuzumab mediated programmed cell death (PCD), antibody dependent cellular phagocytosis and antibody dependent cellular cytotoxicity. Furthermore, such protection required direct contact between stroma and tumor cells. Stromal cells appeared to interfere with obinutuzumab mediated B-cell homotypic adhesion through inhibiting and reversing actin remodelling, potentially as a result of stromal-tumor cell contact leading to downregulation of CD20 on the surface of tumor cells. Further evidence for the potential role of CD20 downregulation comes through the reduction in surface CD20 expression and inhibition of obinutuzumab mediated PCD when tumor cells are treated with Ibrutinib in the presence of stromal cells. The proteomic analysis of tumor cells after contact with stromal cells led to the identification of a number of altered pathways including those involved in cell adhesion and the actin cytoskeleton and remodeling.Discussion: This work demonstrates that contact between tumor cells and stromal cells leads to inhibition of Obinutuzumab effector functions and has important implications for future therapies to improve outcomes to anti-CD20 antibodies. A deeper understanding of how anti-CD20 antibodies interact with stromal cells could prove a useful tool to define better strategies to target the micro-environment and ultimately improve patient outcomes in B-cell malignancies

    Forward genetic screen of homeostatic antibody levels in the Collaborative Cross identifies MBD1 as a novel regulator of B cell homeostasis

    Variation in immune homeostasis, the state in which the immune system is maintained in the absence of stimulation, is highly variable across populations. This variation is attributed to both genetic and environmental factors. However, the identity and function of specific regulators have been difficult to identify in humans. We evaluated homeostatic antibody levels in the serum of the Collaborative Cross (CC) mouse genetic reference population. We found heritable variation in all antibody isotypes and subtypes measured. We identified 4 quantitative trait loci (QTL) associated with 3 IgG subtypes: IgG1, IgG2b, and IgG2c. While 3 of these QTL map to genome regions of known immunological significance (major histocompatibility and immunoglobulin heavy chain locus), Qih1 (associated with variation in IgG1) mapped to a novel locus on Chromosome 18. We further associated this locus with B cell proportions in the spleen and identify Methyl-CpG binding domain protein 1 under this locus as a novel regulator of homeostatic IgG1 levels in the serum and marginal zone B cells (MZB) in the spleen, consistent with a role in MZB differentiation to antibody secreting cells
