    Genome-Wide Identification and Analysis of the Chicken Basic Helix-Loop-Helix Factors

    Members of the basic helix-loop-helix (bHLH) family of transcription factors play important roles in a wide range of developmental processes. In this study, we conducted a genome-wide survey using the chicken (Gallus gallus) genomic database, and identified 104 bHLH sequences belonging to 42 gene families in an effort to characterize the chicken bHLH transcription factor family. Phylogenetic analyses revealed that chicken has 50, 21, 15, 4, 8, and 3 bHLH members in groups A, B, C, D, E, and F, respectively, while three members belonging to none of these groups were classified as ‘‘orphans”. A comparison between chicken and human bHLH repertoires suggested that both organisms have a number of lineage-specific bHLH members in the proteomes. Chromosome distribution patterns and phylogenetic analyses strongly suggest that the bHLH members should have arisen through gene duplication at an early date. Gene Ontology (GO) enrichment statistics showed 51 top GO annotations of biological processes counted in the frequency. The present study deepens our understanding of the chicken bHLH transcription factor family and provides much useful information for further studies using chicken as a model system

    Masking Improves Contrastive Self-Supervised Learning for ConvNets, and Saliency Tells You Where

    While image data starts to enjoy the simple-but-effective self-supervised learning scheme built upon masking and self-reconstruction objective thanks to the introduction of tokenization procedure and vision transformer backbone, convolutional neural networks as another important and widely-adopted architecture for image data, though having contrastive-learning techniques to drive the self-supervised learning, still face the difficulty of leveraging such straightforward and general masking operation to benefit their learning process significantly. In this work, we aim to alleviate the burden of including masking operation into the contrastive-learning framework for convolutional neural networks as an extra augmentation method. In addition to the additive but unwanted edges (between masked and unmasked regions) as well as other adverse effects caused by the masking operations for ConvNets, which have been discussed by prior works, we particularly identify the potential problem where for one view in a contrastive sample-pair the randomly-sampled masking regions could be overly concentrated on important/salient objects thus resulting in misleading contrastiveness to the other view. To this end, we propose to explicitly take the saliency constraint into consideration in which the masked regions are more evenly distributed among the foreground and background for realizing the masking-based augmentation. Moreover, we introduce hard negative samples by masking larger regions of salient patches in an input image. Extensive experiments conducted on various datasets, contrastive learning mechanisms, and downstream tasks well verify the efficacy as well as the superior performance of our proposed method with respect to several state-of-the-art baselines

    Chronic stress induces steatohepatitis while decreases visceral fat mass in mice

    BACKGROUND: Prolonged stress leads over time to allostatic load on the body and is likely to exacerbate a disease process. Long-term of stress exposure is one of a risk factor for metabolism-related diseases such as obesity and type 2 diabetes. However, the relationship between chronic stress and non-alcoholic fatty liver disease (NAFLD) remain unknown. METHODS: To address the hypothesis that chronic stress associate to NAFLD development, we subjected C57bl/6 mice to electric foot shock and restraint stress for 12 weeks to set up chronic stress model. Then the serum and hepatic triglyceride (TG), total cholesterol (TC) were measured. Hepatic HE and Oil red O staining were used to specify the state of the NAFLD. To investigate whether inflammation takes part in the stress-induced NAFLD process, related visceral fat, serum and hepatic inflammatory factors were measured. RESULTS: We observed that chronic stress led to an overall increase of hepatic triglyceride and cholesterol while decreasing body weight and visceral fat mass. Microvesicular steatosis, lobular inflammation and ballooning degeneration were seen in stress liver section. This effect was correlated with elevated hepatic and serum inflammatory factors. Although the amount of visceral fat was decreased in stress group, various adipocytokines were elevated. CONCLUSIONS: We showed that chronic stress is associated to NAFLD and chronic inflammation in visceral fat, though food intake and visceral fat mass were decreased. These results may contribute to better understanding of the mechanism from steatosis to steatohepatitis, and propose a novel insight into the prevention and treatment of NAFLD

    Molecular Analysis of Spring Viraemia of Carp Virus in China: A Fatal Aquatic Viral Disease that Might Spread in East Asian

