    Radio frequency plasma processing effects on the emission characteristics of a MeV electron beam cathode

    Experiments have proven that surface contaminants on the cathode of an electron beam diode influence electron emission current and impedance collapse. This letter reports on an investigation to reduce parasitic cathode current loss and to increase high voltage hold off capabilities by reactive sputter cleaning of contaminants. Experiments have characterized effective radio frequency (rf) plasma processing protocols for high voltage anode–cathode (A–K) gaps using a two-stage argon/oxygen and argon rf plasma discharge. Time-resolved optical emission spectroscopy measures contaminant (hydrogen) and bulk cathode (aluminum) plasma emission versus transported axial electron beam current turn on. Experiments were performed at accelerator parameters: V = −0.7V=−0.7 to −1.1 MV,−1.1MV, I(diode)=3–30 kA,I(diode)=3–30kA, and pulse length=0.4–1.0 μs.length=0.4–1.0μs. Experiments using a two-stage low power (100 W) argon/oxygen rf discharge followed by a higher power (200 W) pure argon rf discharge yielded an increase in cathode turn-on voltage required for axial current emission from 662±174 kV662±174kV to 981±97 kV.981±97kV.     Gravitomagnetism in Quantum Mechanics

    We give a systematic treatment of the quantum mechanics of a spin zero particle in a combined electromagnetic field and a weak gravitational field, which is produced by a slow moving matter source. The analysis is based on the Klein-Gordon equation expressed in generally covariant form and coupled minimally to the electromagnetic field. The Klein-Gordon equation is recast into Schroedinger equation form (SEF), which we then analyze in the non-relativistic limit. We include a discussion of some rather general observable physical effects implied by the SEF, concentrating on gravitomagnetism. Of particular interest is the interaction of the orbital angular momentum of the particle with the gravitomagnetic field.Comment: 9 page

    Effects of pressure on diffusion and vacancy formation in MgO from non-empirical free-energy integrations

    The free energies of vacancy pair formation and migration in MgO were computed via molecular dynamics using free-energy integrations and a non-empirical ionic model with no adjustable parameters. The intrinsic diffusion constant for MgO was obtained at pressures from 0 to 140 GPa and temperatures from 1000 to 5000 K. Excellent agreement was found with the zero pressure diffusion data within experimental error.     Efficient, high-throughput transfection of human embryonic stem cells

    INTRODUCTION: Genetic manipulation of human embryonic stem cells (hESC) has been limited by their general resistance to common methods used to introduce exogenous DNA or RNA. Efficient and high throughput transfection of nucleic acids into hESC would be a valuable experimental tool to manipulate these cells for research and clinical applications. METHODS: We investigated the ability of two commercially available electroporation systems, the Nucleofection(® )96-well Shuttle(® )System from Lonza and the Neon™ Transfection System from Invitrogen to efficiently transfect hESC. Transfection efficiency was measured by flow cytometry for the expression of the green fluorescent protein and the viability of the transfected cells was determined by an ATP catalyzed luciferase reaction. The transfected cells were also analyzed by flow cytometry for common markers of pluripotency. RESULTS: Both systems are capable of transfecting hESC at high efficiencies with little loss of cell viability. However, the reproducibility and the ease of scaling for high throughput applications led us to perform more comprehensive tests on the Nucleofection(® )96-well Shuttle(® )System. We demonstrate that this method yields a large fraction of transiently transfected cells with minimal loss of cell viability and pluripotency, producing protein expression from plasmid vectors in several different hESC lines. The method scales to a 96-well plate with similar transfection efficiencies at the start and end of the plate. We also investigated the efficiency with which stable transfectants can be generated and recovered under antibiotic selection. Finally, we found that this method is effective in the delivery of short synthetic RNA oligonucleotides (siRNA) into hESC for knockdown of translation activity via RNA interference. CONCLUSIONS: Our results indicate that these electroporation methods provide a reliable, efficient, and high-throughput approach to the genetic manipulation of hESC

    LIVE COLOR PATTERNS DIAGNOSE SPECIES: A TALE OF TWO HERICHTHYS

    The Rio Grande Cichlid, Herichthys cyanoguttatus, is native to the drainages of the Gulf Coast of northern Mexico and southern Texas and has been introduced at several sites in the US. Previous observations have suggested that non-native populations in Louisiana that are currently recognized as H. cyanoguttatus resemble another species, the Lowland Cichlid, H. carpintis. Traditional morphological and genetic techniques have been insufficient to differentiate these species, but H. carpintis has been reported to differ from H. cyanoguttatus in color pattern, so we turned to novel electronic photo archives to determine the identity of the species introduced in Louisiana. First, we used the public databases Nonindigenous Aquatic Species Database and Fishes of Texas to infer the historical distributions of these species in the US. We then used museum specimens, live specimens, and two additional databases, The Cichlid Room Companion and iNaturalist, to compare morphology and color patterns among individuals obtained from their native and introduced ranges in Mexico, Texas, and Louisiana. Our general observations found that H. cf. cyanoguttatus from Louisiana tended to have an obliquely oriented mouth and a more rounded ventral profile than H. cyanoguttatus from Texas, consistent with previous descriptions of H. carpintis, but our morphological analyses were unable to identify any significant differences among populations. Our analyses of color patterns found that H. cf. cyanoguttatus from Louisiana had larger iridescent spots than H. cyanoguttatus from Texas as well as black breeding coloration that extended anteriorly to the tip of the mouth, characters consistent with H. carpintis. Our observations indicate that at least some of the cichlids introduced in Louisiana are not H. cyanoguttatus but are instead H. carpintis, and that their presence there is likely due to release by humans. This is the first record of H. carpintis establishing a population in the US.
