The cholesterol-hydroxyecdysone-vitellogenin pathway is involved in the longevity of trophocytes and oenocytes of queen honey bees (Apis mellifera)
International audienceAbstractTrophocytes and oenocytes in the abdomen of honey bees do not divide after eclosion; however, trophocytes and oenocytes of queen bees have a longer lifespan and maintain better cellular function than those of worker bees. To explore this phenomenon, we assayed the molecules involved in the cholesterol-hydroxyecdysone-vitellogenin (Vg) pathway in the trophocytes and oenocytes of young and old worker and queen bees. The results showed that Vg and cholesterol levels in hemolymph and cholesterol levels, 20-hydroxyecdysone (20E) levels, and the messenger RNA levels of cytochrome P450 314A1 20-hydroxylase (Cyp314A1), ecdysone receptor isoform A (EcR-A), ecdysone receptor isoform B1 (EcR-B1), ultraspiracle (USP), ecdysone-induced protein 74 (E74), ecdysone-induced protein 75 (E75), broad-complex (BR-C), Vg, and Vg receptor (VgR) in trophocytes and oenocytes were increased in queen bees compared with worker bees. These findings indicated that queen bees have higher expression of molecules in the cholesterol-hydroxyecdysone-Vg pathway than worker bees
Boosting Factual Consistency and High Coverage in Unsupervised Abstractive Summarization
Abstractive summarization has gained attention because of the positive performance of large-scale, pretrained language models. However, models may generate a summary that contains information different from the original document. This phenomenon is particularly critical under the abstractive methods and is known as factual inconsistency. This study proposes an unsupervised abstractive method for improving factual consistency and coverage by adopting reinforcement learning. The proposed framework includes (1) a novel design to maintain factual consistency with an automatic question-answering process between the generated summary and original document, and (2) a novel method of ranking keywords based on word dependency, where keywords are used to examine the coverage of the key information preserved in the summary. The experimental results show that the proposed method outperforms the reinforcement learning baseline on both the evaluations for factual consistency and coverage
Effects of natto extract on endothelial injury in a rat model
Vascular endothelial damage has been found to be associated with thrombus formation, which is considered to be a risk factor for cardiovascular disease. A diet of natto leads to a low prevalence of cardiovascular disease. The aim of the present study was to investigate the effects of natto extract on vascular endothelia damage with exposure to laser irradiation. Endothelial damage both in vitro and in vivo was induced by irradiation of rose bengal using a DPSS green laser. Cell viability was determined by MTS assay, and the intimal thickening was verified by a histological approach. The antioxidant content of natto extract was determined for the free radical scavenging activity. Endothelial cells were injured in the presence of rose bengal irradiated in a dose-dependent manner. Natto extract exhibits high levels of antioxidant activity compared with purified natto kinase. Apoptosis of laser-injured endothelial cells was significantly reduced in the presence of natto extract. Both the natto extract and natto kinase suppressed intimal thickening in rats with endothelial injury. The present findings suggest that natto extract suppresses vessel thickening as a synergic effect attributed to its antioxidant and anti-apoptosis properties
Chinese Herbal Medicine as an Adjunctive Therapy Ameliorated the Incidence of Chronic Hepatitis in Patients with Breast Cancer: A Nationwide Population-Based Cohort Study
We conducted a National Health Insurance Research Database-based Taiwanese nationwide population-based cohort study to evaluate whether Chinese herbal medicine (CHM) treatment decreased the incidence of chronic hepatitis in breast cancer patients receiving chemotherapy and/or radiotherapy. A total of 81171 patients were diagnosed with breast cancer within the defined study period. After randomly equal matching, data from 13856 patients were analyzed. Hazard ratios of incidence rate of chronic hepatitis were used to determine the influence and therapeutic potential of CHM in patients with breast cancer. The patients with breast cancer receiving CHM treatment exhibited a significantly decreased incidence rate of chronic hepatitis even across the stratification of age, CCI score, and treatments. The cumulative incidence of chronic hepatitis for a period of seven years after initial breast cancer diagnosis was also reduced in the patients receiving CHM treatment. The ten most commonly used single herbs and formulas were effective in protecting liver function in patients with breast cancer, where Hedyotis diffusa and Jia-Wei-Xiao-Yao-San were the most commonly used herbal agents. In conclusion, our study provided information that western medicine therapy combined with CHM as an adjuvant modality may have a significant impact on liver protection in patients with breast cancer
The risk of false inclusion of a relative in parentage testing – an in silico population study
Aim To investigate the potential of false inclusion of a
close genetic relative in paternity testing by using computer
generated families.
Methods 10 000 computer-simulated families over three
generations were generated based on genotypes using 15
short tandem repeat loci. These data were used in assessing
the probability of inclusion or exclusion of paternity
when the father is actually a sibling, grandparent, uncle,
half sibling, cousin, or a random male. Further, we considered
a duo case where the mother’s DNA type was not
available and a trio case including the mother’s profile.
Results The data showed that the duo scenario had the
highest and lowest false inclusion rates when considering
a sibling (19.03 ± 0.77%) and a cousin (0.51 ± 0.14%) as
the father, respectively; and the rate when considering a
random male was much lower (0.04 ± 0.04%). The situation
altered slightly with a trio case where the highest rate
(0.56 ± 0.15%) occurred when a paternal uncle was considered
as the father, and the lowest rate (0.03 ± 0.03%) occurred
when a cousin was considered as the father. We also
report on the distribution of the numbers for non-conformity
(non-matching loci) where the father is a close genetic
relative.
Conclusions The results highlight the risk of false inclusion
in parentage testing. These data provide a valuable
reference when incorporating either a mutation in the father’s
DNA type or if a close relative is included as being
the father; particularly when there are varying numbers of
non-matching loci
A novel strategy for sibship determination in trio sibling model
Aim To use a virtually simulated population, generated
from published allele frequencies based on 15 short tandem
repeats (STR), to evaluate the efficacy of trio sibship
testing and sibling assignment for forensic purposes.
Methods Virtual populations were generated using 15 STR
loci to create a large number of related and unrelated genotypes
(10 000 trio combinations). Using these virtual populations,
the probability of related and unrelated profiles
can be compared to determine the chance of inclusions
of being siblings if they are true siblings and the chance of
inclusion if they are unrelated. Two specific relationships
were tested – two reference siblings were compared to a
third true sibling (3S trio, sibling trio) and two reference siblings
were compared to an unrelated individual (2S1U trio,
non-sibling trio).
Results When the likelihood ratio was greater than 1,
99.87% of siblings in the 3S trio population were considered
as siblings (sensitivity); 99.88% of non-siblings in the
2S1U trio population were considered as non-siblings
(specificity); 99.9% of both populations were identified correctly
as siblings and non-siblings; and the accuracy of the
test was 99.88%.
Conclusions The high sensitivity and specificity figures
when using two known siblings compared to a putative
sibling are significantly greater than when using only one
known relative. The data also support the use of increasing
number of loci allowing for greater confidence in genetic
identification. The system established in this study could
be used as the model for evaluating and simulating the
cases with multiple relatives
