    What does the structure-function relationship of the HIV-1 Tat protein teach us about developing an AIDS vaccine?

    The human immunodeficiency virus type 1 (HIV-1) trans-activator of transcription protein Tat is an important factor in viral pathogenesis. In addition to its function as the key trans-activator of viral transcription, Tat is also secreted by the infected cell and taken up by neighboring cells where it has an effect both on infected and uninfected cells. In this review we will focus on the relationship between the structure of the Tat protein and its function as a secreted factor. To this end we will summarize some of the exogenous functions of Tat that have been implicated in HIV-1 pathogenesis and the impact of structural variations and viral subtype variants of Tat on those functions. Finally, since in some patients the presence of Tat-specific antibodies or CTL frequencies are associated with slow or non-progression to AIDS, we will also discuss the role of Tat as a potential vaccine candidate, the advances made in this field, and the importance of using a Tat protein capable of eliciting a protective or therapeutic immune response to viral challenge

    Biomass Increases Go under Cover: Woody Vegetation Dynamics in South African Rangelands

    <div><p>Woody biomass dynamics are an expression of ecosystem function, yet biomass estimates do not provide information on the spatial distribution of woody vegetation within the vertical vegetation subcanopy. We demonstrate the ability of airborne light detection and ranging (LiDAR) to measure aboveground biomass and subcanopy structure, as an explanatory tool to unravel vegetation dynamics in structurally heterogeneous landscapes. We sampled three communal rangelands in Bushbuckridge, South Africa, utilised by rural communities for fuelwood harvesting. Woody biomass estimates ranged between 9 Mg ha<sup>-1</sup> on gabbro geology sites to 27 Mg ha<sup>-1</sup> on granitic geology sites. Despite predictions of woodland depletion due to unsustainable fuelwood extraction in previous studies, biomass in all the communal rangelands increased between 2008 and 2012. Annual biomass productivity estimates (10–14% p.a.) were higher than previous estimates of 4% and likely a significant contributor to the previous underestimations of modelled biomass supply. We show that biomass increases are attributable to growth of vegetation <5 m in height, and that, in the high wood extraction rangeland, 79% of the changes in the vertical vegetation subcanopy are gains in the 1-3m height class. The higher the wood extraction pressure on the rangelands, the greater the biomass increases in the low height classes within the subcanopy, likely a strong resprouting response to intensive harvesting. Yet, fuelwood shortages are still occurring, as evidenced by the losses in the tall tree height class in the high extraction rangeland. Loss of large trees and gain in subcanopy shrubs could result in a structurally simple landscape with reduced functional capacity. This research demonstrates that intensive harvesting can, paradoxically, increase biomass and this has implications for the sustainability of ecosystem service provision. The structural implications of biomass increases in communal rangelands could be misinterpreted as woodland recovery in the absence of three-dimensional, subcanopy information.</p></div

    Are there differences in performance, metabolism, and quadriceps muscle activity in black African and Caucasian athletes during brief intermittent and intense exercise?

    The purpose of the present study was to determine whether there are any differences in power output (PO) and/or quadriceps muscle (Quad) activity between black African and Caucasian football players during a force-velocity (fv) exercise test, which consisted of performing maximal 6-s sprints against an increasing load. Each subject started the test with a load of 2 kg and then recovered for 5 min before repeating the same test with a load increased by 2 kg. When the pedal frequency did not exceed 130 rev·min–1, the load was increased by only 1 kg. Each subject attained the load corresponding to his maximal power if an additional increase in load (+1 kg) induced a power decrease. Nine black Africans (mean age 24.2 ± 3.3 years) and nine Caucasians (24.7 ± 4.2 years) (matched for stature and aerobic fitness) participated in the fv exercise test. During the test, PO, blood lactate, and the quadriceps electromyography (EMG) root mean square (Quad RMS) were assessed. Higher blood lactate was observed in Caucasians than in black Africans for POs over the load range from 4 kg up to the maximal power. However, PO and Quad RMS values were similar in Caucasians and black Africans. They also had similar lean leg volume (LLV) and consequently produced similar PO/LLV and Quad RMS/LLV values. Overall, our results suggest that Caucasians and black Africans matched for stature, max, and training background have similar PO and Quad RMS values, but different blood lactate concentrations during brief, intermittent, intense exercise performed on a cycloergometer

    Effects of Strength Training on the Physiological Determinants of Middle- and Long-Distance Running Performance: A Systematic Review

The efficacy of strength training (ST) for distance runners has received considerable attention in the literature. However, to date, the results of these studies have not been fully synthesized in a review on the topic. Objectives: This systematic review aimed to provide a comprehensive critical commentary on the current literature that has examined the effects of ST modalities on the physiological determinants and performance of middle- and long-distance runners, and offer recommendations for best practice. Methods: Electronic databases were searched using a variety of key words relating to ST exercise and distance running. This search was supplemented with citation tracking. To be eligible for inclusion, a study was required to meet the following criteria: participants were middle- or long-distance runners with ≥ 6 months experience, a ST intervention (heavy resistance training, explosive resistance training, or plyometric training) lasting ≥ 4 weeks was applied, a running only control group was used, data on one or more physiological variables was reported. Two independent assessors deemed that 24 studies fully met the criteria for inclusion. Methodological rigor was assessed for each study using the PEDro scale. Results: PEDro scores revealed internal validity of 4, 5, or 6 for the studies reviewed. Running economy (RE) was measured in 20 of the studies and generally showed improvements (2–8%) compared to a control group, although this was not always the case. Time trial (TT) performance (1.5–10 km) and anaerobic speed qualities also tended to improve following ST. Other parameters [maximal oxygen uptake (V˙ O 2 max), velocity at V˙ O 2 max, blood lactate, body composition] were typically unaffected by ST. Conclusion: Whilst there was good evidence that ST improves RE, TT, and sprint performance, this was not a consistent finding across all works that were reviewed. Several important methodological differences and limitations are highlighted, which may explain the discrepancies in findings and should be considered in future investigations in this area. Importantly for the distance runner, measures relating to body composition are not negatively impacted by a ST intervention. The addition of two to three ST sessions per week, which include a variety of ST modalities are likely to provide benefits to the performance of middle- and long-distance runners
