The COVID-19 vaccination campaign in Bhutan: strategy and enablers
Bhutan has reported a total of 2596 COVID-19 cases and three deaths as of September 15, 2021. With support from India, the United States, Denmark, the People’s Republic of China, Croatia and other countries, Bhutan was able to conduct two rounds of nationwide vaccination campaign. While many countries struggle to overcome vaccine refusal or hesitancy due to complacency, a lack of trust, inconvenience and fear, escalated in some countries by anti-vaccine groups, Bhutan managed to inoculate more than 95% of its eligible populations in two rounds of vaccination campaign. Enabling factors of this successful vaccination campaign were strong national leadership, a well-coordinated national preparedness plan, and high acceptability of vaccine due to effective mass communication and social engagement led by religious figures, volunteers and local leaders. In this short report, we described the national strategic plan and enabling factors that led to the success of this historical vaccination campaign
Hypercholesterolemia is associated with hyperactive cardiac mTORC1 and mTORC2 signaling
Nutritional excess and hyperlipidemia increase the heart’s susceptibility to ischemic injury. Mammalian target of rapamycin (mTOR) controls the cellular response to nutritional status and may play a role in ischemic injury. To explore the effect of hypercholesterolemia on cardiac mTOR signaling, we assessed mTOR signaling in hypercholesterolemic swine (HC) that are also susceptible to increased cardiac ischemia-reperfusion injury. Yucatan pigs were fed a high-fat/high-cholesterol diet for 4 weeks to induce hypercholesterolemia, and mTOR signaling was measured by immunoblotting and immunofluorescence in the non-ischemic left ventricular area. Total myocardial mTOR and raptor levels were markedly increased in the HC group compared to the normocholesterolemic group, and directly correlated with serum cholesterol levels. mTOR exhibited intense perinuclear staining in myocytes only in the HC group. Hypercholesterolemia was associated with hyperactive signaling upstream and downstream of both mTOR complexes, including myocardial Akt, S6K1, 4EBP1, S6 and PKC-alpha, increased levels of cardiac hypertrophy markers, and a trend toward lower levels of myocardial autophagy. Hypercholesterolemia can now be added to the growing list of conditions associated with aberrant mTOR signaling. Hypercholesterolemia produces a unique profile of alterations in cardiac mTOR signaling, which is a potential target in cardiac diseases associated with hypercholesterolemia and nutritional excess
Short-Term Forecasting with Mixed-Frequency Data: A MIDASSO Approach
In this paper we extend the targeted-regressor approach suggested in Bai and Ng (2008) for variables sampled at the same frequency to mixed-frequency data. Our MIDASSO approach is a combination of the unrestricted MIxed-frequency DAta-Sampling approach (U-MIDAS) (see Foroni et al., 2015; Castle et al., 2009; Bec and Mogliani, 2013), and the LASSO-type penalised regression used in Bai and Ng (2008), called the elastic net (Zou and Hastie, 2005). We illustrate our approach by forecasting the quarterly real GDP growth rate in Switzerland
The homeobox gene Hex is required in definitive endodermal tissues for normal forebrain, liver and thyroid formation
The homeobox gene Hex is expressed in the anterior visceral endoderm (AVE) and rostral definitive endoderm of early mouse embryos. Later, Hex transcripts are detected in liver, thyroid and endothelial precursor cells. A null mutation was introduced into the Hex locus by homologous recombination in embryonic stem cells. Hex mutant embryos exhibit varying degrees of anterior truncation as well as liver and thyroid dysplasia. The liver diverticulum is formed but migration of hepatocytes into the septum transversum fails to occur. Development of the thyroid is arrested at the thyroid bud stage at 9.5 dpc. Brain defects are restricted to the rostral forebrain and have a caudal limit at the zona limitans intrathalamica, the boundary between dorsal and ventral thalamus. Analysis of Hex(−/−) mutants at early stages shows that the prospective forebrain ectoderm is correctly induced and patterned at 7.5 days post coitum (dpc), but subsequently fails to develop. AVE markers are expressed and correctly positioned but development of rostral definitive endoderm is greatly disturbed in Hex(−/−) embryos. Chimeric embryos composed of Hex(−/−) cells developing within a wild-type visceral endoderm show forebrain defects indicating that Hex is required in the definitive endoderm. All together, these results demonstrate that Hex function is essential in definitive endoderm for normal development of the forebrain, liver and thyroid gland
Systematic characterisation of the Herschel SPIRE Fourier Transform Spectrometer
A systematic programme of calibration observations was carried out to monitor
the performance of the SPIRE FTS instrument on board the Herschel Space
Observatory. Observations of planets (including the prime point-source
calibrator, Uranus), asteroids, line sources, dark sky, and cross-calibration
sources were made in order to monitor repeatability and sensitivity, and to
improve FTS calibration. We present a complete analysis of the full set of
calibration observations and use them to assess the performance of the FTS.
Particular care is taken to understand and separate out the effect of pointing
uncertainties, including the position of the internal beam steering mirror for
sparse observations in the early part of the mission. The repeatability of
spectral line centre positions is <5km/s, for lines with signal-to-noise ratios
>40, corresponding to <0.5-2.0% of a resolution element. For spectral line
flux, the repeatability is better than 6%, which improves to 1-2% for spectra
corrected for pointing offsets. The continuum repeatability is 4.4% for the SLW
band and 13.6% for the SSW band, which reduces to ~1% once the data have been
corrected for pointing offsets. Observations of dark sky were used to assess
the sensitivity and the systematic offset in the continuum, both of which were
found to be consistent across the FTS detector arrays. The average point-source
calibrated sensitivity for the centre detectors is 0.20 and 0.21 Jy [1 sigma; 1
hour], for SLW and SSW. The average continuum offset is 0.40 Jy for the SLW
