Panax notoginseng (Burk.) effects on fibrinogen and lipid plasma level in rats fed on a high-fat diet
Several studies have shown that notoginsenoides improve diastolic function in hypertensive subjects, induce the fibrinolytic system in in vitro models and act as antiproliferative agents on vessel leiomyocytes. Our aim was to evaluate their effect on fibrinogen and lipid plasma levels compared with a well-known HMGCoA reductase inhibitor. Seventy Wistar male adult rats on a fat-enriched diet were treated orally with P. notoginseng pulverized root (43 mg/kg/day or 86 mg/kg/day; 20 animals per group), fluvastatin (3 mg/kg/day; 20 animals) or physiological saline (5 mL/kg/day; 10 animals). The ten rats on a normocaloric diet were also treated with 5 mL/kg/day of physiological saline. After a 28-day treatment, the rats were killed and their blood analysed with standard procedures. Treatment with 43 mg/kg/day of P. notoginseng or 3 mg/kg/day of fluvastatin showed similar activity in decreasing total cholesterol (-23.70%, -19.29%, respectively) and triglycerides (-21.59%, -18.55%). The most evident effect of P. notoginseng was the reduction of fibrinogenaemia in treated rats compared with the control values (-38.10%; p < 0.001), no dose-relationship being shown in this effect. Moreover, no significant variation in HDL cholesterol and glucose levels was observed nor did relevant behavioural changes occur in association with the root intake. Besides a moderate, non dose-related decrease in the plasma lipid levels, P. notoginseng appeared to induce a significant reduction in the rat fibrinogenaemia. Copyrigh
Tuning the potential drop at graphene/protic ionic liquid interface by molecular structure engineering
Ionic liquids (ILs) have been extensively employed in many applications involving interfaces with carbon-based electrodes, such as energy storage devices (batteries or supercapacitors) or electrocatalytic devices, where the way each ion of the IL interacts with the electrode has a strong impact on the overall performance of the device. For instance, the amount of potential difference between the electrode and the bulk of the IL is highly sensitive to the IL composition and it is directly related to the device capacitance. The selection of the most suited pair of ions often proceeds by time-consuming and costly trial-and-error approaches. It is necessary to understand the atomistic features of the interface to determine the effect of each ion on the potential drop. By classical molecular dynamics simulations, we show that it is possible to quickly infer the interface potential arising at the carbon electrode by carefully inspecting the molecular structure of the IL. The ion orientation at the interface is, in fact, determined by the distribution of charges within the molecules. Depending on where charges are located, ions can either lie flat or perpendicular to the interface to minimize the surface energy. The interface potential is found to be mainly determined by ion-ion interactions dictating the interface energy minimization process, whereas ion-electrode interactions are found to enforce higher ordering and charge layers stacking but not to induce selective adsorption of an ion over the other
Combined action of SAMe, Folate, and Vitamin B12 in the treatment of mood disorders: a review.
Mood disorders affect more than 500 million people around the world. In the last decade, their prevalence has increased, and many people suffer from nervousness, anxiety, and stress at least once in their lives. The incidence of mood disorders and anxiety increases during perimenopause or under stressful conditions. The social restrictions introduced during the COVID-19 pandemic have significantly increased the normal burden of psychological and psychic disorders. In moderate to severe cases, pharmacological treatment is currently recommended, while in mild disorders, especially in the initial phase, psychological therapy is preferable. It is known that several nutrients are crucial for brain function. Among them, folate (vitamin B9), cyanocobalamin (vitamin B12), and S-adenosyl-L-methionine (SAMe) have been shown to influence various neurobiological processes. Overall, the available evidence suggests that dietary supplementation with folic acid, vitamin B12, and SAMe can be beneficial for people with mild mood disorders
Triptans: the experience of a clinical pharmacologist in clinical practice.
Despite the pharmacokinetic differences among triptans and the variety of ways of administration, the clinical differences in every day use of these drugs lack in an accepted decisional tree. In fact,
there are a number of comparative trials showing conflicting results with regard to efficacy, onset of action, safety and ecurrence incidence. That means that the patient’s preference probably is the main criterion for choosing one triptan vs. others. This point of view is probably correct considering also that the
main cause of therapy failure is noncompliance.
