    Arctic Ocean Simulations in the CMIP6 Ocean Model Intercomparison Project (OMIP)

    oai:publications.copernicus.org:gmdd107357Arctic Ocean simulations in 19 global ocean-sea ice models participating in the Ocean Model Intercomparison Project (OMIP) of the CMIP6 are evaluated in this paper. Our results indicate that no significant improvements were achieved in the Arctic Ocean simulations from the previous Coordinated Ocean-ice Reference Experiments phase II (CORE-II) to the current OMIP. Large model biases and inter-model spread exist in the simulated mean state of the halocline and Atlantic Water layer in the OMIP models. Most of the OMIP models suffer from too thick and deep Atlantic Water layer, too deep halocline base, and large fresh biases in the halocline. The OMIP models largely agree on the inter-annual and decadal variability of the Arctic Ocean freshwater content and volume/heat/freshwater transports through the Arctic Ocean gateways. The models can reproduce observed changes in volume, heat and freshwater transports through the gateways except for the Bering Strait. Overall, the performance of the Arctic Ocean simulations is similar between the CORE2-forced OMIP-1 and JRA55-do-forced OMIP-2.</p

    Idiopathic Male Infertility Is Strongly Associated with Aberrant Promoter Methylation of Methylenetetrahydrofolate Reductase (MTHFR)

    Abnormal germline DNA methylation in males has been proposed as a possible mechanism compromising spermatogenesis of some men currently diagnosed with idiopathic infertility. Previous studies have been focused on imprinted genes with DNA methylation in poor quality human sperms. However, recent but limited data have revealed that sperm methylation abnormalities may involve large numbers of genes or shown that genes that are not imprinted are also affected.Using the methylation-specific polymerase chain reaction and bisulfite sequencing method, we examined methylation patterns of the promoter of methylenetetrahydrofolate reductase (MTHFR) gene (NG_013351: 1538-1719) in sperm DNA obtained from 94 idiopathic infertile men and 54 normal fertile controls. Subjects with idiopathic infertility were further divided into groups of normozoospermia and oligozoospermia. Overall, 45% (41/94) of idiopathic infertile males had MTHFR hypermethylation (both hemimethylation and full methylation), compared with 15% of fertile controls (P<0.05). Subjects with higher methylation level of MTHFR were more likely to have idiopathic male infertility (P-value for trend  = 0.0007). Comparing the two groups of idiopathic infertile subjects with different sperm concentrations, a higher methylation pattern was found in the group with oligozoospermia.Hypermethylation of the promoter of MTHFR gene in sperms is associated with idiopathic male infertility. The functional relevance of hypermathylation of MTHFR to male fertility warrants further investigation

    Investigation of Dynamic Coupling Coordination between Urbanization and the Eco-Environment—A Case Study in the Pearl River Delta Area

    The interaction between urbanization and the eco-environment is usually viewed as an effect–feedback framework. Its coupling system is composed of urbanization and eco-environment subsystems. In this paper, the coupling degree (CD) and the coupling coordinated degree (CCD) are used to reflect the coupling interaction and coupling coordination between the urbanization subsystem and the eco-environment subsystem. Based on the dynamic relative quantities of urbanization and eco-environment data in the Pearl River Delta, CD and CCD values were calculated, and the spatiotemporal evolution trend of coordination was analyzed. The results show that (1) from 2000 to 2015, the nine cities in the Pearl River Delta had high CD values and CCD values. Though they had different performances in different periods, they were all in a coordinated class, including good coordination (GC), moderate coordination (MC), and bare coordination (BC). (2) In terms of temporal evolution, the coupling coordination between urbanization and the eco-environment in the entire Pearl River Delta greatly improved. (3) From the perspective of spatial distribution, the coupling coordination of the central region was higher than that of the peripheral regions, and that of the west bank of the Pearl River was higher than that of the east bank of the Pearl River. These results can help local policy makers enact appropriate measures for sustainable development

    Doping-Promoted Solar Water Oxidation on Hematite Photoanodes

    As one of the most promising materials for solar water oxidation, hematite has attracted intense research interest for four decades. Despite their desirable optical band gap, stability and other attractive features, there are great challenges for the implementation of hematite-based photoelectrochemical cells. In particular, the extremely low electron mobility leads to severe energy loss by electron hole recombination. Elemental doping, i.e., replacing lattice iron with foreign atoms, has been shown to be a practical solution. Here we review the significant progresses in metal and non-metal element doping-promoted hematite solar water oxidation, focusing on the role of dopants in adjusting carrier density, charge collection efficiency and surface water oxidation kinetics. The advantages and salient features of the different doping categories are compared and discussed

