    Induction of apoptosis by the retinoid inducible growth regulator RIG1 depends on the NC motif in HtTA cervical cancer cells

    <p>Abstract</p> <p>Background</p> <p>Retinoid-inducible gene 1 (RIG1), also known as tazarotene-induced gene 3 or retinoic-acid receptor responder 3, is a growth regulator, which induces apoptosis and differentiation. RIG1 is classified into the NC protein family. This study investigated functional domains and critical amino acids associated with RIG1-mediated cell death and apoptosis.</p> <p>Results</p> <p>Using enhanced green fluorescence protein (EGFP)-tagged RIG1 variants, RIG1 proteins with deletion at the NC domain significantly decreased cell death induced by RIG1, and fusion variants containing only the NC domain significantly induced apoptosis of HtTA cervical cancer cells. The EGFP-RIG1-induced apoptosis was significantly decreased in cells expressing N<sup>112</sup>C<sup>113 </sup>motif double- (NC→FG) or triple- (NCR→FGE) mutated RIG1 variants. Using dodecapeptides, nuclear localization and profound cell death was observed in HtTA cells expressing wild type RIG1<sub>111–123 </sub>or Leu<sup>121</sup>-mutated RIG1<sub>111–123</sub>:L→ C peptide, but peptides double- or triple-mutated at the NC motif alone, RIG1<sub>111–123</sub>:NC→FG or RIG1<sub>111–123</sub>:NCR→FGE, were cytoplasmically localized and did not induce apoptosis. The RIG1<sub>111–123 </sub>also induced apoptosis of A2058 melanoma cells but not normal human fibroblasts.</p> <p>Conclusion</p> <p>The NC domain, especially the NC motif, plays the major role in RIG1-mediated pro-apoptotic activity. The RIG1<sub>111–123 </sub>dodecapeptide exhibited strong pro-apoptotic activity and has potential as an anticancer drug.</p

    Glycogen synthase kinase 3α and 3β have distinct functions during cardiogenesis of zebrafish embryo

    <p>Abstract</p> <p>Background</p> <p>Glycogen synthase kinase 3 (GSK3) encodes a serine/threonine protein kinase, is known to play roles in many biological processes. Two closely related GSK3 isoforms encoded by distinct genes: GSK3α (51 kDa) and GSK3β (47 kDa). In previously studies, most GSK3 inhibitors are not only inhibiting GSK3, but are also affecting many other kinases. In addition, because of highly similarity in amino acid sequence between GSK3α and GSK3β, making it difficult to identify an inhibitor that can be selective against GSK3α or GSK3β. Thus, it is relatively difficult to address the functions of GSK3 isoforms during embryogenesis. At this study, we attempt to specifically inhibit either GSK3α or GSK3β and uncover the isoform-specific roles that GSK3 plays during cardiogenesis.</p> <p>Results</p> <p>We blocked <it>gsk3α </it>and <it>gsk3β </it>translations by injection of morpholino antisense oligonucleotides (MO). Both <it>gsk3α</it>- and <it>gsk3β</it>-MO-injected embryos displayed similar morphological defects, with a thin, string-like shaped heart and pericardial edema at 72 hours post-fertilization. However, when detailed analysis of the <it>gsk3α</it>- and <it>gsk3β</it>-MO-induced heart defects, we found that the reduced number of cardiomyocytes in <it>gsk3α </it>morphants during the heart-ring stage was due to apoptosis. On the contrary, <it>gsk3β </it>morphants did not exhibit significant apoptosis in the cardiomyocytes, and the heart developed normally during the heart-ring stage. Later, however, the heart positioning was severely disrupted in <it>gsk3β </it>morphants. <it>bmp4 </it>expression in <it>gsk3β </it>morphants was up-regulated and disrupted the asymmetry pattern in the heart. The cardiac valve defects in <it>gsk3β </it>morphants were similar to those observed in <it>axin1 </it>and <it>apc</it><sup><it>mcr </it></sup>mutants, suggesting that GSK3β might play a role in cardiac valve development through the Wnt/β-catenin pathway. Finally, the phenotypes of <it>gsk3α </it>mutant embryos cannot be rescued by <it>gsk3β </it>mRNA, and vice versa, demonstrating that GSK3α and GSK3β are not functionally redundant.</p> <p>Conclusion</p> <p>We conclude that (1) GSK3α, but not GSK3β, is necessary in cardiomyocyte survival; (2) the GSK3β plays important roles in modulating the left-right asymmetry and affecting heart positioning; and (3) GSK3α and GSK3β play distinct roles during zebrafish cardiogenesis.</p

    Flood Damage Assessment in Taipei City Taiwan

    In this study, we reviewed the literature on flood damage assessment and collected information for related research in Taiwan to analyze the relationships between direct flood damage, flood frequency, flood depth, and land-use. The procedure for flood damage assessment was then developed that includes the following steps: (a) Scenario simulation of inundation potential. (b) Establishment of the relationship between inundation depth and damage loss for varied land-use. (c) Risk analysis of inundation damage. Taipei City in north Taiwan was adopted as the case study to demonstrate the proposed algorithm. Flood events with return periods of 5, 10, 25, 50, 100 and 200 years were used for flood hazard analysis to cover possible flooding scenarios. The inundation hazard maps were first generated via hydraulic modelling. The regional flood damage was then estimated using a relationship between inundation depth and damage. The flood damage exceedance probability (EP) curve for Taipei City was constructed following the association of the loss with its probability of occurrence. The flood damage EP curve was further used to integrate the damage assessments for individual flood events for a full probability range presentation of the flood risk. The expected annual damage was calculated by integrating the area under the EP curve

