An investigation of siloxane cross-linked hydroxyapatite–gelatin/copolymer composites for potential orthopedic applications
Causes of bone deficiency are numerous, but biomimetic alloplastic grafts provide an alternative to repair tissue naturally. Previously, a hydroxyapatite-gelatin modified siloxane (HAp-Gemosil) composite was prepared by cross-linking (N, N′-bis[(3-trimethoxysilyl)propyl]ethylene diamine (enTMOS) around the HAp-Gel nanocomposite particles, to mimic the natural composition and properties of bone. However, the tensile strength remained too low for many orthopedic applications. It was hypothesized that incorporating a polymer chain into the composite could help improve long range interaction. Furthermore, designing this polymer to interact with the enTMOS siloxane cross-linked matrix would provide improved adhesion between the polymer and the ceramic composite, and improve mechanical properties. To this end, copolymers of L-Lactide (LLA), and a novel alkyne derivatized trimethylene carbonate, propargyl carbonate (PC), were synthesized. Incorporation of PC during copolymerization affects properties of copolymers such as molecular weight, Tg, and % PC incorporation. More importantly, PC monomers bear a synthetic handle, allowing copolymers to undergo post-polymerization functionalization with graft monomers to specifically tailor the properties of the final composite. For our investigation, P(LLA-co-PC) copolymers were functionalized by an azido-silane (AS) via copper catalyzed azide-alkyne cycloaddition (CuAAC) through terminal alkyne on PC monomers. The new functionalized polymer, P(LLA-co-PC)(AS) was blended with HAp-Gemosil, with the azido-silane linking the copolymer to the silsesquioxane matrix within the final composite
Self‐potential ambient noise and spectral relationship with urbanization, seismicity, and strain rate revealed via the Taiwan Geoelectric Monitoring Network
AbstractGeoelectric self‐potential (SP) signals are sensitive to natural and anthropogenic factors. The SP spectral characteristics under the different factors in Taiwan were investigated, and the SP spectral scalings were correlated with urbanization level, seismicity, and crustal deformation. The ambient SP noise models were first established by estimating the probability density functions of the spectrograms at each frequency. The effects of the natural and anthropogenic factors on the SP signals are understood by comparing the SP noise models under various conditions, such as precipitation, urbanization, and electric trains. Results show that the SP signals in areas of high industrialization and human activity and areas close to train stations behave as white noises and exhibit a distinct spectral ripple at frequencies around 1 Hz. On the other hand, the SP spectral power law parameters, Gutenberg‐Richter b values, and dilation strain rates were estimated by using the SP, earthquake catalog, and GPS data, respectively, during 2012–2017. By investigating the correlations of the SP spectral parameters with the Gutenberg‐Richter b value, dilation strain rates, and urbanization level, the SP optimal frequency band is found between 0.006 and 1 Hz due to the high correlation between the SP and seismicity data and between the SP and dilation data and the low correlation between the SP and urbanization data. Hence, this study may help the filtering and screening of the SP data and facilitate the understanding of the mechano‐electric behavior in the crust
Drumhead Surface States and Topological Nodal-Line Fermions in TlTaSe2
A topological nodal-line semimetal is a new condensed matter state with
one-dimensional bulk nodal lines and two-dimensional drumhead surface bands.
Based on first-principles calculations and our effective k . p model, we
propose the existence of topological nodal-line fermions in the ternary
transition- metal chalcogenide TlTaSe2. The noncentrosymmetric structure and
strong spin-orbit coupling give rise to spinful nodal-line bulk states which
are protected by a mirror reflection symmetry of this compound. This is
remarkably distinguished from other proposed nodal-line semimetals such as
Cu3NPb(Zn) in which nodal lines exist only in the limit of vanishing spin-orbit
coupling. We show that the drumhead surface states in TlTaSe2, which are
associated with the topological nodal lines, exhibit an unconventional chiral
spin texture and an exotic Lifshitz transition as a consequence of the linkage
among multiple drumhead surface-state pockets.Comment: Related papers at

Direct scaffolding of biomimetic hydroxyapatite-gelatin nanocomposites using aminosilane cross-linker for bone regeneration
Hydroxyapatite-gelatin modified siloxane (GEMOSIL) nanocomposite was developed by coating, kneading and hardening processes to provide formable scaffolding for alloplastic graft applications. The present study aims to characterize scaffolding formability and mechanical properties of GEMOSIL, and to test the in vitro and in vivo biocompatibility of GEMOSIL. Buffer Solution initiated formable paste followed by the sol-gel reaction led to a final hardened composite. Results showed the adequate coating of aminosilane, 11–19 wt%, affected the cohesiveness of the powders and the final compressive strength (69 MPa) of the composite. TGA and TEM results showed the effective aminosilane coating that preserves hydroxyapatite-gelatin nanocrystals from damage. Both GEMOSIL with and without titania increased the mineralization of preosteoblasts in vitro. Only did titania additives revealed good in vivo bone formation in rat calvarium defects. The scaffolding formability, due to cohesive bonding among GEMOSIL particles, could be further refined to fulfill the complicated scaffold processes
Aminosilane as an effective binder for hydroxyapatite-gelatin nanocomposites
Aminosilane has been explored as an alternative chemical linker to facilitate the binding and solidification of hydroxyapatite-gelatin nanocomposite at room temperature, which was synthesized using co-precipitation method in the presence of gelatin. This aminosilane treatment was found effective at low concentration (~25 μL/mL) and the solidification and dehydration of hydroxyapatite-gelatin slurry completes within hours depending on the amount of aminosilane. The resulting sample exhibits compressive strength of 133 MPa, about 40% higher than glutaraldehyde treated samples, and shows good biocompatibility based on cell adhesion, proliferation, alkaline phosphate synthesis, and mineralization studies
Tumour inflammasome-derived IL-1b recruits neutrophils and improves local recurrence-free survival in EBV-induced nasopharyngeal carcinoma
Inflammasomes sense infection and cellular damage and are critical for triggering inflammation through IL-1β production. In carcinogenesis, inflammasomes may have contradictory roles through facilitating antitumour immunity and inducing oncogenic factors. Their function in cancer remains poorly characterized. Here we show that the NLRP3, AIM2 and RIG-I inflammasomes are overexpressed in Epstein-Barr virus (EBV)-associated nasopharyngeal carcinoma (NPC), and expression levels correlate with patient survival. In tumour cells, AIM2 and RIG-I are required for IL-1β induction by EBV genomic DNA and EBV-encoded small RNAs, respectively, while NLRP3 responds to extracellular ATP and reactive oxygen species. Irradiation and chemotherapy can further activate AIM2 and NLRP3, respectively. In mice, tumour-derived IL-1β inhibits tumour growth and enhances survival through host responses. Mechanistically, IL-1β-mediated anti-tumour effects depend on infiltrated immunostimulatory neutrophils. We show further that presence of tumour-associated neutrophils is significantly associated with better survival in NPC patients. Thus, tumour inflammasomes play a key role in tumour control by recruiting neutrophils, and their expression levels are favourable prognostic markers and promising therapeutic targets in patients
