Materials based on BIFEVOX and bismuth or iron simple oxides nanopowders
Received: 22.09.2017; accepted: 17.10.2017; published: 20.10.2017.Compositions of composite materials based on BIFEVOX and nanopowders of bismuth and iron oxides have been obtained. The absence of chemical interaction between the components has been proved, the total electrical conductivity of materials in the average temperature region has been determined. It has been shown that under the selected formation conditions, it has not yet been possible to achieve significant improvement of the functional characteristics of heterogeneous compositions in comparison with individual phases. However positive results on chemical and structural stability give way to further investigations. The work was partially supported by the Scholarship of the President (SP-3376.2016.1) and Russian Foundation for Basic Research (project No 17-53-04098)
Derivation, Characterization, and Stable Transfection of Induced Pluripotent Stem Cells from Fischer344 Rats
The rat represents an important animal model that, in many respects, is superior to the mouse for dissecting behavioral, cardiovascular and other physiological pathologies relevant to humans. Derivation of induced pluripotent stem cells from rats (riPS) opens the opportunity for gene targeting in specific rat strains, as well as for the development of new protocols for the treatment of different degenerative diseases. Here, we report an improved lentivirus-based hit-and-run riPS derivation protocol that makes use of small inhibitors of MEK and GSK3. We demonstrate that the excision of proviruses does not affect either the karyotype or the differentiation ability of these cells. We show that the established riPS cells are readily amenable to genetic manipulations such as stable electroporation. Finally, we propose a genetic tool for an improvement of riPS cell quality in culture. These data may prompt iPS cell-based gene targeting in rat as well as the development of iPS cell-based therapies using disease models established in this species
Status of selective permeability of hematosalivary barrier in individuals of different age groups
Hematosalivary barrier (HSB) maintains the relative im mutability of physical, chemical and biological properties of saliva, which ensures the proper functioning of the oral cavity. GSB is a hallmark of selective permeability. Aim of this study was to investigate some biochemical parameters (calcium, m agnesium , phosphorus, protein, TBC-active products) of oral fluid of individuals of different age groups. 90 people of different age groups were studied. The first main group consisted of 30 people aged 60 to 86 years, the second group consisted of 30 people from 45 to 59 years and the third group included 30 people aged 2 0-30 years. The content of total protein, total Π‘Π°, P, Mg, and TBC - active products were determ ined in oral fluid. Studies carried out demonstrated significant differences in the content of basic m acronutrients and other substances in the oral fluid in different age groups, that allows to speak about the change of functioning and selective permeability of HSB depending on age.ΠΠ΅ΠΌΠ°ΡΠΎΡΠ°Π»ΠΈΠ²Π°ΡΠ½ΡΠΉ Π±Π°ΡΡΠ΅Ρ (ΠΠ‘Π) ΠΏΠΎΠ΄Π΄Π΅ΡΠΆΠΈΠ²Π°Π΅Ρ ΠΎΡΠ½ΠΎΡΠΈΡΠ΅Π»ΡΠ½ΡΡ Π½Π΅ΠΈΠ·ΠΌΠ΅Π½Π½ΠΎΡΡΡ ΡΠΎΡΡΠ°Π²Π° ΡΠΈΠ·ΠΈΡΠ΅ΡΠΊΠΈΡ

Kpna6 deficiency causes infertility in male mice by disrupting spermatogenesis
Spermatogenesis is driven by an ordered series of events, which rely on trafficking of specific proteins between nucleus and cytoplasm. The importin Ξ± family of proteins mediates movement of specific cargo proteins when bound to importin Ξ². Importin Ξ± genes have distinct expression patterns in mouse testis, implying they may have unique roles during mammalian spermatogenesis. Here we use a loss-of-function approach to specifically determine the role of importin Ξ±7 in spermatogenesis and male fertility. We show that ablation of importin Ξ±7 in male mice leads to infertility and has multiple cumulative effects on both germ cells and Sertoli cells. Importin Ξ±7-deficient mice exhibit an impaired Sertoli cell function, including loss of Sertoli cells and a compromised nuclear localization of the androgen receptor. Furthermore, our data demonstrate devastating defects in spermiogenesis including incomplete sperm maturation and massive loss of sperms that are accompanied by disturbed histone-protamine-exchange, differential localization of the transcriptional regulator Brwd1 and altered expression of Rfx2 target genes. Our work uncovers the essential role of importin Ξ±7 in spermatogenesis and hence in male fertility
Evaluation of selective permeability of the hematosalivary barrier with lichen planus of the oral mucosa
The hematosalivary barrier selectively regulates metabolism, including trace elements between the blood and the internal contents of the salivary glands, provides a relatively constant composition of the physical, chemical and biological properties of mouth liquid. The aim was to study the content of elements (copper, zinc) in serum of blood and oral fluid of patients with oral lichen planus (OLP). The study involved only 95 patients: first group consisted of 19 people with the typical form of the OLP, second group -19 people with exudative-hyperemic form of OLP, third group consisted of 27 patients with erosive-ulcerative form, the reference group consisted of 30 healthy individuals. With increasing severity of the clinical course of the OLP (with exudative-hyperemic and erosive- ulcerative forms) function and the selective permeability of the hematosalivary barrier in respect of essential microelements - zinc and copper - are violated.ΠΠ΅ΠΌΠ°ΡΠΎΡΠ°Π»ΠΈΠ²Π°ΡΠ½ΡΠΉ Π±Π°ΡΡΠ΅Ρ ΠΈΠ·Π±ΠΈΡΠ°ΡΠ΅Π»ΡΠ½ΠΎ ΡΠ΅Π³ΡΠ»ΠΈΡΡΠ΅Ρ ΠΎΠ±ΠΌΠ΅Π½ Π²Π΅ΡΠ΅ΡΡΠ², Π² ΡΠΎΠΌ ΡΠΈΡΠ»Π΅, ΠΈ ΠΌΠΈΠΊΡΠΎΡΠ»Π΅ΠΌΠ΅Π½ΡΠΎΠ² ΠΌΠ΅ΠΆΠ΄Ρ ΠΊΡΠΎΠ²ΡΡ ΠΈ Π²Π½ΡΡΡΠ΅Π½Π½ΠΈΠΌ ΡΠΎΠ΄Π΅ΡΠΆΠΈΠΌΡΠΌ ΡΠ»ΡΠ½Π½ΡΡ