    Transcriptome and DNA Methylome Dynamics during Triclosan-Induced Cardiomyocyte Differentiation Toxicity

    Cardiac development is a dynamic process and sensitive to environmental chemicals. Triclosan is widely used as an antibacterial agent and reported to transport across the placenta and affect embryonic development. Here, we used human embryonic stem cell- (hESC-) derived cardiomyocytes (CMs) to determine the effects of TCS exposure on cardiac development. After TCS treatment, the differentiation process was significantly blocked and spontaneous beating rates of CMs were also decreased. Transcriptome analysis showed the dysregulation of genes involved in cardiogenesis, including GATA4 and TNNT2. Additionally, DNA methylation was also altered by TCS exposure, especially in those regions with GATA motif enrichment. These alterations of transcriptome and DNA methylation were all associated with signaling pathways integral to heart development. Our findings indicate that TCS exposure might cause cardiomyocyte differentiation toxicity and provide the new insights into how environmental factors regulate DNA methylation and gene expressions during heart development

    Administration of simvastatin after kainic acid-induced status epilepticus restrains chronic temporal lobe epilepsy.

    In this study, we examined the effect of chronic administration of simvastatin immediately after status epilepticus (SE) on rat brain with temporal lobe epilepsy (TLE). First, we evaluated cytokines expression at 3 days post KA-lesion in hippocampus and found that simvastatin-treatment suppressed lesion-induced expression of interleukin (IL)-1β and tumor necrosis factor-α (TNF-α). Further, we quantified reactive astrocytosis using glial fibrillary acidic protein (GFAP) staining and neuron loss using Nissl staining in hippocampus at 4-6 months after KA-lesion. We found that simvastatin suppressed reactive astrocytosis demonstrated by a significant decrease in GFAP-positive cells, and attenuated loss of pyramidal neurons in CA3 and interneurons in dentate hilar (DH). We next assessed aberrant mossy fiber sprouting (MFS) that is known to contribute to recurrence of spontaneous seizure in epileptic brain. In contrast to the robust MFS observed in saline-treated animals, the extent of MFS was restrained by simvastatin in epileptic rats. Attenuated MFS was related to decreased neuronal loss in CA3 and DH, which is possibly a mechanism underlying decreased hippocampal susceptibility in animal treated with simvastatin. Electronic encephalography (EEG) was recorded during 4 to 6 months after KA-lesion. The frequency of abnormal spikes in rats with simvastatin-treatment decreased significantly compared to the saline group. In summary, simvastatin treatment suppressed cytokines expression and reactive astrocytosis and decreased the frequency of discharges of epileptic brain, which might be due to the inhibition of MFS in DH. Our study suggests that simvastatin administration might be a possible intervention and promising strategy for preventing SE exacerbating to chronic epilepsy

    Insights into the hydrogen-bond cross-linking effects of small multiamine molecules on physical and mechanical properties of poly(vinly alcohol) by molecular dynamics simulations

    Small organic multiamine and multihydroxyl molecules have great potential for enhancing overall properties of poly(vinyl alcohol) (PVA) through the cross-linking effect of hydrogen bonds. However, experimentally there remains a remarkable lack of insightful understanding of the cross-linking effect on a molecular level. In the work, we report molecular dynamics simulations to reveal the cross-linking effect of hydrogen bonds of tetraaminopyrimidine (4N-2456) molecules on the structure, chain dynamics and mechanical properties of the PVA matrix. It was found that the addition of 4N-2456 leads to a nonlinear decrease of the free volume of PVA. A critical concentration of 4N-2456, about 5 wt%, was identified, resulting in the formation of 4N-2456 clusters. At this concentration, the PVA chains show the relatively slow mobility, the higher glass transition temperature and elastic modulus. Further increasing the 4N-2456 concentration enhances aggregation, and conversely weakens the interactions of hydrogen bonds between the PVA chains. Our work offers an understanding of how the 4N-2456 molecules influence the PVA chain dynamics and mechanical properties of the PVA matrix on molecular level