    Seismic behavior of pile in liquefiable soil ground by centrifuge shaking table tests

    Dramatic failure of pile foundations caused by the soil liquefaction was founded and leading to many studies on the seismic behavior of pile. The failures were often accompanied with settlement, lateral displacement and tilting of superstructures. Therefore soil-structure interaction effects must be properly considered in the design of pile. Two centrifuge models were conducted by shaking table at an acceleration field of 80 g. The purpose of this study was to investigate the seismic response of piles attached with different tip mass and embedded in liquefied or non-liquefied deposits. From the results, it was found that the maximum bending moment of pile occurs at the depth of 4 m and 5 m for dry sand and saturated sand models, respectively. The more tip mass leads to the more permanent lateral displacement and the more residual bending moment

    Coexistent squamous cell carcinoma and adenoid basal carcinoma in the uterine cervix and infection with human papillomavirus (HPV 31)

    AbstractObjectiveAdenoid basal carcinoma (ABC) is an uncommon neoplasm of the uterine cervix. ABC can be accompanied by carcinoma in situ or invasive carcinoma. Most cases are discovered accidentally during radical hysterectomy. ABC is associated with a high risk of human papillomavirus infection (HPV), most often HPV 16 infection.Case reportWe present a rare case of an 86-year-old Taiwanese married woman who suffered from bloody vaginal discharge and occasional lower abdominal pain and received cervical biopsy. The pathological report revealed squamous cell carcinoma (SCC) of the uterine cervix. After radical hysterectomy, bilateral salpingo-oophorectomy, and bilateral pelvic and para-aortic lymph node dissection, the final pathological report revealed SCC coexisting with ABC, and both of the components were infected by HPV 31. After receiving radiotherapy, she maintained outpatient department follow-up.ConclusionA literature review revealed that this was a rare case of combined ABC–SCC associated with HPV 31 infection. In this case, the ABC component did not affect the tumor stage because it was confined to the cervix. However, we must avoid overestimating the clinical stage because the ABC component is thought to be a benign lesion

    Porcine circovirus type 2 (PCV2) infection decreases the efficacy of an attenuated classical swine fever virus (CSFV) vaccine

    The Lapinized Philippines Coronel (LPC) vaccine, an attenuated strain of classical swine fever virus (CSFV), is an important tool for the prevention and control of CSFV infection and is widely and routinely used in most CSF endemic areas, including Taiwan. The aim of this study was to investigate whether PCV2 infection affects the efficacy of the LPC vaccine. Eighteen 6-week-old, cesarean-derived and colostrum-deprived (CDCD), crossbred pigs were randomly assigned to four groups. A total of 105.3 TCID50 of PCV2 was experimentally inoculated into pigs through both intranasal and intramuscular routes at 0 days post-inoculation (dpi) followed by LPC vaccination 12 days later. All the animals were challenged with wild-type CSFV (ALD stain) at 27 dpi and euthanized at 45 dpi. Following CSFV challenge, the LPC-vaccinated pigs pre-inoculated with PCV2 showed transient fever, viremia, and viral shedding in the saliva and feces. The number of IgM+, CD4+CD8-CD25+, CD4+CD8+CD25+, and CD4-CD8+CD25+ lymphocyte subsets and the level of neutralizing antibodies against CSFV were significantly higher in the animals with LPC vaccination alone than in the pigs with PCV2 inoculation/LPC vaccination. In addition, PCV2-derived inhibition of the CSFV-specific cell proliferative response of peripheral blood mononuclear cells (PBMCs) was demonstrated in an ex vivo experiment. These findings indicate that PCV2 infection decreases the efficacy of the LPC vaccine. This PCV2-derived interference may not only allow the invasion of wild-type CSFV in pig farms but also increases the difficulty of CSF prevention and control in CSF endemic areas

    Flood Vulnerability and Risk Maps in Taipei City, Taiwan

    This paper presents the process of constructing a flood risk map in Taipei City. The study calculates the social vulnerability index (SVI) for flooding at a district level, based on five factors including (1) female population (2) alone living elderly (3) low-income households (4) household income and (5) house-hold possessions. The index is determined according to the factor ratios in a district comparing to the statistical average across Taipei City. By combining the SVI with spatial varied flood potential information simulated by a hydraulic model, the flood risk index is obtained for district level that has an area of about 0.2 km2. Results show that the flood risk in Taipei City changed from 2002 to 2010 due to changes in the demographic structure. During the period from 2002 to 2004, the most obvious change of flood risk occurred in the Wanhua district due to the increase in the ratio of household possessions, which escalated the vulnerability to flooding. Between 2005 and 2007, the ratio of household possessions dropped in the Nankang district such that the flood risk reduced mostly in the region