The therapeutic effectiveness of the drug "Longidaza" in the complex treatment of oral lichen planus
The aim of the work was to determine the therapeutic effectiveness of the drug 'Longidaza' in the complex treatment of oral lichen planus. The study was performed involving 112 patients with various forms of oral lichen planus in age from 24 to 70 years. The first group included 54 patients who in the complex treatment used the drug 'Longidaza.' The second (control) group consisted of 58 people, who as part of an integrated treatment did not use the drug 'Longidaza.' It was found a significant improvement of the effectiveness of treatment when the drug 'Longidaza' was in the complex treatment of oral lichen planus, which was confirmed by positive clinical dynamics: faster flattening of papules, reducing the severity of the pattern on the mucosa, reduction of terms of epithelialization of erosions, longer remission in all forms of the disease.Π¦Π΅Π»ΡΡ ΡΠ°Π±ΠΎΡΡ ΡΠ²ΠΈΠ»ΠΎΡΡ ΠΎΠΏΡΠ΅Π΄Π΅Π»Π΅Π½ΠΈΠ΅ ΡΠ΅ΡΠ°ΠΏΠ΅Π²ΡΠΈΡΠ΅ΡΠΊΠΎΠΉ ΡΡΡΠ΅ΠΊΡΠΈΠ²Π½ΠΎΡΡΠΈ ΠΏΡΠ΅ΠΏΠ°ΡΠ°ΡΠ° Β«ΠΠΎΠ½Π³ΠΈΠ΄Π°Π·Π°Β» Π² ΠΊΠΎΠΌΠΏΠ»Π΅ΠΊΡΠ½ΠΎΠΌ Π»Π΅ΡΠ΅Π½ΠΈΠΈ ΠΊΡΠ°ΡΠ½ΠΎΠ³ΠΎ ΠΏΠ»ΠΎΡΠΊΠΎΠ³ΠΎ Π»ΠΈΡΠ°Ρ ΡΠ»ΠΈΠ·ΠΈΡΡΠΎΠΉ ΠΎΠ±ΠΎΠ»ΠΎΡΠΊΠΈ ΡΡΠ°. Role of ortopedic correction of dentition in complex treatment of patients with oral lichen planus
Most patients suffering from Oral Lichen Planus (OLP) need orthopedic care. The aim of this study was to evaluate the role of prosthesis in patients OLP. We observed 78 patients (8 men and 70 wom en) with oral lichen planus aged from 35 to 76 years with typical, exudative -hyperem ic, erosive-ulcer form s of oral lichen planus. At the same tim e they were divided into 2 groups: 45 patients who underwent a comprehensive com mon m edication with further rational prosthesis (group 1), 33 patients who underwent complex conventional medication without prosthesis (because of financial capabilities of patients) (group 2). The biocompatibility of materials was evaluated, before preparing oral prosthetics for m anufacturing of prosthetic. Prosthetics held after epithelialization of erosions. Patients in Group 1 after treatm ent and prosthetics (the 45th day) reported im provem ent of the oral mucosa. In Group 2, all form s of OLP in the mucosa remained papular pattern, but erosion and ulcers arose again. Patients OLP efficiency when the rational prosthesis in the treatm ent scheme confirm ed by the increase of disease remission period in all forms of the CPL (typical form: in prosthetics - 1 5 (3-24) m onths, the traditional treatm ent - 5 (2 -10 ) months: exudative -hyperem ic form: 8 (3-15) months and 3.5 (1.5 -7) months: erosive- ulcerative form: 9 (4 -10 ) m onths and one (2 -6) month respectively).ΠΠΎΠ»ΡΡΠΈΠ½ΡΡΠ²ΠΎ ΠΏΠ°ΡΠΈΠ΅Π½ΡΠΎΠ², ΡΡΡΠ°Π΄Π°ΡΡΠΈΡ

Generation of Trophoblast Stem Cells from Rabbit Embryonic Stem Cells with BMP4
Trophoblast stem (TS) cells are ideal models to investigate trophectoderm differentiation and placental development. Herein, we describe the derivation of rabbit trophoblast stem cells from embryonic stem (ES) cells. Rabbit ES cells generated in our laboratory were induced to differentiate in the presence of BMP4 and TS-like cell colonies were isolated and expanded. These cells expressed the molecular markers of mouse TS cells, were able to invade, give rise to derivatives of TS cells, and chimerize placental tissues when injected into blastocysts. The rabbit TS-like cells maintained self-renewal in culture medium with serum but without growth factors or feeder cells, whilst their proliferation and identity were compromised by inhibitors of FGFs and TGF-Ξ² receptors. Taken together, our study demonstrated the derivation of rabbit TS cells and suggested the essential roles of FGF and TGF-Ξ² signalings in maintenance of rabbit TS cell self-renewal
