    Cure of chronic viral infection and virus-induced type 1 diabetes by neutralizing antibodies

    The use of neutralizing antibodies is one of the most successful methods to interfere with receptor–ligand interactions in vivo. In particular blockade of soluble inflammatory mediators or their corresponding cellular receptors was proven an effective way to regulate inflammation and/or prevent its negative consequences. However, one problem that comes along with an effective neutralization of inflammatory mediators is the general systemic immunomodulatory effect. It is, therefore, important to design a treatment regimen in a way to strike at the right place and at the right time in order to achieve maximal effects with minimal duration of immunosuppression or hyperactivation. In this review, we reflect on two examples of how short time administration of such neutralizing antibodies can block two distinct inflammatory consequences of viral infection. First, we review recent findings that blockade of IL-10/IL-10R interaction can resolve chronic viral infection and second, we reflect on how neutralization of the chemokine CXCL10 can abrogate virus-induced type 1 diabetes

    Perception and Use of Communication Care Plans by Certified Nursing Assistants in Nursing Homes: The Role of Professional Support

    The majority of individuals in nursing homes have cognitive-communication impairments which impact quality of care because direct care providers, certified nursing assistants, (CNAs) are unsure how to respond to resident’s communication behaviors. One intervention that facilitates staff-resident communication in nursing homes is communication plans; however, the research to date about communication plans has not specifically involved CNAs. The purpose of this study, using a grounded theory qualitative approach, was to describe development, implementation and evaluation of communication care plans (CCPs) for residents with cognitive-communication impairments in nursing homes by CNAs who did and did not receive professional support. Communication care plans are communication plans with the addition of autobiographical information. Twenty residents and ten CNAs from two nursing homes participated in the study. Once CCPs were created, CNA participants in one facility received support each day. Following two weeks, CNAs participated in a semi-structured interview. Interviews were transcribed and analyzed using open, axial and selective coding. Findings revealed a core category, meeting resident’s needs through professional support and communication, which describes the progressive process these CNAs underwent to effectively communicate with residents in nursing homes using CCPs. Evolution of this process occurred as CNA participants became familiar with residents. An underlying component facilitating this familiarity was support during CCP implementation. Initially, these CNAs had negative views about nursing home care because they were unsure how to communicate with residents and received little support from higher levels of nursing authority. Over time and with application of CCPs, CNAs became familiar with residents and their communication behaviors. Application of specific communication strategies on CCPs required ongoing support from the investigator/speech-language pathologist which was evident by the comments between CNA participants from each facility. Participants from both facilities reported positive experiences during application of the autobiographical information on the CCPs. This personal information coupled with increased knowledge about resident’s specific communication abilities fostered the formation of a relationship between residents and CNAs. In summary, support during application of CCPs supplements CNAs’ abilities to meet residents’ needs

    ERBlox: Combining Matching Dependencies with Machine Learning for Entity Resolution

    Entity resolution (ER), an important and common data cleaning problem, is about detecting data duplicate representations for the same external entities, and merging them into single representations. Relatively recently, declarative rules called matching dependencies (MDs) have been proposed for specifying similarity conditions under which attribute values in database records are merged. In this work we show the process and the benefits of integrating three components of ER: (a) Classifiers for duplicate/non-duplicate record pairs built using machine learning (ML) techniques, (b) MDs for supporting both the blocking phase of ML and the merge itself; and (c) The use of the declarative language LogiQL -an extended form of Datalog supported by the LogicBlox platform- for data processing, and the specification and enforcement of MDs.Comment: To appear in Proc.     Blockade but not overexpression of the junctional adhesion molecule C influences virus-induced type 1 diabetes in mice