    Spring viraemia of carp (SVC) is a fatal viral disease for cyprinid fish, which is caused by spring viraemia of carp virus (SVCV). To date, no SVC outbreak has been reported in China. Between 1998 and 2002, outbreaks of SVC were reported in ornamental and wild fish in Europe and America, imported from multiple sources including China. Based on phylogenetic analysis, the viral strain isolated from America was shown to be originated from Asia. These outbreaks not only resulted in huge economic losses, but also raise an interesting question as to whether SVCV really exists in China and if so, is it responsible for SVC outbreaks? From 2002 to 2006, we screened 6700 samples from ornamental fish farms using the cell culture method of the Office International des Epizooties (OIE), and further verified the presence of SVCV by ELISA and real-time quantitative RT-PCR. Two infected samples were found and the complete genome of SVCV was sequenced from one of the isolates, termed SVCV-C1. Several unique hallmarks of SVCV-C1 were identified, including six amino acid (KSLANA) insertion in the viral RNA-dependent RNA polymerase (L) protein and ten nucleotide insertion in the region between glycoprotein (G) and L genes in European SVCV strains. Phylogenetic tree analysis of the full-length G protein of selected SVCV isolates from the United Kingdom and United States revealed that G proteins could be classified into Ia and Id sub genogroups. The Ia sub genogroup can be further divided into newly defined sub genogroups Ia-A and Ia-B. The isolates derived from the United States and China including the SVCV-C1 belongs to in the Ia-A sub genogroup. The SVCV-C1 G protein shares more than 99% homology with the G proteins of the SVCV strains from England and the United States, making it difficult to compare their pathogenicity. Comparison of the predicted three-dimensional structure based on the published G protein sequences from five SVCV strains revealed that the main differences were in the loops of the pleckstrin homology domains. Since SVCV is highly pathogenic, we speculate that SVC may therefore pose a serious threat to farmed cyprinid fish in China

    Antioxidant and Anticancer Activities of Wampee (Clausena lansium (Lour.) Skeels) Peel

    Antioxidant activities of wampee peel extracts using five different solvents (ethanol, hexane, ethyl acetate, butanol and water) were determined by using in-vitro antioxidant models including total antioxidant capability, 1,1-diphenyl-2-picryl hydrazyl (DPPH) radical scavenging activity, reducing power, and superoxide scavenging activity. Ethyl acetate fraction (EAF) exhibited the highest antioxidant activity compared to other fractions, even higher than synthetic antioxidant butylated hydroxyl toluene (BHT). In addition, the EAF exhibited strong anticancer activities against human gastric carcinoma (SGC-7901), human hepatocellular liver carcinoma (HepG-2) and human lung adenocarcinoma (A-549) cancer cell lines, higher than cisplatin, a conventional anticancer drug. The total phenolic content of wampee fraction was positively correlated with the antioxidant activity. This is the first report on the antioxidant and anticancer activities of the wampee peel extract. Thus, wampee peel can be used potentially as a readily accessible source of natural antioxidants and a possible pharmaceutical supplement

    Adversarial Examples with Unlimited Amount of Additions

    Malware detection methods based on gray images and deep learning have the characteristics of high detection accuracy and no need of feature engineering. Unfortunately, adversarial examples (AEs) can deceive such detection methods. However, it is difficult to reduce the detection accuracy of this kind of detection method greatly without destroying the functional integrity of the original file. By analyzing the structure and loading mechanism of portable executable (PE) files, this paper proposes an unrestricted add-amount bytecode attack (BAUAA). BAUAA generates adversarial samples by adding bytecode to a “section additional space” in the PE file that is scattered after each section and is not loaded into memory, and because of the unlimited amount of this space that can be added, the generated adversarial samples can be transformed into grayscale images that vary in size and texture, which can affect the discrimination accuracy of gray images and deep learning-based malware detection methods. The experimental results show that the detection accuracy of the malware detection method based on gray images and deep learning for the AEs generated by BAUAA is significantly lower than that for the non-AEs. To avoid the abuse of BAUAA in reality, it proposes a targeted AE detection method