A good migraine care strategy requires a balance in what the patient views as satisfactory, a reasonable compromise between
efficacy and tolerability, and a careful follow-up. Improvement in compliance should be the main and more immediate goal for the treatment of migraine attacks
Ab initio Study of Misfit Dislocations at the SiC/Si(001) Interface
The high lattice mismatched SiC/Si(001) interface was investigated by means
of combined classical and ab initio molecular dynamics. Among the several
configurations analyzed, a dislocation network pinned at the interface was
found to be the most efficient mechanism for strain relief. A detailed
description of the dislocation core is given, and the related electronic
properties are discussed for the most stable geometry: we found interface
states localized in the gap that may be a source of failure of electronic
devices
Prediction of the structural and electronic properties of MoxTi1−xS2 monolayers via first principle simulations
Two-dimensional transition metal dichalcogenides have gained great attention because of their peculiar physical properties that make them interesting for a wide range of applications. Lately, alloying between different transition metal dichalcogenides has been proposed as an approach to control two-dimensional phase stability and to obtain compounds with tailored characteristics. In this theoretical study, we predict the phase diagram and the electronic properties of MoxTi1−xS2 at varying stoichiometry and show how the material is metallic, when titanium is the predominant species, while it behaves as a p-doped semiconductor, when approaching pure MoS2 composition. Correspondingly, the thermodynamically most stable phase switches from the tetragonal to the hexagonal one. Further, we present an example which shows how the proposed alloys can be used to obtain new vertical two-dimensional heterostructures achieving effective electron/hole separation
Progression-free survival as a surrogate endpoint of overall survival in patients with metastatic colorectal cancer
Background: In many clinical trials designed to assess the efficacy of anticancer treatments, overall
survival (OS) is often used as a primary endpoint despite its several points of weakness.
Methods: This study evaluated the role of progression-free survival (PFS) in the first three
lines of treatment as a potential surrogate endpoint of OS in patients with metastatic colorectal
cancer (MCRC). One hundred and twenty patients with MCRC were enrolled in this study. The
median PFS of the first-, second-, and third-lines of treatment and the OS were evaluated. The
correlation between the time to progression and the OS was analyzed. The median PFS of the
three lines of treatment were 8.5, 5, and 3 months, respectively.
Results: The median OS was 32.4 months. A modest correlation was found between the PFS
to the first-line treatment with Folfox\u2013avastin and OS. Similar data were obtained with the
second-line treatment. However, no correlation was found between the PFS and OS during the
third-line treatment. The regression analysis revealed that PFS is predictive of OS.
Conclusion: In brief, the PFS of the first- and second-lines of treatment could be a good candidate as a surrogate endpoint of OS in patients with MCRC
Cardiovascular autonomic function and MCI in Parkinson's disease
Introduction: dysautonomic dysfunction and cognitive impairment represent the most disabling non-motor features of Parkinson's Disease (PD). Recent evidences suggest the association between Orthostatic Hypotension (OH) and PD-Dementia. However, little is known on the interactions between cardiovascular dysautonomia and Mild Cognitive Impairment (MCI). We aimed to evaluate the association between cardiovascular dysautonomia and MCI in patients with PD. Methods: non-demented PD patients belonging to the PACOS cohort underwent a comprehensive instrumental neurovegetative assessment including the study of both parasympathetic and sympathetic function (30:15 ratio, Expiratory-Inspiratory ratio [E-I] and presence of Orthostatic Hypotension [OH]). Diagnosis of MCI was made according to the MDS criteria level II. Results: we enrolled 185 PD patients of whom 102 (55.1%) were men, mean age was 64.6 ± 9.7 years, mean disease duration of 5.6 ± 5.5 years with a mean UPDRS-ME score of 31.7 ± 10.9. MCI was diagnosed in 79 (42.7%) patients. OH was recorded in 52 (28.1%) patients, altered 30:15 ratio was recorded in 39 (24.1%) patients and an altered E-I ratio was found in 24 (19.1%) patients. Presence of MCI was associated with an altered 30:15 ratio (adjOR 2.83; 95%CI 1.25–6.40) but not with an altered E-I ratio, while OH was associated only with the amnestic MCI subgroup (OR 2.43; 95% CI 1.05–5.06).