    Type 1 diabetes (T1D) results from the autoimmune destruction of insulin-producing beta-cells in the pancreas. Recruitment of inflammatory cells is prerequisite to beta-cell-injury. The junctional adhesion molecule (JAM) family proteins JAM-B and JAM–C are involved in polarized leukocyte transendothelial migration and are expressed by vascular endothelial cells of peripheral tissue and high endothelial venules in lympoid organs. Blocking of JAM-C efficiently attenuated cerulean-induced pancreatitis, rheumatoid arthritis or inflammation induced by ischemia and reperfusion in mice. In order to investigate the influence of JAM-C on trafficking and transmigration of antigen-specific, autoaggressive T-cells, we used transgenic mice that express a protein of the lymphocytic choriomeningitis virus (LCMV) as a target autoantigen in the β-cells of the islets of Langerhans under the rat insulin promoter (RIP). Such RIP-LCMV mice turn diabetic after infection with LCMV. We found that upon LCMV-infection JAM-C protein was upregulated around the islets in RIP-LCMV mice. JAM-C expression correlated with islet infiltration and functional beta-cell impairment. Blockade with a neutralizing anti-JAM-C antibody reduced the T1D incidence. However, JAM-C overexpression on endothelial cells did not accelerate diabetes in the RIP-LCMV model. In summary, our data suggest that JAM-C might be involved in the final steps of trafficking and transmigration of antigen-specific autoaggressive T-cells to the islets of Langerhans

    The effect of vineyard long-term monoculture soil on production of volatile compounds and photosynthetic apparatus in grapevine leaves

    The effect of soil collected from long-term (1007 years of duration of monoculture) and short-term (55 years) grapevine monoculture on production of volatile organic compounds (VOC’s) and on functional parameters of photosystem II in grapevine leaves was analyzed. Grapevine plantlets grown in tested soils showed differences in VOC’s production after five months cultivation. Chlorophyll a fluorescence measurements by JIP-test revealed that the photosystem II was less efficient but the fluorescence intensity increased in plant growing in soil from the long-term monoculture compared to plants growing in the short term monoculture soil. Pseudomonas spp. carrying the biocontrol genes phlD and hcnAB were isolated from long-term monoculture soil. A consortium of ten of these isolates was added to the short term monoculture soil. The plants grown in this inoculated soil showed similar changes in fluorescence intensity and photosystem efficacy as the plants growing in long term monoculture. In this study, simple tools for measurement of a “soil effect” by measuring only a leaf have been tested successfully. They have allowed exhibiting the influence of long-term monoculture on plant physiology.

    A geothermal plant from a time-scale perspective

    In recent years, geothermal energy use from low-temperature sandstone reservoirs has sharply increased. Nonetheless, the injection of heat-depleted geothermal fluids has not been an easy task because of well/formation damage and operational/economic issues. Sønderborg geothermal plant is a case example of heat-mining from a low-temperature reservoir. It is in the northeast of Sønderborg towards Augustenborg Fjord. The present work takes into consideration the regional and local geology of the Sønderborg area, construction of the wells, field experience and water chemistry. The main issues of the geothermal plant appear to be related to the construction of the wells and reinjection of the heat-depleted brine. Our water chemistry analysis and PHREEQC simulations indicate that geothermal brine was saturated with respect to carbonate and barite minerals. The excess of Ca2+ and SO42− ions could have led to the formation and precipitation of carbonate and sulfate scales. Moreover, the increment of iron concentration over time could suggest the ingress of oxygen and pitting corrosion due to the presence of halide ions

    Low Frequency Quantum Transport in a Three-probe Mesoscopic Conductor

    The low frequency quantum transport properties of a three-probe mesoscopic conductor are studied using B\"uttiker's AC transport formalism. The static transmission coefficients and emittance matrix of the system were computed by explicitly evaluating the various partial density of states (PDOS). We have investigated the finite size effect of the scattering volume on the global PDOS. By increasing the scattering volume we observed a gradual improvement in the agreement of the total DOS as computed externally or locally.     Charge injection instability in perfect insulators

    We show that in a macroscopic perfect insulator, charge injection at a field-enhancing defect is associated with an instability of the insulating state or with bistability of the insulating and the charged state. The effect of a nonlinear carrier mobility is emphasized. The formation of the charged state is governed by two different processes with clearly separated time scales. First, due to a fast growth of a charge-injection mode, a localized charge cloud forms near the injecting defect (or contact). Charge injection stops when the field enhancement is screened below criticality. Secondly, the charge slowly redistributes in the bulk. The linear instability mechanism and the final charged steady state are discussed for a simple model and for cylindrical and spherical geometries. The theory explains an experimentally observed increase of the critical electric field with decreasing size of the injecting contact.
